Herein, we develop a virtual cathode display as a molecular-resolution mixed reality interface, which can exhibit electric field phenomena in a dynamical nanopattern. A thin membrane window of SiN is employed as a display surface for the presentation of electric field. Backside projection of a scanning focused electron beam generates nanoscale electric, chemical, and mechanical phenomena on the SiN surface.     Gliding of microtubules (MTs) on kinesins has been applied to lab-on-a-chip devices, which enable autonomous transportation and detection of biomolecules in the field of bioengineering. For rapid fabrication and evaluation of the kinesin–MT based devices, optical control techniques have been developed for control of kinesin activity and density; however, use of caged molecules lacks spatial controllability for long-term experiments, and direct irradiations of UV light onto kinesin-coated surfaces are inherently damaging to MTs due to their depth limit since the heights of the kinesin–MT systems are at the tens of a nanometer scale. Considering surface electric fields in electrolytic solutions are shielded at the nanometer scale due to Debye shielding, in this study, we show that fine spatial control of kinesin density and activity is enabled using surface-limited electrochemical reactions induced by indirect irradiations of an electron beam (EB). An EB is indirectly irradiated onto the kinesins through a 100-nm-thick silicon nitride membrane, and the electrons scattered in the membrane can cause localized electrochemical effects to the kinesins. We show that these localized electrochemical effects cause both ablation of kinesins and motility control of kinesin activity by changing the EB acceleration voltage. In particular, the latter is achieved without complete ablation of MTs, though the MTs are indirectly irradiated by the EB. As a demonstration of on-demand control of gliding MTs, we show the accumulation of the MTs on a target area by scanning the EB. The proposed accumulation technique will lead to rapid prototyping of microdevices based on MT–kinesin motility assay systems.

    Environmental and biomedical monitoring bio-applications need the multi-step processing of the chip designs for patterning the biomolecules. This multi-step processing takes a lot of costs to change the biochip design and reconstruct them therefore optimization of the chip function is difficult. So, for reducing processing steps, we propose the on-demand and electrically switched steering of the gliding non-tagged microtubules by the virtual cathode that can generate the electrochemical reactions freely in two dimensional. This method is simpler processing of prototyping than the conventional methods and could be lead to develop the biomedical bio-applications.

    The dynamic electromechanical control of spatial structures of biomolecules in aqueous solutions was demonstrated using a nano virtual cathode display. By generating a focused electric field around the biomolecules using an electron beam (EB), the molecules' spatiotemporal responses to the electrical stimuli, such as globule transition of DNA random coils and deformation of planar lipid bilayers and vesicles, were successfully observed. The proposed system may be applied to high resolution and high degree-of-freedom manipulations to measure the mechanical and structural properties of bio-nanomaterials.

    Placing cells in the proper position is important for tissue engineering. Previous works addressed this subject in the way of controlling cell migration by micro- or nano-patterning the substrates. However, the problem of changing spatial cell density freely under co-culture conditions is remaining. To solve this problem, in this work, we report that C2C12 spatial cell density changes by the patterning geometric boundary of the topographical structures. In 48 h after seeding cells, at the linear boundary (ridge-groove) structures, C2C12 Groove/Ridge ratio was under 0.70 both under monoculture conditions and under co-culture conditions. In contrast, at the combining the linear boundary and the round boundary (ridge-groove + hole) structures, the ratio was over 0.89 under both culture conditions. This our finding will provide a new device which enables to manipulate spatial cell density under co-culture conditions for heterogeneous tissue engineering.

    Electron beam (EB) induced fine virtual cathode (VC) on a 100-nm-thick SiN membrane has been developed using inverted-EB lithography (I-EBL). Here we present electrokinetic property of the VC for biological materials and instantly control their spatio-temporal responses using the VC. Negatively charged biomolecules (proteins, DNAs) respond to locally injected negative electrons of VC, and change their electrostatic interaction which affects the molecular aggregation/dissociation and adhesion/detachment in water solution. So, we call this technique virtual cathode display for biomolecules. This technique was applied to in-situ molecules patterning for mechanical strain microscopy and pinpoint transmembrane molecules delivery.

    In our study, we used the dorsal vessel (DV, heart-like tissue) of Drosophila melanogaster (Dmel, fruit fly) larva as a bioactuator. This paper reports on the closed loop visual feedback system to control the contraction rates of the DV in vivo. We investigated the response of the DVs that express channelrhodopsin(ChR2) and/or halorhodopsin(NpHR) to blue and red light. We found that DV expressing ChR2 increased contraction rates under blue light stimulation, while DV expressing NpHR stopped contracting under red light stimulation. In our feedback experiments, we managed to keep the contraction frequency of the DV in vivo above an arbitrary threshold value using the visual feedback system we developed.

