    Plant growth and development are sustained by continuous cell division in the meristems, which is perturbed by various environmental stresses. For the maintenance of meristematic functions, it is essential that cell division be coordinated with cell differentiation. However, it is unknown how the proliferative activities of the meristems and the coordination between cell division and differentiation are maintained under stressful conditions. Here, we report that a rice protein, RSS1, whose stability is controlled by cell cycle phases, contributes to the vigour of meristematic cells and viability under salinity conditions. These effects of RSS1 are exerted by regulating the G1-S transition, possibly through an interaction of RSS1 with protein phosphatase 1, and are mediated by the phytohormone, cytokinin. RSS1 is conserved widely in plant lineages, except eudicots, suggesting that RSS1-dependent mechanisms might have been adopted in specific lineages during the evolutionary radiation of angiosperms.

    Crop production is threatened by global climate change, and recent demands for crops to produce bio-fuels have started to affect the worldwide supply of some of
    the most important foods. How can we support a growing human population in such circumstances? One potential solution is the improvement of crops to increase yield from both irrigated and non-irrigated lands, and to create novel varieties that are more tolerant to environmental stresses. Recent progress has been made in the isolation and functional analyses ofIgenes controlling yield and tolerance to abiotic stresses. In addition, promisingnew methods are being developed for identifying additional genes and variants of interest and putting these to practical use in crop improvement.

    DNA repair associated with DNA replication is important for the conservation of genomic sequence information, whereas reconstitution of chromatin after replication sustains epigenetic information. We have isolated and characterized mutations in the BRU1 gene of Arabidopsis that suggest a novel link between these underlying maintenance mechanisms. Bru1 plants are highly sensitive to genotoxic stress and show stochastic release of transcriptional gene silencing and show increased intrachromosomal homologous recombination and constitutively activated expression of AtPARP-2, the induction of which is associated with elevated DNA damage. Bru1 mutations affect the stability of heterochromatin organization without changing in genome-wide DNA methylation. BRU1 encodes a novel nuclear protein with two predicted protein-protein interaction domains. Comparison of bru1 with mutations in genes encoding subunits of CAF-1, the condensin complex, or MRE11 indicates cooperative roles in the replication and stabilization of chromatin structure, providing a novel link between chromatin replication, epigenetic inheritance, S-phase DNA damage checkpoints, and the regulation of meristem development.
