    うつ病など心の病には社会からのストレスや老化など後天的な要因が深く関わる。我々は、マウスの慢性ストレスが脳内外の炎症反応によりうつ・不安様行動を誘導することを示してきた。一方、慢性ストレスや老化による脳機能障害には個体差があり、レジリエンスの存在が示唆されるが、実態は不明である。本研究では、神経活動・炎症反応の全脳イメージング、一細胞オミクス解析、神経活動・細胞・分子操作など先端技術を駆使し、慢性ストレスや脳機能老化に対するレジリエンスを担う神経回路・脳組織リモデリング・脳末梢連関を解明する。

    本研究では、申請者が見出した、リズミックな応答をとる神経細胞に着目する。この神経細胞を単一細胞レベルで同定するため、遺伝学ツールを用いた解剖学的手法、およびイメージングを行う。解剖学的な情報からこの神経細胞を含む上流下流の神経回路を明らかにする。また、この神経細胞の神経活動を操作し、行動を解析することにより、リズミックな応答の必要性・十分性を検討する。さらに、リズミックな応答の基盤となる分子機構を明らかにすることで、この細胞が行う情報の統合機構の分子・神経基盤を明らかにする。

    本研究課題では、神経発達障害や精神疾患の病態発症機序となる分子・神経回路異常を明らかにし、治療法を開発することを目的として、in vivoおよびin vitroにおける霊長類精神疾患モデルの作製に必要な基盤技術を構築する。静脈注射で全脳感染が可能なアデノ随伴ウイルスベクター(AAV)にCRISPR/Cas9システムを搭載させることで、in vivoゲノム編集を可能にする。加えて、マーモセット胚性幹細胞(ES細胞)から大脳皮質オルガノイドおよび視床オルガノイドを誘導し、それら2領野のオルガノイドを融合させることで領野間神経回路をin vitroで再構成する。

    大脳皮質には機能局在があり、領野毎に特定の機能を有する。大脳皮質視覚野では高次視覚野毎に異なる視覚属性を処理する。大脳皮質の神経回路は、入力された感覚情報を選り分け、異なる情報を異なる脳領域へ並列的に送出する働きを持つ。送出される情報は、情報の送出を担う投射細胞においてシナプス入力の統合処理により生み出される。つまり、投射細胞のシナプス前細胞群の情報統合により、投射ターゲットに固有の視覚応答性を生み出され、その情報を特定の投射部位に伝達すると考えられる。しかし、V1の神経回路がどのような演算により異なる領野へ新しい並列的な出力を生み出すのかは不明である。そこで本研究では、マウス一次視覚野をモデルとして、異なる視覚情報を異なる高次視覚野に伝達する役割を担う微小神経回路の構造と演算機構を明らかにすることを目的とする。
    高次視覚野であるALに投射するV1ニューロンは空間周波数が低く早い動きの視覚情報を、PMに投射するV1ニューロンは空間周波数の高い遅い動きの視覚情報を伝達する。そこでALに投射するV1ニューロンとPMに投射するV1ニューロンの機能を評価する目的で、それら投射ニューロンに逆行性に緑色カルシウム感受性蛍光タンパク質であるGCaMP6を導入した。V1の第2/3層および第5層に分布するGCaMP発現投射ニューロンの視覚刺激に対する応答を2光子顕微鏡イメージングにより評価したところ、それらは方位選択性応答や方向選択性応答を示した。続いて、第2/3層ニューロンと第5層ニューロンの対応関係を明らかにするために、第2/3層および第5層の多焦点面同時イメージングを行った。相互相関を解析することで、第2/3層内の細胞の結合、第5層内の細胞の結合、第2/3層細胞と第5層細胞の層間の結合を評価することができた。以上の結果より、投射特異的に相互結合細胞群の存在が示唆された。

