    <p>Background: Flavonoids form a diverse class of naturally occurring polyphenols ascribed various biological activities, including inhibition of amyloid β (Aβ) fibrillisation and neurotoxicity. In this study we evaluated the neuroprotective and anti-aggregative properties of the flavones transilitin, quercetin and the novel semi-synthetic flavone and SUMOylation inhibitor, 2-D08 using neuronal PC-12 cells exposed to Aβ1-42 and in mouse cortical neural stem cells (mNSC).</p><p>Methods: Flavone effects on neuronal viability were assessed morphologically using live cell imaging and morphometric analysis in mNSC, and biochemically in PC-12 cells using the MTT assay following incubation with Aβ (0-2 microM), alone or in the presence of each flavone (10-100 microM) for 48 hr. Effects of each flavone on Aβ fibril formation and aggregation were directly assessed using the Thioflavin T (ThT) fluorometric assay and transmission electron microscopy (TEM) over 48 hr.</p><p>Results: 2-D08 and transilitin (10-100 microM) evoked concentration-dependent inhibition of neurotoxicity elicited by Aβ (0-2 microM). Quercetin was neuroprotective at the low concentration (10 microM), but evoked intrinsic neurotoxicity at higher concentrations (50-100 microM). Morphometric imaging analysis demonstrated that 2-D08 and transilitin were both non-toxic to mNSCs and confirmed that quercetin was neurotoxic at the higher concentrations. The neuroprotective effects were associated with a marked inhibition of Aβ fibrillisation, as evidenced by attenuated ThT fluorescence kinetics and microscopic evidence of both altered and diminished Aβ aggregate and fibril morphology.</p><p>Conclusions: These findings highlight the neuroprotective role of select flavones on Aβ-mediated toxicity associated with an inhibition of Aβ fibrillisation. In particular, the efficacy of the novel flavone 2-D08 in this regard may direct further lead development with multiple biological actions targeting neurodegeneration in Alzheimer's disease.</p>

    <p>Currently, various protocols have been developed to differentiate stem cells into target cells for applications in regenerative medicine and drug discovery. In many protocols, factors to control cellular differentiation are now replaced by small chemical compounds by their stable and cost- effective performance compared to protein-based factors. There are also reports that indicate such differentiation controlling compounds can be used as medical drug seed molecules. Therefore, great attention is attracted in screening chemical compounds that stimulate stem cells either negatively/positively. However, the evaluation of such compounds for stem cell culture is time-consuming, costly, and cell-invasive (endpoint assay). In our group, we have been applying image processing and bioinformatics analysis for quantitative evaluation of cellular phenotypic changes, such as evaluation of cellular quality during their culture process. In this study, we have applied this evaluation technique to assay the stem cell response to the small compounds rapidly, cost-effectively, and non-invasively (real-time assay). By converting the phase contrast microscopic images of cells into multiple parameters, designated as "fingerprint", we found that cellular response to positive/negative stimulating compounds could be quantitatively assessed, and in some conditions, their differentiation controlling potential could be screened in extremely early stage. Our image-based early evaluation method demonstrates high potential to be applied for screening useful compound that enhance stem cell culture.</p>

    <p>Alzheimer's disease (AD) is a progressive and chronic neurodegenerative disease characterized by the presence of Beta-Amyloid Protein (Abeta) aggregates in the brain. Macroalgal-derived phlorotannins (PT) are polyphenolic compounds that resemble plant flavonoids known to be neuroprotective in Alzheimer's disease models. In this study we characterized the anti-aggregatory and neuroprotective effects of a phlorotannin extract from Ecklonia radiata against amyloid-beta (Abeta) aggregation and neurotoxicity. E. radiata was sourced fresh from regional coastal wrack (Rivoli Bay, South Australia), where crude PT extract was produced via solid-liquid extraction and liquid-liquid fractionation to reach a purity of 87% by biomass dry weight. Abeta anti-aggregatory effects of PT extract were subsequently evaluated using the thioflavin T assay of Abeta kinetics and transmission electron microscopy (TEM) to directly visualise Abeta fibrils. Neuroprotection was assessed in neuronal PC12 cells exposed to Abeta1-42 for 48 hrs in the presence of PT extract (0-100 microg/ml) and in mouse cortical neural stem cells (mNSCs) exposed to extract alone to assess its intrinsic properties. The PT extract demonstrated significant anti-aggregative properties, inhibiting fibril formation at 50 and 100 microg/ml (P<0.05) and inhibiting Abeta-induced toxicity in PC12 cells from 10 microg/ml. At <10 microg/ml, PT extract elicited differentiation of mNSCs, but at higher concentrations (>40 microg/ml), Ecklonia extract was neurotoxic in mNSCs. Further refinement and analysis of the extract revealed a novel constituent phlorotannin that may aid in the development of novel therapeutic treatments in Alzheimer's disease.</p>

