    皮膚形成では、表皮細胞の分化に伴ってケラチン等の様々なタンパク質が接着(架橋)して不溶化し、表皮細胞膜を裏打ちすることにより皮膚の硬度が保たれている。この接着反応を担うのが、タンパク質接着酵素(トランスグルタミナーゼ)で、遺伝子欠損も含め異常な活性レベルは多様な皮膚疾患をもたらす。またこの酵素反応は細胞外マトリクス(ECM)の強化による傷害修復にも寄与している。そのため各種皮膚疾患の再生回復には本酵素の活性上昇が必須である。
     これまで申請者は、接着酵素に対する「高反応性基質ペプチド」をペプチドライブラリから同定して多面的に活用し、蛍光標識した基質ペプチドを用いて、組織での接着反応を可視化するシステムを確立した。
     本研究ではその成果を基に、遺伝的要因による皮膚疾患や皮膚再生における接着反応(産物)の可視化システムを展開・活用し、病的皮膚形成への対処法と創薬シーズ開発のための情報基盤を提供する

    動物の生体内にはタンパク質を接着させる酵素反応が存在する。これらは血液凝固や皮膚形成、傷害修復など多彩な生命現象に関与し、その異常な活性レベルは疾患症状をもたらす。申請者は本酵素を対象に、天然に存在するタンパク質基質よりも遥かに反応性の高い「高反応性基質ペプチド」をペプチドライブラリから同定してきた。また、これらを蛍光標識したペプチドを用い、組織内の接着反応の可視化に成功している。この成果を基に、1)蛍光共鳴現象(FRET)による細胞内活性の可視化、2)疾患診断への広汎な活用を目指す。これらの高反応性基質ペプチドの可視化システムの活用は、疾患要因の特定や傷害修復における有効な因子探索に寄与する。
