    代表研究者はメダカ生殖細胞の性決定研究においてメス特有の減数分裂機構が存在することを発見し(Science 2015, PNAS 2020)、染色体高次構造を作り出すコヒーシン(REC8)が関わることを見出した。コヒーシンの量的あるいは質的な性差が染色体高次構造を変え、組換えランドスケープの性差をもたらすと考えられたことから、減数分裂動態が解析可能なイメージング手法の開発を行い、Rec8 や他の変異体と組み合わせて減数分裂染色体構造を解析し、減数分裂の性差をもたらす分子機構を明らかにする。
    申請者はメダカ生殖細胞の性決定研究においてメス特有の減数分裂機構が存在することを発見し(Science 2015, PNAS 2020)、染色体高次構造を作り出すコヒーシン(REC8)が関わることを見出した。メダカには2つのrec8 遺伝子(rec8a, rec8b)が存在し、rec8a変異体はメスでのみ減数分裂が停止することから、コヒーシンの量的あるいは質的な性差が染色体高次構造を変え、組換えランドスケープの性差をもたらすと考えた。
    減数分裂の性差は、染色体あたりの組換えの数や位置(組換えランドスケープ)の差として認められ、その組換えランドスケープは以下に述べる減数分裂染色体の「高次構造」と「核内動態」を基盤として形成される。本研究ではこの高次構造と核内動態に着目して解析を行う。
    本研究では、高輝度高解像度のライブイメージング手法と条件的タンパク質分解法を確立し、独自の変異体を用いた核内動態解析と超解像度顕微鏡による染色体構造解析を行い、組換えランドスケープの性差をもたらす分子機構を明らかにする。
    相同染色体軸を構成するタンパク質 SCYP3 と EGFP とを融合させた遺伝子をメダカの SCYP3 遺伝子の制御領域に組み込んだコンストラクトを作製し、メダカに導入してトランスジェニックメダカSYCP3_EGFP 系統を確立した。そこでSYCP3_EGFP 系統を用いて、減数分裂期の染色体動態の可視化を試みた。
    この系統とスピニングディスク共焦点レーザー顕微鏡を用いて最終的に数分にわたる減数分裂前期相同染色体動態のイメージングに成功した。減数分裂前期は相同染色体の形成過程でレプトテンやザイゴテン、パキテンなどに分けられるが、以前のマウスの報告とは異なる時期、パキテン期にメダカの相同染色体は大きな回転運動を行うことが示された。
    また AID を用いた特定タンパク質の誘導的分解がメダカの生殖細胞でも適応できるかの検討を行なった。AID をターゲットとした分解を媒介するTIR と、AID でタグ付した EGFP のRNA をメダカ受精卵にマイクロインジェクションし、分解誘導物質オーキシンアナログの存在、非存在下で生殖細胞でのEGFP蛍光強度を調べた。オーキシンアナログを加えた場合インジェクションRNAから翻訳されたEGFP蛍光は加えない時よりも明らかに減弱していた。
    また組換えランドスケープを調べるために、組換えマーカーMLH1に対する抗体、雌雄の生殖細胞で発現の異なるRec8a/bに対する抗体の作製を行なった。Rec8a/bのそれぞれに対する特異抗体を得ることには成功した。
    特定タンパク質をAIDにより誘導分解するシステムはメダカ でも可能との予備結果が得られたが、これはRNAから翻訳されたタンパク質での結果である。トランスジェニックメダカも用いて、de novo でタンパク質が発現した時の詳細なデータの取得を引き続き行う。
    減数分裂前期の相同染色体動態の解析に成功し、他の動物では報告のない知見も得ることができた。今後はこの培養時間とイメージ取得時間を長くすることを試みる。パキテン以前の染色体動体がどうであるのかの解析も推し進める。そしてこの動きに性差があった場合、terb1や rec8 などのその他の減数分裂因子変異体によっても動態や組換えランドスケープの性差が影響を受けるのかを解析する。また組換えランドスケープを解析するための抗体作製をおこなう。
    また動態とは別にrec8a/bなどの減数分裂因子変異体の相同染色体軸長や組換え位置や個数の性差を調べ、これらのどの要素が減数分裂生さと繋がるのかを明らかにしていく。

    The goal of the project was to reveal how chromosome and nucleolus dynamics differ between male and female germ cells and the mechanism behind the sexual dimorphism. To achieve the goal, live imaging of medaka germ cells was established in collaboration with Dr. Huynh in the College de France, who has pioneered live imaging technique using Drosophila germ cells. By culturing testes in a microfluidic device during live imaging with a spinning disc confocal laser microscopy, we successfully monitored medaka germ cells for more than 24 hours at a high resolution. We also generated transgenic medaka in which synaptonemal complex, a protein structure forms between homologous chromosomes during meiotic prophase, was able to be visualized by EGFP, which allows monitoring the dynamics of chromosome by live imaging.

    Starve treatment after hatching makes XX female medaka functional male. We revealed that the starvation causes a decrease of lipogenesis via a reduced activity of a CoA-producing pathway. The reduction of lipogenesis then leads to repression of dmrt1 expression that is a critical gene for testis development. This result is the first mechanistic demonstration that genetically determined vertebrate is affected by the change of metabolism induced by environment.
    We also found genetic regulation on the spectrum of germ cell sex. After expression of germ cell sexual switch gene, foxl3, in germline stem cells, germ cells initiate oogenesis. However, germ cells remains feasible in sexual state and could develop into spermatogenesis. We found that feminization of germ cells establishes by pachytene stage. The mechanism underlying this spectrum demonstrates flexible sex that organisms originally possess and may ensure the base for creation of naturally sex reversed organism during evolution.

    The number of original papers by internal collaboration was only 3 at the time of the mid-term evaluation in 2019, but as of May 2022, it has increased dramatically to 21. A technical seminar on Single cell RNA-seq, DamID-seq, and ChIP-seq technology was held at the young researchers meeting. A technical seminar on metabolome analysis and CUT&RUN technology was held at the area meeting.     A series of studies have successfully revealed the presence of factors that regulate initiation of spermatogenesis at the timing of secondary sex characteristics. These include lhx8b and figla, which have been recognized as folliculogenic factors in mammals, and fbxo47, a component of ubiquitin ligase complex, and function downstream of sexual switch gene of germ cells, foxl3. Loss of these factors allowed spermatogenesis before onset of secondary sex characteristics. Regulations of chromosomal structures and membrane trafficking are involved in this mechanism. In addition, we are getting evidences that these factors are under control of somatic signal downstream of dmrt1.
    The initiation of gametogenesis has been studied in terms of endocrinological regulation in the brain. However, these studies, for the first time, reveal that the initiation of spermatogenesis is associated with the mechanism of sex differentiation in the gonad.