    In recent years, cell and tissue therapy in regenerative medicine have advanced rapidly towards commercialization. However, conventional invasive cell quality assessment is incompatible with direct evaluation of the cells produced for such therapies, especially in the case of regenerative medicine products. Our group has demonstrated the potential of quantitative assessment of cell quality, using information obtained from cell images, for non-invasive real-time evaluation of regenerative medicine products. However, image of cells in the confluent state are often difficult to evaluate, because accurate recognition of cells is technically difficult and the morphological features of confluent cells are non-characteristic. To overcome these challenges, we developed a new image-processing algorithm, heterogeneity of orientation (H-Orient) processing, to describe the heterogeneous density of cells in the confluent state. In this algorithm, we introduced a Hessian calculation that converts pixel intensity data to orientation data and a statistical profiling calculation that evaluates the heterogeneity of orientations within an image, generating novel parameters that yield a quantitative profile of an image. Using such parameters, we tested the algorithm's performance in discriminating different qualities of cellular images with three types of clinically important cell quality check (QC) models: remaining lifespan check (QC1), manipulation error check (QC2), and differentiation potential check (QC3). Our results show that our orientation analysis algorithm could predict with high accuracy the outcomes of all types of cellular quality checks (>84% average accuracy with cross-validation).

    As the number of available cell types grows, it becomes necessary to develop more effective ways to optimize the cell-culture medium for each cell line and culture condition. However, because of the vast number of parameters that must be decided, such as the combination of components, optimization is both laborious and costly. Microdevices are a cost-effective way to perform such evaluations because they use only a small volume of media and enable high-throughput analyses. However, assays performed in microdevices are themselves minimized, and each assay unit (well/chamber) commonly contains an insufficient number of cells for comprehensive evaluations such as gene-expression or flow-cytometry analyses. To address this issue, we introduced image-based analysis in conjunction with microdevice assays; this approach allows quantification of every cell in each assay unit. To quantitatively profile differences in cellular behaviors in a microdevice under different culture media conditions, we developed a non-staining image-based analysis method that utilizes cellular morphology. Our approach combines the structural advantages of microdevices, which can increase the stability of images, and the quantitative advantages of an image-based cell evaluation technique that utilizes time-course population change in several morphological features. Our results demonstrate that cellular changes due to small alterations in the concentration of serum in medium or differences in the basal medium can be profiled using only microscopic images.

    Despite the growing numbers of successful applications in regenerative medicine, biotechnologies for evaluating the quality of cells remain limited. To evaluate the cultured cells non-invasively, image-based cellular assessment method holds great promise. However, although there are various image-processing algorithms, very few studies have focused to prove the effectiveness of phase contrast images with risk assessment example that reflects actual difficulties in regenerative medicine products. In this study, we developed a simple image-processing method to recognize the number of dividing cells in time-course phase-contrast microscopic images, and applied this method to assess the irregular proliferation behavior in normal cells. Practically, as a model, rapid proliferating human fibrosarcoma cells were mixed in normal human fibroblasts in the same culture dish, and their sarcoma existence was evaluated. As a result, the existence of sarcoma population in normal cell sample could be feasibly detected within earliest period of cell culture by their irregular rise of accumulated counts of dividing cells. Our image-processing technique also illustrates the technical effectiveness of combining intra-frame and inter-frame image processing to accurately count only the dividing cells. Our concept of focused counting of dividing cells shows a successful example of image-based analysis to quickly and non-invasively monitor the regular state of regenerative medicine products.

