    第3の酸化還元補酵素として同定されたピロロキノリンキノン(PQQ)は、哺乳動物を含む高等動物におけるビタミン様物質として知られているものの、その多彩な生理作用を補酵素としての機能だけでは説明することができない。本研究では、ある種のGPCRがビタミン様物質PQQの受容体候補分子であると捉え、これを実証することによりPQQの新規な生理活性とその作用機序の解明に挑戦する。
    Gタンパク質共役型受容体(GPCR)の中でも、腸管上皮などに発現する代謝物感受性GPCRに注目し、ピロロキノリンキノン(PQQ)による活性化を評価した。6種類の代謝物感受性GPCRに関して、PQQによる活性化をtransforming growth factor-α(TGF-α)切断アッセイにより評価したところ、GPR35のみがPQQにより顕著に活性化されることがわかった。さらに、 PQQはその投与濃度依存的にGPR35を活性化すること、PQQの部分構造体では全く活性を示さないこと、ヒトGPR35だけでなくマウスGPR35も活性化することがわかった。
    ヒト結腸癌由来細胞株であるCaco-2細胞をモデルとしてPQQの機能について検討した。分化させたCaco2細胞にPQQを処理したところ、GPR35の下流に位置すると考えられるERKのリン酸化が認められた。このリン酸化は、GPR35のアゴニストであるCID 2745687の前処理により部分的に抑制された。このことからPQQによるERKのリン酸化にはGPR35が少なくとも部分的には関与していることが示唆された。
    GPR35とPQQが直接相互作用をするかを証明するため、また新規なPQQ受容体探索を探索するため、PQQの化学プローブを作製した。PQQと受容体タンパク質との非共有結合による相互作用を捉えるため、光反応性官能基であるベンゾフェノンおよびクリック反応のためのアルキニル基を導入したPQQプローブを作製した。さらにこのプローブを用いて、培養細胞におけるラベル実験を行ったところ、複数のタンパク質がラベル化されることがわかった。
    当初の計画通り、PQQ受容体の候補タンパク質を見出すことができた。またPQQプローブの作製も完了した。
    PQQによるGPR35活性化の意義についてさらに詳細に解析を行う。あわせて、PQQプローブを用いた標的タンパク質の探索を進めていく予定である。

    Bile acids are secreted into the upper small intestine upon ingestion, and then taken up in the lower small intestine and returned to the liver. Since a portion of bile acids flow into the systemic bloodstream, the bile acid concentration in the blood increases after eating, which is considered to be a feeding signal. The target molecule that recognizes this is the bile acid receptor TGR5. For functional analysis in skeletal muscle, transgenic mice expressing human TGR5 in skeletal muscle were established and analyzed. It was found that activation of TGR5 by bile acid resulted in a muscle hypertrophy effect. When an oral glucose tolerance test was performed, a rapid decrease in blood glucose was confirmed, and it was clarified that an increase in muscle mass leads to an improvement in glucose intolerance. Part of the increase in muscle mass associated with the feeding response may be due to TGR5 function.

    (1) We identified a novel lysine adduct as an oxidative stress-related biomarker candidate. We also unambiguously detected 2-oxo-histidine-containing dipeptides in mouse tissue homogenates. In addition, an enhanced S-thiolation of HSA in the hyperlipidemia patients was observed.(2) We identified complement component C1q as a novel AGE-binding protein. We also discovered that the EGCG could be an endogenous source of the electrically-transformed proteins that could be recognized by the natural antibodies. In addition, lactate dehydrogenase was identified as a novel a mammalian PQQ-binding protein.(3) We found that the filamin A acted as a guanine nucleotide exchange factor for Drp1 and mediated mitochondrial fission-associated myocardial senescence in mice after myocardial infarction. We also demonstrate that TRPC3 acts as a positive regulator of ROS that stabilizes Nox2 activity through physical interaction, leading to amplification of maladaptive signaling in cardiomyocytes.
