    タンパク質含量の異なる食餌を摂取させ、アミノ酸代謝フローの追跡を行い、脂肪酸合成への関与を明らかとする。in vitroでの膵臓ランゲルハンス島を用いたアミノ酸刺激実験により、内分泌ホルモンの分泌に必要なアミノ酸を同定し、アミノ酸感知システムの制御タンパク質などの関与を明らかにし、阻害剤などを用いたin vivoでの実験により、それらの関与を検証する。また、in vitroでの肝細胞を用いた内分泌ホルモンの刺激実験により、変化する代謝物などを明らかにし、阻害剤などを用いたin vivoでの実験により、それらの関与を検証する。

    Loss or mutation of YggS/PROSC protein, a pyridoxal phosphate (PLP)-binding protein, disrupts a wide variety of metabolic systems, especially amino acid and vitamin B6 metabolism. This study has demonstrated that the YggS/PPROSC protein plays an important role in the homeostasis of vitamin B6, especially pyridoxine phosphate (PNP), in various organisms. In E. coli, the abnormal accumulation of PNP inhibits the glycine cleavage system and various metabolic pathways, including amino acid metabolism. The present study suggests that the YggS/PROSC protein may be involved in the conversion process between PLP and pyridoxamine phosphate (PMP).

    We analyzed the catalytic and regulatory mechanisms of D-Asp synthetic activity of mammalian serine racemase (SR). Data inidicaed that SR is one of mammalian D-Asp biosynthetic enzymes. In addition, we analyzed urinary D-Ser using our D-, and L-Ser enzymatic assay method. The results obtained here suggested that D-Ser levels could be used as a predictable biomarker of renal function reduction in some situations. In addition, we studied the molecular mechanism of developmental abnormalities of Dsd-deficient Dicsyotelium strains and revealed the involvment of D-Ser in cAMP signaling. We found a novel Dsd substrate D-amino acid in the fruiting body of Bunashimeji mushroom, and identified it as (2R, 3S) 2-amino-3,4-dihydroxybutanoic acid.

    現行のD-セリン定量法における様々な問題を解消する新規なD-セリン定量法を提供すること。また、D-セリン定量法に利用される新規な酵素、それをコードする遺伝子などを提供すること。(a)配列番号1で示されるアミノ酸配列を有するタンパク質、又は(b)配列番号1で示されるアミノ酸配列と相同なアミノ酸配列を有し、D-セリンデヒドラターゼ活性をもつタンパク質からなる新規なD-セリンデヒドラターゼが提供される。また、当該酵素を試料に反応させた後、生じたアンモニア又はピルビン酸を定量し、定量値から試料中のD-セリン量を算出することを特徴とするD-セリン定量法が提供される