    Although regenerative medicine is rapidly evolving toward clinical application in recent years, the fact that no effective technique to non-invasively and objectively monitor the state of cultured cells is existent hampers its realization. For this problem, an image processing/recognition technique using phase-contrast microscopy images acquired non-invasively, which first segments individual cells and then quantitatively evaluates the quality of each cell morphologically, will be a solution. However, for confluent cells it has been ineffective because the confluent cells have no decisive boundary, and then each cell cannot be segmented and characterized by the morphological features. In this research, we devise a method to quantify the state of confluent cells by scoring each pixel based on the collective feature in terms of orientation and then statistically characterizing the heterogeneity of the score distribution throughout an image. We demonstrate the efficacy of the proposed method by classifying actual microscopic images of confluent cells into proper classes using a support vector machine based on the feature derived from the orientation and heterogeneity.

    Based on the hypothesis that development of cow's milk allergy (CMA) in children can diagnosed from species of IgE epitope in patients, significant peptide epitope for IgE was analyzed by comprehensive combination analysis using the graphic assessment method. The Adaboost algorithm was used for classification of decision stump as weak classifier, and 6044060 models were constructed. Classification accuracies by 10-fol CV (10-fold cross-validation) were determined. Total distribution of accuracies was depicted and it was analyzed by Gaussian mixture distribution using the EM algorithm. It was found that the distribution could be divided into 5 sub-distributions by the Bayesian information criterion. The Gaussian sub-distributions were grouped into &ldquo;significant sub-distributions&rdquo; and &ldquo;non-significant sub-distributions&rdquo; and classifiers that belong to &ldquo;significant distribution&rdquo; were identified. Analysis of the assignment frequency of individual inputs assigned to the &ldquo;significant distribution&rdquo; identified 16 peptide epitopes with high assignment frequency, including aS1C30 and aS1C47 peptides. Since SERYLGYL from aS1C30 has already been reported as significant peptide epitope for CMA diagnose, our proposed method, CC-GAM, was suggested to be a useful tool for selection of significant epitopes.

To design such polymer by learning the mechanism of native extracellular matrix (ECM), we had investigated ECM-specific peptides with cell-selectivity to control the adhesion balance of endothelial cells (ECs) and smooth muscle cells (SMCs). From our high-throughput comparative screening using <i>in silico</i> analysis and peptide array screening platform, we had found that there are various types of EC-selective and SMC-selective peptides in human collagen type IV. In this report, we mapped the distribution of obtained EC-and SMC-selective peptides in the collagen, and extracted a new design policy of cell-selective polymers for medical implant surface modification.<br>

    We preliminarily constructed a software system to automatically segment cellular regions from phase-contrast microscopic imagery and then to extract the cellular shape features at high speed. Generally, it is difficult to segment cellular regions from phase-contrast microscopic images, because of halo around object and inhomogeneous background, while phase-contrast microscopy is suitable for real-time cellular monitoring. First, we proposed a segmentation method with morphological filters. Then, we devised fast algorithms to calculate cellular features based on chain-code acquired from the segmented image. Finally, we evaluated the performance of proposed method using F-value by comparing the result by the proposed method with the result obtained manually by an expert.

    The effects of freezing treatments on the activity of the lactobacilli-aggregating peptide QRCVNLQA were studied. The aggregation activity of the peptide solution was enhanced by rapid deep freezing in liquid nitrogen followed by storage for at least 1 d below -10°C. The treated peptide strongly induced the aggregation of suspended cells of <i>Lactobacillus rhamnosus</i> GG to provide a clear supernatant after 1.5 h. The peptide was activated by oxidation of thiol groups, probably through dimerization of the peptide molecules to form disulfide bonds. Addition of the freeze-oxidized peptide to a suspension of lactobacilli promoted the adhesion of the lactobacilli to a monolayer of Caco-2 intestinal epithelial cells.

