    核多角体病ウイルス(NPV)に感染した昆虫細胞に誘導される全タンパク質合成停止のメカニズムを明らかにすることを目的として,NPV感染によって誘導されるカイコ(Bombyx mori)由来培養細胞におけるリボソームRNA(rRNA)の分解機構とその誘導機構について解析を行った.
    リボソーム分解経路の解析:NPV感染カイコ細胞では,約1,400 ntのrRNA分解断片が検出されることから,rRNA分解の実行因子はエンドリボヌクレアーゼであると予想した.カイコゲノムデータベース(KAIKOBase)を用いて,エンドリボヌクレアーゼをコードすることが推定される18個の遺伝子を選抜し,rRNA分解への関与を明らかにするため,Autographa californica NPV(AcMNPV)感染カイコ細胞におけるノックダウン解析を行った.その結果,いずれの遺伝子をノックダウンした場合もrRNA分解は抑制されず,エンドリボヌクレアーゼを特定することはできなかった.
    AcMNPVのP143と相互作用するカイコ細胞因子の探索:AcP143の機能ドメインの絞り込みを行い,325-599アミノ酸残基からなる領域(AcP143(325-599))でrRNA分解が誘導されることを明らかにした.さらに,325番目のグルタミン残基(Q325)の重要性を調査するため,Q325をアラニンに置換した変異体(AcP143(Q325A))を用いて一過性発現解析を行った.その結果,AcP143(Q325A)を発現させた細胞は,AcP143を発現させた細胞と同様にrRNA分解を誘導したことから,Q325はrRNA分解において直接的な機能を持つのではなく,AcP143の構造維持などに機能していると考えられた.AcP143(325-599)と相互作用するカイコ細胞因子を探索するため,大量発現系の構築を検討した.
    カイコゲノムの遺伝子情報からrRNA分解に関わるエンドリボヌクレアーゼの探索を進めた.18遺伝子をエンドリボヌクレアーゼの候補遺伝子として実験を進めたが,エンドリボヌクレアーゼを特定することができなかった.したがって,エンドリボヌクレアーゼを特定するために,別のアプローチを検討する必要が生じた.
    リボソーム分解の誘導機構解析を目的として,引き金となるウイルス因子と相互作用するカイコ細胞因子の探索を行った.まず,全長のAcP143を用いて大量発現系の構築を検討したが,タンパク質発現を確認することができなかった.そこで,AcP143の325-599アミノ酸残基からなる領域でrRNA分解が誘導されることから,この領域を大量発現させる系の構築を行った.まず,大腸菌によるタンパク質発現系を試みたが,標準的な方法では発現を確認することができず,さまざまな方法を試行することになった.最終的に,バキュロウイルス発現系を用いることによって,タンパク質発現を確認することができた.
    バキュロウイルス発現系を用いてAcP143の機能ドメイン(325-599アミノ酸残基)を大量発現させ,アフィニティー精製法により発現タンパク質と相互作用する細胞因子の探索を進める.
    蛍光標識したリボソームタンパク質をカイコ細胞に一過性発現させることでリボソームを可視化し,rRNA分解時のリボソームの挙動を調査する.これにより,rRNA分解が進行する細胞内の位置の特定および分解経路の予測を行う.

    We previously showed that rRNA degradation occurs in Bombyx cells (BM-N cells) as an antiviral immune response during abortive infection with heterologous nucleopolyhedroviruses (NPVs). In the present study, we examined the induction mechanisms of rRNA degradation in Autographa californica NPV (AcMNPV) infected BM-N cells. We identified AcMNPV p143 (ac-p143) as an inducer of rRNA degradation. And then, we investigated the domain of Ac-P143 responsible for rRNA degradation, the cellular factors interacting with Ac-P143, and the signal transduction factors responsible for induction of rRNA degradation.