    This paper proposed a Multiple Microfluidic Stream based Manipulation (MMSM) system for biological object using micro hydrodynamics and Lab on a Chip (LOC) technology. Our method can implement the function of micro manipulation and micro assembly of bio-objects without contact in an open space. Compared with other conventional bio-micro manipulation and assembly methods, this system is manipulating a micro object by controlling multiple microfluidic streams onto it from various directions. The advantage of this method is open space, multi-function, multi-scale, multi degree of freedom, and non-invasive three dimensions manipulation. These microfluidic streams are generated simultaneously from multiple orifices. By regulating the parameters of the microfluidic stream such as flow rates, position and number of operating orifices, the direction and velocity of the object can be controlled. To verify this principle, we designed an open space fluidic system for on-chip manipulation. The results presented in this paper showed that this MMSM has the capability for Micro Bio-manipulation.

    We have demonstrated hybrid micro mechanical systems (HMMS) using muscle cells. When constructing cells to HMMS, forces between adhesive interface and cells and contraction of muscle cells must be considered to design. When these forces are larger than adhesion force of cells, cells detach from substrates and system is not functional. Measuring the adhesion force of cells could prevent from the breakage of the system. In this paper, we developed cell adhesion force measurement system defined by "International Organization for Standardization (ISO)". As a result, we measured maximum adhesion strength of cells about 10 μN/mm. © 2009 CBMS.

    Bio-actuators using living cells or tissue cannot work outside culture dish filled with culture medium. Therefore, the limitation of bio-actuator's operating environment is difficult problem compared to conventional micro actuator. To solve this problem, we focused on packaging of insect dorsal vessel and its culture medium by exoskeletal structure. By packaging of insect dorsal vessel and its culture medium, the bio-actuator using insect dorsal vessel would work outside culture dish filled with culture medium. We fabricated exoskeletal structure that has micro through-holes. The TC-100 culture medium was filled inside exoskeletal structure, without its leakage due to surface tension. We fabricated micro-manipulator using contracting force of insect dorsal vessel. The open-end displacement by contracting force of insect dorsal vessel was approximately 60 μm. After the assembly of insect dorsal vessel into exoskeletal structure filled with culture medium inside, the contracting of insect dorsal vessel was confirmed. ©2009 IEEE.

    This paper describes assembly of three dimensional cellular structure by optical tweezers. We attempted to construct a three dimensional quasi living machine in small space with muscle cells. Cardiomyocytes were manipulated by using laser optical tweezers at wavelength of 1064 nm. The cellular bridge structure was successful constructed on between PDMS micro pillars. The viability and contraction of that was was confirmed. We also demonstrated a nano assembly of a cell-driven nano-mechanical tweezers by using a optical tweezers. The nano mechanical tweezers was designed as single cardiomyocyte contraction force for a nano actuator. A myocyte contraction force is tiny force such as <10 mu N, however it is enough large power to drive the nano mechanics. This nano mechanics was three-dimensional carbon structure which was designed for nano tweezers. Focused ion beam induced chemical vapor deposition of the carbon nano mechanics. Optical tweezers allowed to combine the nano tweezers and a single beating cardiomyocyte on it. We demonstrated assembly of single cardiomyocyte powered nano tweezers, which had beams of 150 nm diameter and 20 mu m length, and live cardiomyocyte.

    This paper describes assembly of three dimensional cellular structure by optical tweezers. We attempted to construct a three dimensional quasi living machine in small space with muscle cells. Cardiomyocytes were manipulated by using laser optical tweezers at wavelength of 1064 nm. We demonstrated a nano assembly of a cell-driven nano-mechanical tweezers by using a optical tweezers. The nano mechanical tweezers was designed as single cardiomyocyte diven nano-machine.     We have demonstrated hybrid micro mechanical systems (HMMS) using muscle cells. When constructing cells to HMMS, there are some forces between adhesive interface and cells, dur to contraction of muscle cells must be considered to design. When these forces are larger than adhesion force of cells, cells detach from substrates and system is not functional. Measuring the adhesion force of cells could prevent from the breakage of the system. In this paper, we developed cell adhesion force measurement system "90 degree detachment examination" which defined by "International Organization for Standardization (ISO)". To attempt 90 degree detachment examination, we noticed that to use cell sheet which cultured confluent on two-dimensional surface. And to use cell sheet, we develop four types of cell sheet handling tools. As a result, we measured maximum adhesion strength of cells about 10 N/mm. ©2009 IEEE.

    Mechano-bionic micro tool is a fusion of the micro mechanical components with the bionic systems. Mechanical output from mechano-bionic system could be caused by combination of multiple functions via artificial and bionic components. In this study, we demonstrated muscular cells driven micro tweezers-like structure with artificial three-dimensional micro/nano structure as a mechano-bionic system. We employed structural depend movements for the tweezers with precise three-dimensional micro scaffold to control principal movement without complex molecular biological cellular differentiation and adhesion techniques. The asymmetric inner structure with isometric stiffness could work for guide structure such as joints and skeleton. Focused ion beam technique for three-dimensional fabrication was employed to manufacture biocompatible three-dimensional scaffold on dog's body hair. Skeletal myoblast of rat primary culture were attached on the rapid prototyped structures, and the cells successfully differentiated to myotubes. The deformation of the manipulator was not enough, however the micro structure was driven by muscular contraction due to electrical stimulation. This study was the first trial to realize endoskeletal mechanical micro system using the asymmetric three-dimensional structure.