    骨切開を伴う手術では骨切断面から多くの出血を伴うため、一般的にはボーンワックスで骨切断面にパッキングする止血法が用いられている。しかしボーンワックスは非分解・非吸収性で、残存による骨癒合・治癒阻害や術後感染等の合併症、骨癒合・再生の遅延を助長する可能性がある。従って、新たな骨髄止血材料の開発は多くの患者に有益である。申請者らは、独自技術で開発したポリカプロラクトン(PCL)をベースとした生体吸収性ポリマーと骨再生誘導ペプチドによる新規骨髄止血材料を創出してきた。本研究では、材料の質的向上を図るため、各設計条件を見直す基礎研究を行い、前臨床研究も含めた動物実験にて有用性を検証する。

    大動脈瘤に対する人工血管置換術は侵襲が大きく、新たな低侵襲な治療法の開発が望まれる。我々は、これまでに間葉系幹細胞(MSC)静脈内投与による大動脈瘤治療の有効性を示してきた。治癒メカニズムにはMSCのパラクライン作用が示唆され、MSC産生因子の中に抗炎症作用・組織修復に関わる因子としてProgranulin(PGRN)およびSLPIを同定し、それらによる大動脈瘤治療効果の可能性を見出した。
    本研究は、リコンビナントタンパクPGRN, SLPI投与による効果検証を行ったのち、合成ペプチド医薬として大動脈瘤治療の検討を行い、ペプチド医薬としての有効性を明らかにする。

    ヒトが摂取した食品たんぱく質は、消化管での加水分解の後、ペプチドもしくはアミノ酸として小腸上皮細胞より吸収される。特にペプチドの吸収効率はアミノ酸よりも高く、窒素源の吸収実体として中心的な役割を果たしている。ヒトにおいて体内へのペプチド吸収を担う輸送体は、プロトン共役型オリゴペプチドペプチド輸送体(POT)ファミリーのhPEPT1、hPEPT2であり、8,400種類のジ・トリペプチドが、小腸ではhPEPT1を介して吸収され、腎臓ではhPEPT2を介して原尿中から再吸収される。膨大なバリエーションの基質群を認識するPOTの“基質多選択性”はペプチド栄養の根幹であるが、その詳細は未だ十分には解明されていない。本研究では、申請者らが開発したペプチド輸送体(POT)解析システムを基盤技術として、ヒト小腸での食品由来ペプチドの吸収を担うhPEPT1について、8,400種類のジ・トリペプチドの網羅的吸収特性解析を実施し、“基質多選択性”の全貌を解明する。2018年度にはPOT発現細胞を構築した。しかしその一方で我々が開発したPOT解析法は蛍光ペプチドの取り込みの競合阻害を原理とする間接的な輸送能評価系である。そこで2019年度は2018年度に化学合成によって調整したジ・トリペプチドを用いて各種真核生物由来POTの網羅的親和性解析を進めるとともに、実際にペプチドの輸送効率を調べるため、ジペプチドの網羅的解析法(ジペプチドミクス)を確立した。ジペプチド分析にはLC/MS/MSを用い、誘導体化した合成ジペプチドをサンプルとして網羅的な定量解析が可能なこと、また混合物サンプルを用いた場合でも成功裏に定量解析できることが示された。
    これまでにPOT発現細胞、POT解析法、また実際の輸送を調べるためのジペプチドミクスの開発に至った。2018年度は実験試薬の調達の遅れが生じたものの、ここまでの段階で目標達成に必要な解析系や素材はほぼ揃ってきていることから、現時点での研究進捗状況としては概ね順調と評価した。
    本年度の研究では、ヒトを含めた真核生物POTのin silico親和性予測モデルの構築と、基質認識メカニズムの解析を進める。具体的には、ペプチド群のPOT親和性解析データを重回帰分析する際、ペプチド全体を一つの分子として捉え、“各空間座標における物理化学的因子(“OH基の数”や“logP値”等)の総和”をアルゴリズムとした、汎用性の高いin silico基質親和性予測モデルの構築を検討する。また基質認識メカニズムとして、POTへの結合配向についても解析する。

