    植物にとって部分冠水による低酸素状態や完全冠水による無酸素状態は死に直結する深刻なストレスである。植物は数分から数日の幅で変動する冠水の長さや強度(程度?)に応じて、多段階に通気組織の発達や茎葉伸長を適切に調節する柔軟な応答機構(段階的なステージゲート応答機構)を持ち合わせている。しかし、植物が環境ストレスの時間や強度をどこでどのように受容し複数の応答からどれを選択し発動するのか?は不明である。本課題では、植物の酸素や水分のストレス感受機構と段階的なステージゲート応答の最適な選択機構を明らかにし、植物の冠水に対するレジリエンス機構の解明を目指す。
    これまでに、浮きイネの冠水依存的な節間伸長はアクセルとして働くACE1とブレーキとして働くDEC1によってantagonisticに制御されていることを明らかにした。そこで、ACE1とDEC1による茎伸長制御が双子葉植物に保存されているかを明らかにするために、まず、ACE1、ACL1(ACE1 like 1)を双子葉のモデル植物であるシロイヌナズナで過剰発現体を作成した。しかしながら、ACE1, ACL1過剰発現体は栄養生殖期には植物ホルモンのジベレリンを投与しても全く節間伸長を示さず、開花期や花茎長においても顕著な差を示さなかった。一方、シロイヌナズのACE1ホモログであるFPF1は花芽分化を促進することが報告されているが、FPF1をイネに過剰発現させ個体にGAを投与しても節間伸長も花芽分化の誘導も観察されなかったことから、イネACE1とシロイヌナズナFPF1は機能分化をしていることが示唆された。
    イネの通気組織形成には、恒常的通気組織形成と誘導的通気組織形成の2種類の制御機構が存在し、それぞれオーキシンとエチレンがトリガー因子となることが明らかになっている。エチレンの制御下で、カルシウム依存性プロテインキナーゼOsCDPK5, 13の両方が、OsRBOHHをリン酸化することによって、通気組織形成を促進することが明らかになり、OsCDPK5, 13が誘導的通気組織形成に必須であることが示唆された。
    また、シロイヌナズナを用いた化合物スクリーニングを行った結果、低酸素時に作動する仕組みを介して胚軸伸長を促進すると期待できる化合物を同定した。この化合物は既知の植物ホルモン類のどれにも似ていない分子構造を持つなどのユニークな特徴を持つことから、その作用機序の解明を通じ、低酸素下の伸長応答に対して従来の理解にはない仕組みを導入できると期待される。
    1.イネのACE1およびDEC1が制御する遺伝子群を明らかにするためACE1およびDEC1過剰発現体や機能欠損変異体のRNA-seqを進めている。DEC1が直接制御する遺伝子の探索を併せて進めており、概ね順調に進捗している。
    2.イネのACE1およびDEC1の機能を明らかにするために、コケ植物のモデルであるゼニゴケやヒメツリガネゴケを用いて過剰発現体を作成中である。またACE1およびDEC1のパラログの過剰発現体や機能欠損変異体も作成中であり、これらの植物の形態と遺伝子発現変化を調査することでACE1とDEC1の機能を探る。それぞれの形質転換体が準じ作成されつつあり、概ね順調に進捗している。
    3.イネにはACE1ホモログが計6つ存在しており、現在それぞれの過剰発現体を作成中である。T1世代の植物を用いて表現型と遺伝子配列比較により機能ドメインを明らかにする予定で概ね順調に進捗している。
    4.イネの2種類の通気組織形成機構の分子レベルでの共通点と相違点を明らかにするために、レーザーマイクロダイセクションを用いて、それぞれの通気組織形成変異体の根の皮層組織の単離を進めている。今後RNA-Seq解析を行い、各々の下流因子の遺伝子同定を行う予定である。また、通気組織形成遺伝子のプロモーターにレポーター遺伝子をつないだコンストラクトを導入したイネ形質転換体作出の計画も概ね順調に進捗している。
