    To obtain cells with high dibenzothiophene (DBT) degrading activity, two phase fed-batch culture of Rhodococcus erythropolis KA 2-5-1 was carried out, which consisted of growth phase in the medium containing sulfate ion as a sulfur source and induction phase for expression of DBT degrading enzyme by DBT addition. Cell concentration of 24.3 g-dry cells/L was obtained after 30 h culture and the maximum specific degrading activity of 18.2 mmol/kg-dry cells/h was achieved at 10 h after induction. In order to simplify the system and lower an operational cost, next, in situ induction of the enzyme activity was conducted by means of sulfur limitation since the activity of the strain can only be induced under sulfur starvation. The strain was cultivated in fed-batch mode in the modified A medium containing 96 mg/L of sulfate ion. At 22 h, sulfate concentration decreased below 10 mg/L and approximately 10 g/L of cell concentration was obtained. When 125 mg/L of DBT was added at 22 h, the enzyme activity increased rapidly and the maximum specific activity of 14.3 mmol/kg-dry cells/h was obtained at 28 h. However, the DBT degrading activity decreased rapidly thereafter. From the assay of remaining concentration of medium components in the culture broth, it was found that Fe and Mg ions were exhausted at the end of cultivation. Fed-batch culture with feeding of concentrated solution of Fe and Mg ions was then conducted. After 23 h, 125 mg/L of DBT was added, and the enzyme activity increased rapidly. In this case, the activity did not decrease, and the maximum specific degrading activity of 18.5 mmol/kg-dry cells/h and cell concentration of 41.7 g-dry cells/L were obtained at 34 h.

    To reduce the hydrodynamic stress in plant cell culture and enhance the production of regenerated plantlets, a liquid medium containing a viscous additive was newly designed and plantlet production from embryogenic carrot callus cultivated in the medium was examined. Na-Alginate or carboxymethyl cellulose (CMC) was used as the viscous additive. The viscosity of the medium increased with increasing additive content and the number of regenerated plantlets also increased. When carrot calli were cultivated in the medium containing 0.4% CMC, designated as N medium (viscosity, 3 mPa・s), the maximum enhancement of plantlet regeneration, approximately 2.5 times higher than that in the control medium, was obtained. Enlargement of callus size observed in N medium is considered to be the main reason for the enhanced plantlet regeneration. Regeneration enhancement was sufficiently induced after calli were cultivated once in N medium, but this regenerarion ability rapidly disappeared after once cultivation in the conventional medium. In repeated batch culture using N medium, plantlet production continued at a high level for 18 batches (250 d) with no significant decrease, while in the control culture without CMC the number of plantlets produced dropped to almost zero by the sixth batch (84 d).

    Efficient lipase production by two-step fed-batch culture of an organic solvent-tolerant bacterium, Pseudomonas aeruginosa LST-03, was investigated. When FB synthetic medium was used in flask culture, no lipase activity was detected, whereas lipase was produced at 2.3 I.U./ml in C2 complex medium. However, lipase production was induced in FB medium when a fatty acid was added to the culture broth in the stationary phase. Among fatty acids tested, long chain saturated fatty acids, such as C18 (stearic acid) and C20 (arachidic acid), were found to function as effective inducers for the production of lipase, giving an activity level almost the same as that obtained in C2 medium in flask culture. Two-step lipase production, comprised of a growth phase in fed-batch mode and a production phase in which lipase was induced by the addition of 5 %(v/v) stearic acid, was carried out in a jar-fermentor. In the growth phase, the maximum cell concentration at 16 h was only 20 in terms of the optical density at 660nm (OD660), and a low level of lipase production (8 I.U./ml) was obtained after 167 h. This was considered to be due to the exhaustion of several medium components brought about by the use of an unsuitable medium or feeding solution. After analyzing the contents of the compounds in the culture broth by inductively coupled plasma spectrometry for metal ions and HPLC for anions, a modified FB medium was designed. When this modified FB medium was used in two-step fed-batch culture, the maximum cell concentration reached an OD660 of 55 (30.2 g-dry cells/l) at 16.5 h, and lipase was produced at 96 I.U./ml after 35 h, which is approximately 40 times higher than the production level obtained in flask culture using C2 medium.

    磁性微粒子ラベリング技術を用いた1藻類細胞アレイを作製し、イソプロパノール高生産、高耐性を持つ変異藻類を迅速に探索する。このため、培養藻類が磁気分離できる磁性ナノ微粒子を作製し、1藻類細胞を液滴に隔離し、オイルで封入した培養器中で孤立培養する。また、磁気分離を用いて数万細胞の特性を一気に探索できるハイスループット選択システムを構築し、高機能細胞をスクリーニングする。本研究の成功には高性能藻類を迅速に探索・分離する技術の確立が重要である。磁気分離は1細胞の機能評価には優れた方法であり、磁気ハンドリングと液滴操作は名古屋大学で確立された他に類を見ない手法である。

    It is not clear how exercise alters the number and quality of EVs secreted by skeletal muscle cells. In this study, we used an in vitro exercise model to examine whether exercise affects the amount of EVs released from skeletal muscle cells. The effects of 1 Hz and 30 Hz electrical stimulation applied under different exercise conditions for 24 hours were examined. The results showed that the amount of EVs secreted increased and the type of mircoRNAs contained in EVs also fluctuated when the subjects were exercised with 30 Hz electrical stimulation. Furthermore, it was suggested that changes in Alix expression and calcium ion concentration were involved in this.

    Bioactive peptides with moderate length of 5 to 10-mer are potentially existing into the sequences of plentiful edible proteins. In oral administration of such peptides, development of direct delivery without hydrolysis to the intestine space is strongly desired in the field of healthcare, nutrition and cosmetics. In the present study, following 3 topics were investigated, 1) screening of meso-length bioactive peptides from plentiful edible protein by informational analysis, 2) establishment of selective separation and concentration method of such peptides by optimization of protease hydrolysis, and 3) development of direct delivery without hydrolysis to the intestine using heat treated porous silica gel which can adsorb bioactive peptides in inner space at acidic pH and can release them at neutral pH.

    We have discovered that lactostatin (IIAEK), a peptide that improves cholesterol (CHOL) metabolism, affects the intestinal CHOL metabolism-improving gene (ABCA1) through specific activation of alkaline phosphatase. The first CHOL metabolism-improving dipeptide FP (phenylalanine-proline) was discovered from 400 dipeptides in the world. Nine soybean β-conglycinin-derived peptides with 3 to 8 amino acid residues were detected in rat whole body plasma.

    Peptides that have biological activities are called functional peptides. Many types of functional peptides have been found to date and has been designed for development of peptide drug. In the present study, following 3 topics were investigated, 1) fabrication of peptide library including unnatural amino acid, 2) screening of intracellular functional peptide conjugated with cell penetrating peptide (CPP), 3) development of the smart machinery for removal of CPP in intracellular space. Related to the last topic, we have established the methodology for disulfide bond formation of all peptides on peptide array. In addition, we have been developed a novel synthesis method of hetero-dimerized peptide using intramolecular disulfide bonding between main- and sub-chain of lysin.