    Fabrication of elastic modulus controlled honeycomb films was successfully realized and applied them to the substrates for cultivating hMSC, iPS, and neuron cells with controlling their adhesion, proliferation, and differentiation. By using those data, extraction of conditions for controlling differentiation of stem cells was achieved. Furthermore, theoretical simulation based on coupled Cahn-Hilliard equation revealed the nanoscale structural control of polymer particles, and inorganic-organic composite particles with nanostructures were successfully prepared. By using magnetic nanoparticles and gold nanoparticles as composite materials with polymers, nanostructured composite particles were successfully prepared by self-organization process and applied them to site selective Raman scattering measurement probes.

    Peptides that have biological activities are called functional peptides. Many types of functional peptides have been found to date and has been designed for development of peptide drug. In the present study, following 3 topics were investigated, 1) fabrication of peptide library including unnatural amino acid, 2) screening of intracellular functional peptide conjugated with cell penetrating peptide (CPP), 3) development of the smart machinery for removal of CPP in intracellular space. Related to the last topic, we have established the methodology for disulfide bond formation of all peptides on peptide array. In addition, we have been developed a novel synthesis method of hetero-dimerized peptide using intramolecular disulfide bonding between main- and sub-chain of lysin.

    Aortic aneurysm (AA) is a life-threatening disease associated with risk of aortic rupture. AA is caused by atherosclerosis and chronic inflammation, which contribute to expansion of the aorta. We have reported that ATII-induced AA model mice that received intravenous injection of MSCs exhibited regression of AA via immunomodulation, anti-inflammatory and tissue repair properties, mediated by paracrine effects. To identify soluble proteins secreted from MSCs, the conditioned medium of MSCs was analyzed using a protein array. We identified Progranulin (PGRN) and secretory leukocyte protease inhibitor (SLPI) which participate in anti-inflammation. Intraperitoneal injection of recombinant protein PGRN and SLPI exhibited regression of AA. Using our original peptide array technology, we discovered a short-chain peptide derived from amino acids sequence of SLPI. We demonstrated that the short-chain peptide had a potency of anti-inflammation in vitro.

    Using peptide array-based interaction assay of solid-bound peptide and anchorage-dependent cells, we listed up some peptides enhancing umbilical cord derived mesenchymal stem cells (UCSCs) differentiation for osteogenesis and chondrogeneses. We modified self-assembling peptide hydrogel with these peptides, and seeded UCSCs into this scaffold. After cultivation, we transplanted them into mice subcutaneous tissue. Samples were harvested, and evaluated concerning to osteogenesis and chondrogenesis with biochemical and immunohistological methods. We identified poor bone-specific and cartilage-specific extracellular matrix formation in each samples. In conclusion, we couldn't induce efficient osteogenesis and chondrogenesis using UCSCs-seeded self-assembling peptide hydrogel modified with peptides we selected.

    New bone formation was significantly increased by hyperthermia using magnetite cationic liposome embedded in alginate beads with an alternating magnetic field (AMF) in a rat tibial defect model. Heating materials were changed to Resovist containing ferucarbotoran and hydroxyapatite scaffold, which have been already approved in clinical use in Japan. Combination materials were grafted in tibial defect of rat and rabbit and subjected to the multiple hyperthermia in AMF. Results demonstrated that newly formed bone was observed in the grafted materials from the periphery toward the center, and more osteoclasts were found in the once a week hyperthermia group compared with non-hyperthermia group.
    Experiments of hyperthermia were performed in vitro using ATDC-5 and MC3T3 cells. Heat stress also induced enhanced alkaline phosphatase expression in cultured osteoblastic cells, MC3T3. On the other hand, heat stress had no obvious effects on chondrogenic differentiation using ATDC5 cells.

