    近年、低温でプラズマを発生する技術が進歩したことにより、生命科学への応用が盛んに行われている。WHOが2020年3月11日に新型コロナウイルス感染症はパンデミックであると宣言して以来、感染症対策は世界中で喫緊の課題となっている。本研究では、感染症拡大抑制に資する低温プラズマ及びプラズマ活性溶液の開発を目指す。具体的には豚コロナウイルスや新型コロナウイルスに対する低温プラズマ照射及びプラズマ活性溶液による不活化試験を行う。更に、ウイルスのエンベロープの構造の変化を調べることにより、エンベロープウイルスに対する低温プラズマ及びプラズマ活性溶液による不活化の分子機構を明らかにする。

    近年、低温プラズマのライフサイエンスへの応用研究が活発に進められている。我々は低温プラズマによるがん治療研究を活発に進め、プラズマ照射した溶液による抗腫瘍効果を発見し、この溶液をプラズマ活性溶液と名付けた。その後、プラズマ活性溶液による細胞死の分子機構が世界中で研究され、様々な既知のシグナル伝達経路の関与が明らかになってきた。本研究では、マイクロアレイ解析により未知のプラズマ応答遺伝子を探索し、定量リアルタイムPCR(qRT-PCR)法により検証する。更に質量分析に基づくプロテオミクス解析などを行うことにより低温プラズマやプラズマ活性溶液が活性化する新規シグナル伝達経路を探索する。

    卵巣がんにおける腹膜播種は難治性・治療抵抗性の象徴的な病態として着目されており、がん細胞と腹膜微小環境に存在する腹膜中皮細胞やその他の免疫系細胞との細胞間相互作用が腹膜播種の形成・進展に重要な役割を担っていることが明らかになっている。本研究において、大気圧プラズマの抗腫瘍活性を液体化したプラズマ活性液が誘導する腹膜微小環境における腫瘍抑制機構を解明し、新規治療技術としての基礎的知見の集積を目指す。
    卵巣がんは腹膜播種を伴った進行がんとして発見される場合が多く、故に難治性であり、新規の治療法が求められている。一方、大気圧プラズマの抗腫瘍活性を汎用性の高い液体化したプラズマ活性液は、これまでの抗腫瘍療法とは全く異なる作用機序でがん細胞を殺傷することが示唆されている。本研究では、腹膜播種の形成・進展において重要な役割を果たす腹膜中皮細胞・免疫系細胞を介した腹膜微小環境におけるプラズマ活性液の抗腫瘍効果を明らかにし、新規治療技術としての基礎的知見の集積を目的とする。
    腹膜播種は、がん細胞のみならず本来なら恒常性維持に関わる細胞が、癌関連腹膜中皮細胞、腫瘍浸潤リンパ球、腫瘍関連マクロファージ等に変化し、難治性・治療抵抗性の基盤となって存在している。一方、これまでの研究において、プラズマ活性液の腹腔内投与は、マウス卵巣がん腹膜播種モデルにおいて、腹腔内へのがん細胞の播種を抑制し、マウスの生存期間を有意に延長すること明らかにしてきた。そこで、本研究において、プラズマ活性液による腹膜微小環境への影響を介した播種抑制効果を明らかにするために、がん細胞の播種巣、特に大網組織中のがん微小環境に着目し、組織中に存在するマクロファージとその特徴について解析を行った。その結果、がん細胞の播種3日後には治療対象群のマウス大網組織において、がん細胞の播種および増殖が確認されたのに対し、プラズマ活性液治療群では、がん細胞の増殖が有為に抑制されていることが明らかとなった。組織学的解析の結果、顕著なM1型マクロファージの浸潤が見られ、抗腫瘍免疫が活性化された可能性が示唆された。これらの結果より、初期のがんの播種に対しプラズマ活性液が免疫応答に変化をもたらすことでがんの進展を抑制する可能性を示した。
    コロナ渦における、実験実施の制限や、物品などの入手が一部困難であった。
    今年度の研究成果により、プラズマ活性液が、がん微小環境に抗腫瘍免疫応答を誘導する可能性を示した。しかし、がんの進展に対する腹膜微小環境は種々の細胞が存在し、それらが相互に影響し活性化や分化を誘導している。次年度以降は、中皮細胞やリンパ球にも焦点をあて、正常腹膜とがん性腹膜での微小環境の違いに着目し、プラズマ活性液が、がん細胞が生きづらい微小環境へと変化を誘導することができるかどうかを、培養中皮細胞並びにリンパ球細胞を用いて検討する。一方で、プラズマ活性液によりがん細胞に誘導されたオートファジー細胞死が、腫瘍内免疫原性細胞死を誘導するかどうかについても検討を行い、がん細胞を介した間接的な免疫活性化の効果とそのメカニズムについても明らかにしていく。

