    腱,血管,椎間板などは損傷すると自己再生しない.自家組織移植による修復には移植片採取回数の限度,移植片採取部の治癒などの問題があり,また現在の人工代替材料を用いた修復術にも生体適合性や長期使用による機能劣化の問題が残る.したがって,適切な生理的・力学的機能を有し,最終的に組織再生を導く人工組織が渇望されている.そこで本研究では腱,血管,椎間板を対象とし,其々に特有の線維配向を模倣し,かつ生理的・力学的機能を有する人工組織を,力学負荷を用いて線維形成・配向誘導と構造体を一体形成する技術開発に挑戦する.
    腱,血管,椎間板などは損傷すると自己再生しない.自家組織移植による修復には移植片採取回数の限度,移植片採取部の治癒などの問題があり,また現在の人工代替材料を用いた修復術にも生体適合性や長期使用による機能劣化の問題が残る.これらの問題は人工組織の力学特性が周囲組織と一致しないことも一因と考えられている.したがって,適切な生理的生体適合性(低異物反応)と機能的生体適合性(線維配向,弾性,強度の周囲との一致性)を有し,最終的に組織再生を導く人工組織を効率的に作製する技術の登場が渇望されている.力学負荷を支持する軟組織はコラーゲンやエラスチンなどの線維タンパク質によって構成され,組織の力学特性,特に非線形な応力-ひずみ関係は構成タンパク質の成す構造の配向性に依存することが知られている.
    これまでに提案されている組織形成方法は,例えばエレクトロスピニング法や生体内組織形成法がある.エレクトロスピニング法はkVオーダーの高電圧を用いて線維タンパク質をナノメートルオーダーの極細線維として射出し,無配向または特定方向に揃った線維組織を作製する方法である.人工高分子の繊維組織形成には有用であるものの,熱変性を生じるコラーゲンには不適であり,エレクトロスピニング法でコラーゲン組織を作ることは難しい.生分解性の鋳型を動物の皮下に埋殖し,生体が異物を包み込む作用を利用して特定の形状を有する組織を形成させる方法も提案されているが,作製に時間を有するため現実的な方法とは言えない.
    そこで本研究では,生体組織は作用する力学負荷に適応した最適な構造を形成する原理から,短時間に任意の線維配向をもつ組織を形成する独自の方法を考案し,人工の腱,血管,椎間板を作製することのできる基盤技術の開発に挑戦する.
    これまでに,酸性コラーゲン溶液を中和したものを鋳型に入れ,ダンベル型コラーゲンゲルを作製した.次いでこのゲルに2 gfの静的荷重を作用させて24時間インキュベートし,引張試験で弾性率,破断応力を調べたところ,無負荷インキュベートしたゲルと比べて弾性率,破断応力共に有意に上昇した.また,2光子顕微鏡でコラーゲンSHG光を観察して線維構造を評価したところ,負荷インキュベーションゲル内の線維が負荷方向に配向した.続いて,インキュベート中に架橋剤を導入したところ,弾性率と破断応力の更なる上昇が確認された.また負荷インキュベート時間を48時間に延ばした場合でも,破断応力の上昇が確認された.このように,力学負荷と架橋剤を組み合わせることで,コラーゲン組織の弾性率と破断応力を短時間で飛躍的に上昇させる方法を見出した.一方で,生体への埋殖を考えると弾性率,強度にまだ改善の余地があることも確認した.
    今後は架橋条件や力学負荷量の最適化などを行って,弾性率と破断応力の更なる上昇を図る.また初年度に確立した方法を元に,円筒形ゲルに力学負荷を与えて血管や椎間板線維輪に類似した線維配向を有する人工組織を作製する技術開発に挑む.

    This study was conducted to control cell function utilizing intranuclear dispersion of DNA induced with mechanical deformation of the nucleus. We fabricated a PDMS substrate having a narrow groove with a depth of 35 μm and a width of about 10 μm, and stretched it to widen the groove to make cultured osteoblastic cells (MC3T3-E1) into the groove. The substrate was then released to compress cell nuclei along with the cytoplasm by 15-20%. No change was observed in the number or volume of DNA aggregates in one compression, but the total volume of the aggregates decreased when they are compressed five times. The number of the aggregates increased or decreased depending on the sample. The total volume of aggregates may decrease when the nuclei receive enough amount of compression stimulus. With regard to their number, the change was not simple for it may increase when the aggregates split and may decrease when the split volume become below the measurement limit.

    Two-year study was conducted to control cell function utilizing intranuclear dispersion of DNA induced with mechanical stimulation to the nucleus. We compressed the nucleus of cultured osteoblastic cell line MC3T3-E1 with a micropipette tip by 5 microns, and found that the number of aggregates of chromatin decreases in 3 minutes. We then fabricated a PDMS substrate having wells (100 micron x 100 micron, depth 10 micron) with soft photolithography. We put the cells in the wells, one per each well, and tried to compress them with a cover glass, but failed because the parallelism between the substrate and the cover glass was not enough.

    We investigated the mechanism of a mechanical memory of the intracelualr forces and structures with consideration for cell aging. We used vascular smooth muscle cells and found that cell aging increased cell projected area while it reduced intracellular tension and migration ability. We further found that tensional memory of actin stress fibers in vascular smooth muscle cells tended to decrease with cell aging. These results indicate that cell aging not only affects cell morphology and biochemical functions but also mechanical propeties of cells involving in cellular mechanotransduction.

    In order to estimate 3D stress distribution in Xenopus laevis embryos, we have established two methods, a sectioning method and a piercing method. In the sectioning method, an embryo is cut flat with a thin wire to measure the surface topography of the section along with the stiffness distribution on the surface. Stress distribution is estimated from these data with a finite element analysis as the stress distribution necessary to make the surface flat again. In the piercing method, a fine needle is pierced into an embryo and removed. Then, 3D shape of the pierced column is measured by serial sectioning of the frozen embryo. We have found in gastrula that the embryonic surface is in tension and embryonic core is in compression, and that the level of stresses changes during the process of development, and in tailbud that the notochord produces high compressive force which may cause tissue elongation.

    In the planned research in this area, we aimed to elucidate the principles of macroscopic 3D morphogenesis, which have not been addressed in previous morphogenesis research, by focusing on (1) folding and unfolding of cell sheets and (2) rotation of cell populations. In addition to the above two themes, we also adopted research related to 3D morphogenesis as open research. For both themes, we provided support in terms of equipment and opportunities for discussion so that the experimental and mathematical groups could work together smoothly. Although this was a very challenging research theme, we were able to show with actual examples that it is possible to analyze 3D morphogenesis without losing dimensions, and we believe that we were able to open up a new avenue for morphogenesis research.

    Biological tissues change themselves actively in response to force applied to them. To elucidate this mechanism, we need to know 3D stress and strain distributions at cellular level and compare it with proteins produced by each cell. We studied 3D deformation of intramural microscopic structure in the rabbit thoracic aortas, and found that 1) the longitudinal waviness of the elastic lamina diminishes in response to longitudinal stretch and the circumferential waviness decreases in response to pressurization; and 2) decrease in the waviness of collagen fibers in response to stretch is larger in the smooth muscle layer than in the elastic lamina. We also found that actin filaments are less abundant in the cells reside in the area with high waviness of the elastic lamina.
