    アスベスト線維の国内消費量と悪性中皮腫発症患者数の相関が疫学的に示され、本邦で患者数の増加が危惧されている。中皮腫の発症にメチル化やゲノム安定性に関連する分子が重要であることが明らかになって来ている。研究代表者が所属する研究室で、種々の線維状発がん物質を用いBrown-NorwayとF344のF1 hybrid ratに腹膜中皮腫を作成してきた。これらの凍結保存検体やパラフィン包埋切片を用いてマイクロサテライト解析を行うことで、LOH(loss of heterozygosity)やホモ欠失の有無を検討する。新しい中皮腫感受性領域の有無を検討し、中皮腫の治療法開発の基礎となることを目的とする。

    Transport of iron is carried by divalent metal transporter 1(DMT1) in mammalian cell. In this grant, K562 was employed as an erythroid precursor model to study DMT1 Associated Protein(DAP). An analysis of DAP amino acid sequence revealed the binding domain to PBR. Protoporpyrin IX(PPIX) is presumed to bind with PBR, suggesting the interaction between DAP and PPIX via PBR. The expression levels of DAP, DMT1, transferrin receptor 1(TfR1), ferritin heavy chain(FTH) were examined after exposure of PPIX. PPIX decreased the protein level of DAP and DMT1 at 1 h. mRNA level of DAP was induced at 8 h and protein level was recovered at 24 h, while, mRNA of DMT1 was not increased until 24 h after PPIX exposure. C/EBPa, which transcribe DMT1 mRNA, were decreased by PPIX treatment. PPIX-induced degradation of DAP, FTH, DMT1 was presumed to be carried in lysosome and proteasome. The combination of lysosomal and proteasomal inhibitor were most effective way to prevent the degradation of FTH.

    This project was planned to establish a basis for novel cancer prevention by evaluating iron dynamics multidimensionally. We established the concept of ferroptosis, as catalytic Fe(II)-dependent regulated necrosis, based on our experience of iron-induced carcinogenesis. We used phlebotomy to remove excess iron in crocidolite-induced mesothelial carcinogenesis, which significantly prolonged survival and reduced tumor burden. We used knockout (KO) mice of 3 repair genes for 8-oxoguanine (Ogg1, Mutyh and Mth1) and found that Mutyh KO showed a tendency to increase iron-induced renal carcinogenesis whereas Mth1 KO significantly reduced crocidolite-induced malignant mesothelioma (MM) in females. MM induction by crocidolite was delayed in Dmt1 transgenic mice. We also found that carbonic anhydrase 9 (CA9) confers ferroptosis-resistance to MM cells in hypoxia. Thus, CA9 can be another target for cancer therapy. In conclusion, iron metabolism regulates not only carcinogenesis but tumor biology.