    5.シロイヌナズナの胚軸伸長促進化合物に関しては、化合物添加後の網羅的遺伝子発現解析、伸長制御に関わることが既に知られている植物ホルモン経路や光シグナル経路とこの化合物による効果の関係性の解析、この化合物を固相化したアフィニティー樹脂を用いた標的タンパク質の同定、耐性変異体群の獲得とその原因遺伝子の同定、の各方向性の実験を行う準備が概ね順調に進捗している。
    これまで、イネの冠水時における茎伸長は低酸素と植物ホルモンのエチレンがトリガーとなっている可能性が提唱されている。そこで、イネの冠水時における酸素動態を明らかにするために、イネの節間内に酸素電極センサーを取り付け、程度の異なる冠水に暴露したイネ節間内の酸素動態を計測するとともに、節間伸長の計測を行う。またこのとき、エチレン関連遺伝子やこれまでに同定された節間伸長関連遺伝子の発現を同時に解析することで、冠水時における酸素濃度とエチレンおよび節間伸長の関係を明らかにする。イネの節間伸長は、節間に存在する介在分裂組織の細胞分裂によって開始するが、介在分裂組織の位置は大まかにしか示されておらず、その実体となる細胞群と、その制御機構は全く不明である。そこで、この介在分裂組織の実体を明らかにするために、冠水による節間伸長時に細胞分裂が活発になる細胞群を、細胞分裂マーカーを用いて同定する。また、介在分裂組織の維持に関わると予想されるWOX遺伝子群の発現動態を観察するとともに、WOX遺伝子を破壊して節間伸長に影響があるか調査する。また、これまでに浮きイネを用いた遺伝学的解析から、節間伸長制御の重要遺伝子群を同定してきたが、これらの遺伝子を共発現解析、yeast-two-hybrid解析、G-SELEX法などの分子生物学的解析に供して、節間伸長に関わる新規遺伝子を同定する。恒常的通気組織形成と誘導的通気組織形成はそれぞれオーキシンとエチレンがトリガー因子となるものの、最終的には類似したプログラム細胞死によって形成されることから、これら2種類の通気組織形成機構の分子レベルでの共通点と相違点を抽出する。そのために、各々の通気組織形成の変異体の根の皮層組織を、レーザーマイクロダイセクションで単離し、RNA-Seq解析を行う。この解析によって、各々の下流因子の遺伝子同定を行い、2種類ある通気組織形成の制御機構の解明を目指す。

    2017年度はRAE3のクローニングを試みた。ファインマッピングにより4つ遺伝子に絞り込んだ後、塩基配列の比較から候補遺伝子を1つ見いだした。また、染色体断片置換系統群を用いた解析から、RAE1/An-1とRAE2は芒伸長に対して独立に機能し、両方を機能型で持つ系統は片方だけを持つ系統より長い芒を呈する、すなわち相加的に働くことが示された。一方でRAE3を機能型で持っていても芒は伸長せず、RAE1/An-1もしくはRAE2とペアになって働くことが示された。RAE1/An-1-RAE2-RAE3の遺伝的上位性を明らかにするため、RAE1/An-1&#8211;RAE3をペアでもつ系統(A)、RAE2 -RAE3 をペアでもつ系統(B)、RAE3のみをもつ系統(C)、SGR19系統を用いてqRT-PCRにより各遺伝子発現レベルを検証した。その結果、AとBではRAE1/An-1、RAE2が高発現しているのに対し、CとSGR19では両方の遺伝子発現は低く抑えられていた。このことからRAE3はRAE1/An-1、RAE2の遺伝的上位にあり両方の遺伝子発現を制御していることが示唆された。また前年度までは、O. rufipogonまたはO. glaberrimaを供与親、O. sativaを背景親としたCSSLを用いて解析を行っていた。今年度はO. sativaを背景親に固定し、AAゲノム種のイネ7つを供与親とする11のCSSLを解析することで、上記の2種以外のAAゲノム種イネがもつ芒関連遺伝子について広く議論しようと試みた。各系統における芒長と一穂あたりの有芒種子割合を調査した結果、複数の染色体領域が保存されていることが明らかとなった。特に第4、第8染色体が供与親に置換した系統はほぼ全て有芒であり、配列解析の結果RAE1/An-1とRAE2が機能型であることが明らかとなった。