    This paper proposes a nano-needle intracellular interface which was assembled by using a cell's own locomotion force. This interface was designed for large scale intracellular recording for development of an artificial synaptic device. We previously demonstrated a method to insert a nanostructure into a living cell using cell locomotion. To achieve an intracellular nanoelectrode, we employed such cell migration on an extracellular matrix as the driving force to insert a nanoelectrode into a plasma membrane. In this study, we describe intracellular nanoelectrodes inserted by the cell migration force which are used to construct intracellular recording devices. Each microwell had a low cell binding glass plate which had cellular adhesive molecules, and there was an electrically conductive tungsten nanoneedle about 190 nm in diameter on the bottom of the microwell. Cells can adhere only in the microwell; thus they are pulled down by their own body and are inserted into the nanoneedle by their own migration force. Then, the living cellular activity can be measured from the electrically conductive nanoneedle. Using this concept, we attempted to achieve a long-lasting intracellular multi channel interface between living cells and artificial implements without making a precise mechanical alignment and without an invasive procedure. This report explains the fabrication and electrical characteristics of the interface for development of the physical part of the cell-machine interface.

    Two types of intracortical multichannel flexible neural probes coated with a hybrid biodegradable polymer were designed and fabricated. The hybrid biodegradable polymer was made of polyethylene glycol and biodegradable polymeric microspheres containing nerve growth factor (NGF) and incorporated to improve the stiffness for flexible neural probe insertion, and promote regrowth of damaged neural tissues around the probe. The type-A neural probe has a groove structure designed to be seeded with a large amount of the hybrid biodegradable polymer. The type-B neural probe has a unique configuration like skeleton to minimize the volume of the flexible probe and buffer injurious micromotion between the probe and the tissues after implantation. In this preliminary study, the efficacy of the released NGF from the microspheres with the PC12 cells was examined. Further, neural probe implantation and neural signal recording with an acute experiment were studied.

    A motion support system is important for improving the quality of life for people with inadequate muscle power for walking or standing. Such a system must maintain coordination between the movement of the living body and that of the motion support system with respect to timing and power. We have developed a preliminary assistance device for human locomotion that does not restrict movement of the user's lower extremities. To determine the appropriate timing and applied force for this assistance device, we have use two types control mode; (1) surface EMG signals from the lower limb in preliminary controller. Each assistance leg has a three-axis magnetic sensor near lower end which tracks a magnet attached to each ankle of the user's legs. This sensor attracted to a magnet attached to the human ankle in the leg swing phase. The mechanical leg supports the user's weight when the leg is in the standing phase.

    Nano-tools are very important for the subcellular operation and analysis in the bio-experiments. For giving the high-performance to nano-tool, we think that it is necessary to make tools the three-dimensional (3-D) nano-structure devices. Therefore, we have performed the 3-D nano tools fabrication by using Focused-Ion-Beam Chernical-Vapor-Deposition (FIB-CVD). This time, a nano-net as a novel 3-D nano-tool was fabricated by using FIB-CVD. And we succeeded in scooping the polystyrene micro-sphere witha diameter of 2.0 mu m by using a nano-net.

    A motion support system is very important for improving the quality of life for the people who have inadequate muscle power for walking or standing. One of the most important aspects of the development of such systems is how to keep coordination between the movement of the living body and that of the motion support system with respect to timing and power. In this study, we tried to develop an algorithm that would be capable of solving this problem by measuring the torque and the viscosity of the movements of the living body from a surface electromyogram (EMG), and varying the torque and viscosity of the support system in proportion to those of the living body. The results demonstrated that the torque of the joint of the living boas, coincided very well with the estimated value calculated from integrated surface EMG and it was possible to support the subject's motion such that he could perform smooth motions while exerting with a small amount of power.

    In this paper, we report fabrication and electrical evaluation of various air-wirings formed by FIB-CVD. The FIB system utilizing a beam of 30 keV Ga/sup +/ ion was applied to make air-wiring. We fabricated two kinds of air-wirings. One was fabricated using two phenanthrene (C4H10) source gases. The other was fabricated using a mixture gas of tungsten carbonyl (W(CO)6) and C4H10 source gases. Then the electrical resistivities were compared.

    A master-slave manipulation system with a force-feedback function was developed for endoscopic surgery. The slave manipulation system, which is located at the head of a robot arm system, uses a forceps system which is capable of being opened and closed by a DC motor-driven arrangement. The master manipulator involves a mechanical link system which has the same structure and scale as the slave robot-arm system. The value of this system was verified experimentally using a bench-test system with one degree of freedom which has the same force-feedback mechanism as the master-slave manipulation system. Although the system became oscillatory and unstable when the master manipulator was operated using a force feedback system (simple force reflection servo controller), it was possible to make the system stable improving the force feedback system by adding impedance term (improved force reflection servo controller).