  11. A New development of Controlling Stem Cells by Using Honeycomb Scaffolds

    Honeycomb patterned scaffolds have been fabricated by casting solutions of polybutadiene (PB) and polystyrene (PS) containing amphiphiles under humid conditions. Flat films of these materials were also prepared as reference films. By irradiation of UV light, PB honeycomb and flat scaffolds were cross linked. By using an atomic microscope, surface modulus of each sample has been measured. The measurement revealed that surface modulus increased by UV irradiation. Human mesenchymal stem cells were cultured on these scaffolds and we found that adhesion and differentiation of human mesenchymal stem cells were modulated by changing scaffold parameters including surface modulus and pore sizes. These results will open a path to a new development on controlling behavior of stem cells.

    Pluripotent stem cells, such as iPS cells, require highly trained culture skills, therefore the present cell culture production process greatly rely on experts' skills and experiences. To widely distribute the stem cell technology, the quantitative understanding and standardization is essential. In this study, we have developed an image-based profiling method, "Total culture fingerprint", to record and monitor the quality changes during their culture. By our method, we have found that the stress levels and effects on disturbance of iPS cellular quality can be monitored and modeled by real-time analysis of their colony morphologies, which are known to be an indicator of their quality change.

    Cell death inducing peptide was explored from CPP(cell penetrating peptide) conjugated peptide library and effective cell death of MCF-7 was observed for newly designed octamer peptide, LNLIWKLF, in which fifth amino acid, serine, of the original peptide was substituting to tryptophan and it’s activity was approximately 4 times higher compared with that of the original peptide. When serine residue was replaced to hydrophobic amino acid, F, V, and Y, cell death activity was also further increased. In the case of cell-cycle targeting peptide, newly designed peptide with higher activity was also successfully designed. In addition, minimized peptide library for screening was studied based on 4 amino acid groups categorized from 20 amino acids. Using 2 sets of 256 tetramer peptides, IL-2 binding peptide was assigned and high binding role was identified by principal component analysis. It was found that acquired role could be utilized for screening of octamer functional peptides.

    This research was conducted to develop effective and rapid screening method for designing novel cell culture medium for regenerative medicine or medical development. Briefly, we applied the cellular morphological bioinformatics analysis method to numerically convert the cellular response to indicate the surrounding effects from medium. As a result, we found that clustering-based morphological analysis can categorize the medium effects, also with the combination with cellular activity, and established a screening strategy to evaluate the medium additives non-invasively and rapidly. We also found several compounds could stably control the stem cell differentiation through our method.

    (研究の目的)本研究の目的は、革新的な大動脈瘤治療であるステントグラフト内挿術の重大な術後晩期合併症のステント位置移動やそれに伴うエンドリークを抑制するための自己組織化を目指した「インテリジェント化ステントグラフト」の開発である。
    (本年度研究結果)
    (1) ペプチド付与担体の開発:ペプチドインフォマティクスを活用し、単一アミノ酸配列において、組織化を促すと考えられる線維芽細胞の接着・増殖選択性の高い配列を検索(内皮細胞、平滑筋細胞との比較)すると、7残基のチロシンやアルギニンにおいて、線維芽細胞選択性が比較的高いことがわかった。
    ペプチド混和した、ポリグリコール酸、ポリ乳酸の共重合ポリマーを、エレクトロスピニング(E-spin)法を用いて不織布を作製した。この不織布を用いても、細胞選択性は維持されることがわかった。(2)ドキシサイクリン徐放担体の作製:マトリクスメタロプロテアーゼ(MMP)に拮抗して、動脈のリモデリングを抑制するドキシサイクリンを生体吸収性ポリマーに混和し、E-spinで紡糸したところ、ドキシサイクリンは、in vivoで、2週間で約50%を、4週間で約70%を徐放した。この担体は、平滑筋細胞やマクロファージのMMPやサイトカイン分泌に抑制的に働き、エラスチンを維持し、アポリポプロテインEノックアウトマウス動脈瘤モデルおいて動脈瘤進展抑制効果を示した。
    今後これらの技術を応用し、ステントグラフトの作製を試みる予定である。