  5. 大気圧プラズマのケロイド・肥厚性瘢痕に与える影響に関する基礎的研究

    ケロイドや肥厚性瘢痕は、拘縮・疼痛や醜状など患者の負担は大きく、また治療難渋する症例も少なくなく、新たな治療法の開発が期待されている。近年、大気圧プラズマ研究の進歩により、細胞増殖・分化などに対する様々な影響が報告され始めた。その中で、筋線維芽細胞への分化やその増殖を、大気圧プラズマが制御できないか、という着想に至った。本研究では、①大気圧プラズマのラジカル分布パターン解析、②大気圧プラズマの皮膚創傷治癒関連細胞に与える影響の解析、③ケロイド・肥厚性瘢痕動物モデルで、大気圧プラズマによる発症・進展予防や治療の効果検討を行い、大気圧プラズマによるケロイド・肥厚性瘢痕に与える影響について検討する。
    本年度は、細胞レベルでの大気圧プラズマの肥厚性瘢痕・ケロイドに対する影響を調べるため、ケロイド・肥厚性瘢痕組織において重要な挙動を示す線維芽細胞に引き続き焦点を当てた。昨年市販のヒト正常真皮由来およびケロイド由来線維芽細胞の比較で、細胞増殖は両細胞とも希釈するに従い亢進し、1/32倍以下の濃度では一定となった結果を得ていた。本年度はプラズマ照射液の培地に添加する濃度の最適化をめざし、多くのドナー由来細胞で検討しようと計画したが、COVID19感染症蔓延のためドナー細胞収集に難渋している。現状で得られたサンプル数の細胞では1/4から1/128と結果のバラツキが大きく、条件の至適化ができていないのが現状である。
    ケロイド・肥厚性瘢痕動物モデルで、大気圧プラズマによる発症・進展予防や治療の効果検討を行うため、その作成を行った。昨年度に行った肥厚性瘢痕モデル(肥厚性瘢痕の深部を半分切除、皮下ポケット内に移植、2週後に移植片上の皮膚を切除)を継続的に作成した。ケロイドモデルに関してはCOVID感染症蔓延の影響か、ドナーが1名もおらず作成できなかった。プラズマ照射した培地の2倍希釈系列を作成し、これを肥厚性瘢痕モデルに注入した。注入後1ヶ月では、色調や肥厚に関して差を認めなかった。また2ヶ月後では移植した肥厚性瘢痕自体が潰瘍化から壊死に陥り、観察継続が困難であった。動物モデルの再検討を要する状態である。
    正常線維芽細胞増殖を抑制せず、筋線維芽細胞増殖を抑制するプラズマ照射液の条件がドナーや細胞腫各ごとに異なり、ドナーの確保が最重要課題であったが、COVID19感染症蔓延のためドナー数確保が困難を極め、現時点で最適化できていない。俣長期経過観可能なモデルも再検討を要する状態であり、実験が計画通りに進行できていない。
    まずは研究分担者らと検討の上、本年度以上のプラズマ照射培地を作成し、ヒト正常線維芽細胞の増殖を抑制せず、筋線維芽細胞の増殖を抑制するような条件を検討し、プラズマ照射培地の最適化を継続して行う。その際ドナーのリクルートに今年以上に注力する。その上で、申請書の計画に沿って行う。

    Low-temperature plasma has been studied for medical applications. We had focused on the plasma cancer treatments, and we have previously invented plasma-activated solutions. In this study, we used plasma-activated medium (PAM) and plasma-activated Ringer’s lactate solution (PAL) for plasma cancer treatments. We found that PAM induced oxidative stress-dependent cell death on glioblastoma brain tumor cells while PAL induced oxidative stress-independent cell death on glioblastoma cells. These results suggest that plasma-activated solutions might be useful for cancer drug resistant cancer therapy.