    ウイルスベクターでサブネットワークを解剖学的に評価し、イメージングで機能的なシナプス接続の推定を行っている。今後は、動物の個体数を増やし、再現性の確認を行う。続いて、標識した神経回路の解剖学的知見と生理学的知見との対応付けを行う。さらに、投射先の領野を増やし、投射毎のサブネットワークの構造と機能の違いを評価する。

    大脳皮質は6層構造を有し、第2/3層細胞および第5層細胞が出力細胞として大脳皮質の異なる領野間の情報伝達を担う。第2/3層細胞および第5層細胞が特定の領野に固有の情報を伝達する。そこで、我々は視覚情報伝達における領野特異的かつ層特異的なフィードバック結合の役割を検討する目的で、まずフィードバック出力を担う第2/3層細胞と第5層細胞の機能を同時にイメージングする技術を開発した。2光子顕微鏡を改造し空間光変調器(SLM)を搭載することで、異なる2焦点面の同時イメージングに成功した。続いて、領野特異的かつ層特異的なフィードバック結合を特異的に標識する目的で、大脳皮質の第5層特異的にCreを発現するマウス、およびCre依存的かつFlp依存的なウイルスベクターを用いて、大脳皮質視覚野のLMからV1にフィードバック投射をする第5層細胞に特異的にGFPを導入する実験系を確立した。その後、LMからV1にフィードバック投射をする第5層細胞を特異的に神経活動操作する目的で、化合物リガンドであるCNOの投与依存的に神経活動を活性化できるhM3Dqあるいは神経活動を抑制するhM4Diを、GFPの代わりに搭載したAAVを作製した。その結果、V1へフィードバック投射をするLMの第5層細胞に特異的にhM3DqあるいはhM4Diの発現が認められた。これら高次領野からのV1へのフィードバック結合の役割を明らかにするために、V1へフィードバック投射をするLMの第5層細胞の活動を操作した際に、V1の視覚応答の変化を解析した。リガンドのCNOを投与しながら、V1の応答を2光子顕微鏡イメージングで解析したところ、V1ニューロンの視覚応答性に変化が認められた。層特異的な役割を検討するために、第5層細胞と第2/3層細胞をそれぞれ操作して、違いを比較検討した。以上の結果より、領野特異的かつ層特異的なフィードバック結合の視覚情報処理における役割を明らかにすることができた。

  10. Establishment of recombinant rabies virus-mediated retrograde trancing of neural circuits in zebrafish

    Rabies virus (RV) infects neurons through its glycoprotein (G). Recombinant RV that contains the GFP gene instead of the G gene and is pseudotyped with the retroviral envelope protein (EnvA) was developed for neural circuit tracing. The pseudotyped RV can infect neurons that express TVA (the EnvA receptor)-mCherry. If the G is provided in the same cells, the RV particles are generated and transmitted mono-synaptically to input neurons. We generated transgenic lines that express TVA-mCherry and G in Purkinje cells (PCs) or granule cells (GCs). When PCs were infected with the pseudotyped RV, GFP was detected in GCs and neurons in the inferior olive nuclei. When GCs were infected with the virus, GFP was detected in neurons in various brain regions that receive inputs from the visual, vestibular, lateral line sensory systems, or telencephalon. Our findings validated the RV-mediated retrograde tracing in zebrafish and identified input neurons for GCs in zebrafish.

    The aim of this project is to reveal how the receptive fields of higher-order visual cortical areas (HVAs) are shaped by the thalamus. We identified synaptic input to the V1 and each HVA by injecting retrograde viruses into each visual area of mice. Neurons of the lateral geniculate nucleus (LGN) neurons project to both the V1 and various HVAs, whereas neurons of the lateral posterior nucleus (LP) project to various HVAs, but not to the V1. We imaged visual responses from axon terminals of the LP or LGN neurons expressing GCaMP6 by 2-photon microscopy. In addition, we imaged visual responses from the V1 or HVA while manipulating activity of neurons in the LP or LGN with the Designer Receptors Exclusively Activated by Designer Drug (DREADD) system. From these results, we conclude that LP neurons play a pivotal role in formation of the receptive fields of HVA neurons in mice.
