遷移金属カルベン錯体は、有機合成において極めて有用なツールとなっている。本計画では、末端アセチレンとカチオン性ルテニウム錯体から発生するビニリデンカルベン錯体への酸素原子移動反応によるケテンの発生と、同過程を組み込んだ新規触媒系の創成を目指す。旧来のカルベン錯体からのケテン発生法は、予め調製されたカルベン錯体を用いる非触媒的手法であったことに加え、有毒な一酸化炭素の挿入に頼らざるを得なかった。本計画の達成によって、入手容易な末端アセチレンよりビニリデンカルベン錯体を発生させ、その酸化により所望のケテンを触媒的に発生させる実用的な手法が確立される。本手法は学術的に興味深いことに加え、極めて応用範囲の広いものである。さらに、本研究を通して開発される触媒的分子変換プロセスを駆使して、生理活性物質の合成を展開する。

2価ルテニウム錯体を触媒とする[2 + 2 + 2]環化付加反応を駆使して,生理機能が期待されるスピロ環状C-アリールグリコシドの多様性指向型合成法を確立した。また,銅触媒マンニッヒ型三成分カップリングとイリジウム触媒環化異性化/Diels-Alder環化付加タンデムプロセスを統合した環状アミノ酸の新規合成法を開発した。
