    本研究課題は、英国リバプール大学の構造生物学研究チームと共同で病因タンパク質の構造生物学的知見に基づいた筋萎縮性側索硬化症(ALS)の分子病態解明と新規治療法の開発を目指す。具体的に、2種類のALS原因タンパク質SOD1、TDP-43に着目し、SOD1については、その立体構造を安定化することにより疾患の進行を抑制する化合物の開発を行う。TDP-43については、ALSで見られる病理像の鍵となるTDP-43タンパク質の構造異常を英国チームと共同で解析する。本国際共同研究を通じて、新たな視点でのALSの病態解明、治療法開発や、異分野を俯瞰する国際的に視野の広い若手研究者の育成が期待される。
    本研究課題は、英国リバプール大学と共同で構造生物学的知見に基づいた筋萎縮性側索硬化症(ALS)の分子病態解明と新規治療戦略の開発を目指す。具体的に、1) ALS原因タンパク質SOD1の立体構造を安定化させて疾患の進行を抑制する化合物の開発、2) 単量体TDP-43の分子の生物物理学的性状解析と立体構造解析を行う。SOD1の構造安定化を目標に、英国チームが合成した化合物について、本邦チームが英国にて共同で培養細胞を用いたスクリーニング・構造解析を行い、神経保護効果を細胞やモデルマウスを用いて検証する。また、TDP-43タンパク質の解析について、本邦チームによる知見である、ALS病理の鍵タンパク質TDP-43の異常蓄積や核外漏出と蛋白の構造異常の関係性に着目し、この分子機構をタンパク質の物理化学的特性ならびにクライオ電子顕微鏡等による立体構造解析から解明する。
    2019年度は、本研究課題の採択に際して、英国チームとWeb会議、海外学会等において研究計画の詳細な打ち合わせを行った。1)ALS原因蛋白質SOD1の立体構造を安定化させて疾患の進行を抑制する化合物の開発について、英国チームが見出した候補化合物であるエブセレンについて、構造展開によって類縁化合物数種を合成し、その神経保護効果について、培養細胞における予備検討を開始した。2) 単量体TDP-43の分子の生物物理学的性状解析と立体構造解析について、今年度は、TDP-43の単量体化により、ALSに特徴的なTDP-43病理であるTDP-43タンパク質の局在異常とリン酸化TDP-43の異常凝集が惹起されることを見出した。
    また、英国における実験実施のため、英国への渡航を予定していたが、COVID-19蔓延のため、次年度以降に延期となった。その間、Web会議、e-mail等によって共同研究に関わる打ち合わせを継続している。
    COVID-19の蔓延により、英国への渡航が延期になったため、一部の実験計画は延期している。
    双方の研究チームがWeb会議等によって密に連携して、それぞれの担当課題を進めるように努める。ただし、COVID-19による大学の活動制限が再度発生した場合には研究計画の変更も検討せざるを得ない。

    神経変性疾患の病態研究では、神経細胞のみに着目するのではなく、グリア細胞等を含めた神経系における細胞間ネットワークの破綻という観点からの病態解明の重要性が近年認識されている。筋萎縮性側索硬化症(ALS)の一部は遺伝性に発症し、その原因遺伝子の一つとしてTBK1 (TANK binding kinase 1)が最近同定された。TBK1は、免疫系のシグナル制御分子であるとともに、オートファジーにも重要な役割を担うことが知られており、既報によるとTBK1の機能喪失がALSの発症に関わると考えられるがそのメカニズムの詳細は不明である。本研究では、TBK1の機能喪失を再現する新規ALSモデルの作成と解析を通じて、神経炎症の観点から、TBK1異常による神経変性メカニズムの解明を目指す。
    昨年度までに、TBK1ノックアウトマウスの作成を完了することができた。二重欠失マウス(TBK1-DKO)は出生し、約10ヶ月齢まで生存し、明確な表現型に乏しい。現在、長期にわたる表現型・行動解析を行なうとともに、別の遺伝性ALSモデルであるSOD1変異マウスとの交配実験を開始しており、SOD1モデルにおけるTBK1欠失の影響を解析する計画である。さらに、グリア細胞におけるTBK1の役割を解明するため、オリゴデンドロサイトをはじめとしたグリア細胞の初代培養系を用いて、TBK1欠失による表現型、分子プロファイルの変容について詳細に解析を行なった。
    TBK1の異常を再現したモデルマウスおよび初代培養細胞の解析が進捗している。
    次年度も予定通り実験計画を遂行する。

    本研究では、免疫学的背景の異なる2種類のSOD1変異ALSモデルマウスを作成して、それらの分子病態を比較することを通じて、神経変性疾患の病態と末梢免疫反応との連関の解明を目的とする。
    末梢免疫背景の異なる細胞性免疫(Th1)優位なC57BL/6系統、及び液性免疫(Th2)優位なBalb/c系統の2種類のALSモデル(以下G93A(B6)、G93A(Balb))マウスを作成し、生存解析を行った。G93A(Balb)マウスの平均生存期間は、G93A(B6)マウスに対して有意に短縮した (G93A(Balb): 199.1 days; G93A(B6): 224.6 days, n=17-18)。次に、病態への関与が予想される遺伝子群の脊髄における発現を比較し、G93A(Balb)マウスにおいてケモカインや神経栄養因子の発現が低いことを見いだした。また、G93A(Balb)マウスでは、G93A(B6)と比較して免疫細胞の脊髄内浸潤がほとんど起こらず、脊髄内の活性化ミクログリアの細胞数の低下がみられた。その機序の一端として、発症後のG93A(Balb)マウスにおいて、ミクログリアの細胞死が亢進していた。一方、遺伝的背景による影響を検証するため、C57BL/6マウス及びBalb/cマウス由来の初代培養ミクログリアを比較したが、細胞増殖率に差は見られなかった。さらに、血清中の免疫・炎症関連分子の発現解析を行ったところ、G93A(Balb)マウスの末梢血におけるケモカインやある種のサイトカインの低下傾向を認めた。
    以上の結果から、G93A(B6)マウスとG93A(Balb)マウスにおけるミクログリアの表現型や病態の差異は遺伝的背景によるものではなく、免疫反応など神経組織外の環境に由来することが示唆された。

    Using model mice for amyotrophic lateral sclerosis (ALS), we identified cytokine TGF-β1 as a detrimental factor for ALS by interfering a neuroprotective environment mediated by glial cells. TGF-β1 was abnormally upregulated in astrocytes of human ALS and a mouse model for ALS. Administrating TGF-β1 inhibitor slowed disease progression in ALS mice, indicating TGF-β1 as a potential therapeutic target for ALS. In addition, we elucidated a novel role of innate immune adaptor TRIF in eliminating abnormally activated astrocytes through inducing apoptosis.

    Various chronic inflammatory diseases including lifestyle-related diseases eventually result in tissue fibrosis, which has major unmet medical needs. In this study, to understand the molecular mechanism underlying NASH, we employed genetically obese melanocortin-4 receptor-deficient mice, which sequentially developed simple steatosis, NASH, and hepatocellular carcinoma (HCC) when they were fed a Western diet. We found that tissue-resident macrophages in the liver or Kupffer cells aggregate around dead or dying hepatocytes to form a unique histological structure termed crown-like structure, thereby accelerating liver fibrosis. Interestingly, these Kupffer cells showed a phenotypic change to obtain the profibrotic property. We also found that MC4R in the hypothalamus is capable of regulating chemotactic activity of macrophages in the liver, particularly in the resolution phase.
