    本年度は、昨年度までの研究で確立したRNA-protein相互作用検出法tRIPを用いて、RNA結合タンパクFUSとU1 snRNPの相互作用に焦点を当て、研究を行った。FUSとU1 snRNPは、細胞内で複合体を形成するものの、その転写制御における役割は、明らかでなかった。tRIP法を駆使することで、以下の点が明らかとなった。
    1. FUSとU1 snRNPは、RNAP II転写複合体上で、新生RNA上に形成されている。どちらも、とりわけ、選択的転写終結部位周囲で顕著であった。U1 snRNPの結合部位が、転写終結部位から数百塩基上流に形成されることを初めて明らかにした。2. FUSとU1 snRNP、どちらが欠損しても、新生RNAとこれら分子の結合は、形成できない。つまり、FUSとU1 snRNPは複合体として、機能している、と考えられた。3. FUSとU1 snRNPの複合体は、新生RNA上でポリアデニル化抑制を行うことが主要機能であり、転写伸長の維持に必須であることを証明した。
    4. この複合体は、ALS患者に認められるFUS変異で、形成が阻害されていた。5. ALS患者変異では、ポリアデニル化異常が認められ、そのパターンは、U1 snRNP-depletionとよく似ていた。FUS-U1 snRNP複合体の病態形成における意義が、示唆された。
    以上の結果について、EMBO reports誌に論文として発表した (in press)。また、本研究成果について、日本分子生物学会で発表を行い、他研究施設との共同研究が複数、進行するところとなった。その成果の一部は、すでにCell誌の論文発表となっており、他にも3ヵ所で、共同研究が現在進行中である。
    効率、感度ともに従来法を大きく上回り、現時点で世界最高峰である新規RNA-protein相互作用検出法であるtRIP法の開発に成功し、その成果の論文発表を行った。順調に進展している、と考えられる。
    現在、RNA-protein相互作用は、新しい細胞内小分画の概念である相分離の観点から注目されている。本研究で開発したtRIP法を、相分離した分画内でのRNA-protein相互作用検出へ応用できないか、検討していきたい。

  4. MERS-異常言動スペクトラムの分子生物学的基盤の解明

    急性脳炎・脳症の遺伝学的背景の解明のため、遺伝学的解析を施行した。可逆性脳梁膨大部病変を有する軽症脳炎・脳症(MERS)を反復する家族例2家系を発見し、全エクソーム解析によって、MYRF遺伝子に2家系に共通するミスセンス変異を同定した。この知見に基づいて、他のMERSの症例に対し、全エクソーム解析を行った。今年度は、反復性および家族性のMERS症例を対象に、MYRFで制御される可能性のある約700の遺伝子についてのバリアントに注目し、神経疾患をもたない対照群と比較した。その結果、バリアントをもつ遺伝子の数には差を認めなかったが、MERS症例と対照群では、バリアントを認める遺伝子が異なっていた。MERS症例では、GFOD1・HEMK1・CDIP1・PCSK6・LGMN・SLC5A11の変異が、対照群に比べて多い傾向であったが、これらの遺伝子バリアントとMERS発症との関連については現時点では明確な結論には達していない。さらに、男性のみにMERSが発症した家系例について、遺伝学的解析を行った。今年度はMERSに類似した可逆性脳梁膨大部病変を認めることが知られているX連鎖性Charcot-Marie-Tooth病の原因遺伝子であるCx32に注目し、解析を施行した。しかし、Cx32の変異は認めなかった。これらの結果から、MERSの発症については現在報告されているものよりも多様な遺伝学的要因が関与していると考えられた。MERSでは高サイトカイン血症との関連が指摘されており、免疫調節に関与する遺伝子との関係を解析する必要があると思われた。
    今年度中に、遺伝学的解析を概ね終了することができたため。
    今後は、MYRF遺伝子の変異による機能の変化を様々な方法で研究する予定である。温度などの環境やサイトカインなどの液性因子によるMYRF蛋白の機能への影響を、試験管内や動物モデルの作成によって検討する準備を開始している。また、MERSや発熱に伴う異常言動の孤発例に対し全エクソーム解析を行い、MYRF以外の遺伝子の関与を検討中である。これらの研究成果は、発熱に伴う異常言動やMERSの原因を明らかにし、その病態や治療法の解明に有用である可能性がある。また、薬物と発熱に伴う異常言動との関係が明らかになり、その使用の適否を科学的な根拠によって示すことが可能になることが期待される。

However, the transcriptome-wide detection of protein-RNA interactions in a subcellular component is challenging. Here, we developed the tRIP method (targeted RNA immunoprecipitation) to increase efficiency of RNA immunoprecipitation, which enables detection of UV-crosslinked protein-RNA interactions from thousands of cells. Application of tRIP to identification of protein-protein-RNA interactions revealed that FUS, a multifunctional RNA-binding protein associated with ALS, binds specifically upstream to alternative polyadenylation (APA) sites of nascent RNA engaged to RNAP II. Further tRIP analysis demonstrated that the FUS-U1 snRNP-RNA complex is the entity that suppresses APA.
