The project aims to elucidate how the engineered enzyme, phospholipase D (PLD), recognizes and accommodates bulky substrates in a specific orientation to preferably afford certain positional isomers of rare phospholipids, and to understand whether PLD utilizes an alternative catalytic mechanism in the synthesis of cyclic phospholipids.
This research is important since it will provide entirely new knowledge on structure-function relationship of PLD, valuable for the synthesis of bioactive, but otherwise unavailable, phospholipids. Since enzymatic synthesis is advantageous to other methods in terms of cost, yield, and specificity, we expect increased availability of rare and unnatural phospholipids for both, research and industry. This will foster further discovery into the roles and functions of phospholipids in the human body and will assist in drug development, since the same catalytic mechanism of PLD is shared among many enzymes with key functions in humans and microorganisms, such as endonucleases, ribonucleases, and kinases.
