    飛行時間型二次イオン質量分析(TOF-SIMS)によって不均一な生体組織あるいは複合材料中における微量成分の定性的な分布評価が可能である。申請者はこれまでに、急速凍結させた試料を連続的に切削することにより、細胞(細胞壁)内にあるがままに存在するあらゆる構成成分の三次元分布を、分子レベルで可視化する複合装置(クライオTOF-SIMS/SEM システム)を世界に先駆けて開発してきた。本研究ではリグニン前駆物質の輸送・貯蔵に関わる細胞間ネットワークの解明を目的とし、凍結した植物試料内での様々な化合物の動態可視化を試みる。
    (1)二次木部の各種組織の木化における同種・異種細胞間の相互作用
    樹木二次木部ではほとんどの細胞が木化するため、二次壁の木化に必要な多量のリグニン前駆物質が周辺細胞から供給される可能性は低く、液胞内に貯蔵された前駆物質が用いられるか、形成層付近の若い同種細胞から放射組織経由で供給されることが想定される。第4年度では特に姿勢制御応答として生じる特異な化合物に焦点を当て、その輸送・消費状況を追跡するための新たな可視化技術の開発に取り組んだ。
    (2)微量成分・高分子量成分を対象とした新規イメージング法の開発
    生体試料内には様々な微量有機成分ならびに高分子成分が含まれているが、イメージング質量分析法ではある一瞬の状態を可視化しており、内部メカニズムの動的考察には不明な点も残されている。第4年度では高感度検出可能なマーカー化合物を利用することで、生体内における物質輸送の流れの可視化を検討したところ、時間変化による分布差異を明瞭に示すことに成功した。
    可視化技術の開発に複数のアプローチから取り組み、それぞれ良好な結果が得られたことから、当初計画の目標に対しておおむね順調に進展していると判断した。
    今年度までに実証できた新可視化法を用いて、リグニン前駆物質の輸送・貯蔵に関わる細胞間ネットワークの解明に取り組む。

  2. 二次イオン質量分析法による植物細胞における生体分子三次元分布の可視化

    本研究の目的は、生きたままの植物細胞を急速凍結した試料において、飛行時間型二次イオン質量分析(TOF-SIMS)を用いることによって、細胞や細胞壁内にあるがままに存在するあらゆる構成成分(細胞質物質、オルガネラ構成成分、細胞壁中の多糖ならびにリグニン、抽出成分)の分布を、選択的に分子レベルで可視化することである。植物の細胞の構造を、視覚的に、また化学的に理解することは、あらゆる代謝物の生合成を理解する上で非常に重要である。しかしながら、細胞内に存在する分子は、水可溶性であったり、極めて不安定であったり、異質の物質と複合体を形成して存在していたりして、従来の方法では解析困難であった。

    これらの課題を克服するために、前年度までに、窒素雰囲気下で急速凍結した生体試料をスライディングミクロトームを用いて平滑な面を作成する前処理室を設計・構築し、さらに、凍結した状態で試料をTOF-SIMSおよび走査電子顕微鏡(Cryo-SEM)に移動させることのできる搬送装置を備えた世界初の前処理システムを開発した。今年度は、前処理システムに改良をかさね、実際に、生きた植物(針葉樹、広葉樹)の急速凍結試料を用いて、グローブボックス内の凍結スライディングミクロトームを用いた試料切削、TOF-SIMS分析、Cryo-SEM観察を同一系内で連続的に行った。SEM試料室で氷を昇華することにより細胞形状、細胞内小器官レベルの位置情報を入手することが可能となり、スクロースなどの水可溶分子でも組織内マッピングを高解像度で行うことに成功した。

    また、TOF-SIMS分析において欠かすことのできない、二次イオンの同定と定量法についても基盤的な研究を展開し、新規な分析法(リグニンのケミカルマッピング)を開発した。

    遺伝子資源の宝庫であるアマゾン川流域および植物が独自の進化を遂げてきたギニア高地における本植物(おもに被子植物)の木部形態形成に関し、材利用の観点から材質を左右する因子に焦点をあて、環境適応と遺伝学的分類の両面から知見を得ることを目的とした。平成15年9月には、ブラジルアマゾン州の熱帯林、パラナ州の亜熱帯林を、平成16年3月には、ベネズエラギアナ高地において固有樹種の調査を行った。同年9月にマトグロッソ州北部のコトリグアス周辺の熱帯雨林をマトグロッソ連邦大学と共同で調査し、ブラジル環境省(IBAMA)より正式な輸出許可を受け、貴重な原生林より伐採直後の数種の熱帯産材を日本に持ち出すことに成功した。平成17年4月にはマトグロッソ連邦大学において約30名の日伯研究が参加したワークショップを開催した。同年7月には、マトグロッソ州熱帯林において熱帯産樹種のサンプリングを再度おこなった。また、マトグロッソ州クイアバ市近郊、サンパウロ州イタペバ市近郊からも熱帯産樹木および亜熱帯産数種を数種採取した。当初の目的では、遺伝子解析を通して、植物の進化と木部形態形成の関連を探ることを項目のひとつに加えていたが、遺伝子資源の保護の観点から、IBAMA(ブラジル環境省)より木部以外の試料の海外持ち出しの許可が得られず、計画の一部変更を余儀なくされた。しかしながら、樹木の分類に有用な微量成分(抽出成分)を解析項目に加え、アマゾン産材の特徴付けを行った。