    In order to cultivate biotechnology devices and plasma bioelectronics that can control adhesion, morphology and differentiation induction of cells by systematic elucidation interaction between electronic properties developed from nanosheet-edges in carbon nanowall (CNW) and cells, the effects of superimposed electrical stimulation on the CNW scaffold on the proliferation / differentiation (ossification) of osteoblasts were elucidated. Proliferation promotion and suppression of ossification were specifically confirmed only when the electrical stimulation at the frequency of 10 Hz was superimposed. Furthermore, the dependence on wall density and the formation of cell clumps without ossification were also found. These suggest changes in cell proliferation and differentiation due to the synergy between the sheet edge on the CNW surface and electrical stimulation, and establishment of new cell control technology in various regenerative medicine is expected.

    We have previously developed plasma-activated medium (PAM) that is plasma-treated medium for cancer treatments. In this study, we collaborated with department of obstetrics and gynecology and department of gastrointestinal surgery, and have developed plasma-activated Ringer’s lactate solution to treat with ovarian, gastric, and pancreatic cancers which are peritoneally disseminated. In addition, we collaborated with biochemical engineering lab, and found that PAM selectively eliminates undifferentiated human induced pluripotent stem cells. Thus, plasma-activated medium have broadened the applications of plasmas in medicine.

    Recently, medical applications using non-thermal atmospheric pressure plasmas have attracted attention in the field of medicine. We have developed non-thermal atmospheric pressure plasma sources, and applied for cancer therapy. We found anti-tumor effects by plasma treated medium, which we termed “plasma-activated medium or PAM”. In this study, we further developed plasma-activated Ringer’s lactate solution or PAL, and we found that plasma-treated L-sodium lactate is an anti-tumor factor in the PAL. Based on these results, we proposed that PAL is promising as a novel chemotherapy for cancers such as peritoneally disseminated cancers.

    The low-temperature plasma sterilization have advantages for applications. However, complexity of plasma processes arises from the involvement of a myriad of species including: electrons, ions, radicals, and photons. Such plasma components are needed to be measured and analyzed for fine-tuning the process. To determine the best practice for the sterilization, plasma diagnostics were conducted on clarifying the relationship between the control parameters and the plasma parameters.
    Penicillium digitatum spores, which causes green mold on citrus, are inactivated by contribution of extracellular reactive oxygen species (ROS). The stable free radical as semiquinone in the spores decreases during oxygen atom irradiation observed using real time in situ electron spin resonance (ESR). The plasma electrical discharge induce metabolic modification. Also, intracellular metabolites induce the inactivation of spores. These understanding provides the development of plasma sources.

    We found that plasma-irradiated medium induced apoptosis on glioblastoma brain tumor cells, and termed this medium plasma-activated medium (PAM). PAM down-regulated activities of signaling molecules in the survival and proliferation signaling network and induced reactive oxygen species (ROS) on glioblastoma to induce apoptosis. The PAM projects have brought us many collaborative works that include collaborations with departments of Obsterics and gynecology, Gastrointestinal surgery for treatments of peritoneal dissemination of ovarian cancers and gastric cancers, and Ophthalmology for treatments of age-related macular degeneration. We have published 10 papers and applied for 9 patents.

    An innovative approach for producing reactive oxygen and nitrogen species is the use of non-thermal atmospheric pressure plasma. The technique has been applied in a wide variety of fields ranging from the micro-fabrication of electric devices to the treatment of disease. Although non-thermal atmospheric pressure plasmas have been shown to be clinically beneficial for wound healing, blood coagulation, and cancer treatment, the underlying molecular mechanisms are poorly understood. Our researches obtained the current progress in plasma medicine, with a particular emphasis on plasma-activated medium (PAM), which is a solution that is irradiated with a plasma and has broadened the applications of plasmas in medicine.

    Non-thermal plasma has been focused on as a novel medical practice. There have been a number of reports showing the effect of non-thermal plasma on cell death of various cancers. We have succeeded in selective killing of ovarian cancer and glioblastoma brain tumor cells, which are difficult to achieve complete remission under the present therapies, using not only directly but also indirectly plasma referred to as plasma-activated liquids. We have also elucidated that the effects and mechanisms of non-thermal plasma on biological systems using mesothelioma. We have demonstrated that plasma-activated liquids might be more useful for advanced cancers with metastasis and micro-dissemination. On the other hand, for developing medical applications of non-thermal plasma, we have achieved a certain result for regenerative medicine using human adipose tissue-derived stromal cells we established.
    These discoveries have led to the use of non-thermal plasma for alternative clinical applications.