    採取した試料は、これまでに、材比重、抽出成分量、シリンギル・グアイアシルリグニンの比率等について解析をおこない、多種多様な種を分類・序列化するための基礎データとした。また、樹木の特徴的な性質を示す抽出成分の解析をおこない、未利用アマゾン産樹種の利用価値を高めるデータを提供した。

    Cationic and anionic groups were introduced into lignin and their conversion into functional materials was attempted. Acid-hydrolyzed lignin (SAL) was selected as the least reactive lignin, as it seems to be the most reactive. It was found that the SALs were soluble in water when subjected to alkaline hydrothermal treatment, and the chemical structure of the SALs was analyzed, and it was found that phenolic hydroxyl groups and carboxyl groups were introduced.
    The water-soluble HSL was cationized, and it could be applied to the flocculation treatment of the dye waste solution. The carboxyl group was introduced into the water-soluble HSL and the product was applicable to the dispersant of gypsum.
    The bioactivity of water-solubilized HSAL was investigated, and it was confirmed that it promoted the growth of plant roots.

    In this study, the cultivation of plant samples under 13CO2 atmosphere was examined for supersensitive structural analyses. At first, the environmentally controlled growth chamber was designed and built up. Plant samples were cultivated under N2+O2+CO2 (in chamber), N2+O2+13CO2 (in chamber), or air for 2 months. The resultant plant samples had high 13C ratio and supersensitive analyses was achieved successfully.

    Versatile analytical method was developed based on the synergistic utilization of time-of-flight secondary ion mass spectrometry (TOF-SIMS) and quantitative chemical analysis, laser microdissection to collect microscopic samples, and/or multivariate analysis. The developed method was applied to various samples including frozen-hydrated biomaterials. Low-molecular and water-soluble compounds and inorganic metal ions are effectively visualized by the developed method using (cryo-)TOF-SIMS. Obtained remarkable results should provide new insights beyond the field of wood sciences.

    リグニン形成の制御機構を前駆物質の貯蔵と輸送という動的な観点から捉え、モノリグノールの貯蔵と供給機構に着目し、細胞レベルで解明することを目的とした。安定同位体トレーサー法によるリグニン生合成中間物質の選択的な標識と飛行時間型二次イオン顕微質量分析(ToF-SIMS)による分子マッピングを組み合わせて、分子レベルの情報と組織レベルの情報を同時に入手することができるのが本研究の特徴である。

    まず、細胞レベルでの細胞壁成分の顕微レベルでの分子マッピングを可能にするための技術開発を行った。樹木木部細胞壁におけるシリンギル、グアイアシルリグニンのマッピング手法を確立するため、カエデを用いて、飛行時間型二次イオン質量分析を行った。その結果、シリンギルリグニンとグアイアシルリグニンの細胞壁中での分布を正確に可視化する手法を開発した。化学分解法や組織化学的手法とデータを比較検討することにより、本法の妥当性を証明するとともに、顕微領域における定量分析も可能とした。心材成分の樹幹内での分布も細胞レベルで可視化することに成功した。このことは、リグニン生合成中間体である低分子フェノール性化合物の選択的なマッピングにも適用できることを示した。

    次に、リグニン生合成代謝中間物質の細胞内分布を明らかにするため、モノリグノールグルコシドのToF-SIMS を用いた解析法を確立した。LC-MS とToF-SIMS 分析を組み合わせることにより、配糖体の定性・定量分析を行った。また、特定の位置を13C で標識した前駆物質(コニフェリン)を合成し、イチョウ切り枝に投与した。一定期間代謝させた後、新生木部を採取し、標識前駆物質からリグニンへの取り込みを解析した。

    安定同位体元素でラベルしたコニフェリルアルコール、コニフェリルアルデヒドおよびコニフェリンをイチョウ、広葉樹にそれぞれ投与して、生成したリグニンを化学分析した結果、コニフェリンからコニフェリルアルデヒドグルコシド、さらにコニフェリルアルデヒドという経路でリグニン生合成に合流することが示された。オートラジオグラフィーによって、コニフェリン、コニフェリルアルデヒドグルコシドのアグリコン由来のラベルは、コブシ(配糖体貯蔵広葉樹)においては木部細胞壁に取り込みが認められた。またシリンギンアグリコン由来のラベルは少なく、放射柔細胞および道管壁に取り込みが見られた。シナップアルデヒドグルコシドアグリコン由来のラベルも木部で認められるものの、非常に弱く、コニフェリルアルデヒドグルコシドの活発な取り込みとは対照的だった。キョウチクトウ(非糖体非貯蔵広葉樹)においてもコニフェリン、コニフェリルアルデヒドグルコシドのアグリコン由来のラベルは木部細胞壁に取り込みが認められた。コブシとキョウチクトウにコニフェリンおよびコニフェリルアルデヒドグルコシドを投与した結果、グアイアシルリグニン、シリンギルリグニンどちらにもアグリコンの取り込みが見られた。しかしシリンギルリグニンへの取り込みはグアイアシルリグニンへの取り込みに比べて少なかった。またシリンギルリグニンへの取り込みは、コニフェリルアルデヒドグルコシドを投与した場合のみ取り込みが見られた。したがって、コニフェリンよりもコニフェリルアルデヒドグルコシドのほうがリグニン前駆物質として優勢であること、さらにグアイアシルユニットからシリンギルリグニンユニットへの転換にはアルデヒド構造が大きく関与していることが示された。

    また、数種の被子植物のモノリグノール生合成経路をトレーサー法により調べた結果、シナピン酸を経由する種とそうでない種とに分類することができ、リグニン生合成の多様性が示唆された。
