    Wntシグナルの抑制は関節炎の抑制効果など軟骨保護に加えて複合的な効果が期待でき、関節治療薬の開発が期待できる。一方、ヒアルロン酸の主たる受容体であるCD44のADAM10による断片化がOA軟骨において観察され,CD44断片化は細胞外マトリックス形成を阻害し、断片化の結果生成されたICD(intra cellular domain)が生理活性を持ち、MMPなど分解系酵素や炎症性サイトカイン産生に働き関節破壊を招来する。ADAM10阻害はCD44断片化抑制を介して、OA治療に応用可能である。ADAM10の阻害活性を持つ化合物をDrug repositioning により見出すことを目標とする。
    ①Wnt/βカテニン阻害によるOA治療薬の開発
    1186種のFDA既認可薬を用い、Wntシグナル抑制効果のある薬剤を一次スクリーニングし、8種類の候補化合物を得て、二次スクリーニングを行った。HCS細胞株でWnt/βカテニンの抑制効果と標的分子についてmRNA発現量、蛋白発現量の変化を検討、効果を示す化合物候補を探索した。別にWntの調節因子であるDKK-1に注目して、その発現に変化を与える物質の探索を行った。予備的検討では、軟骨細胞では予想に反して、WntとDDK-1の活性が平行して動く結果を得た。そこで、Wnt抑制とDKK-1抑制の両方を満たす調節物質を探索した。以上から軟骨細胞に保護的作用を持つ物質(Raloxifene)を見出した。しかし、この物質については既に我々が過去に軟骨保護作用を報告しており、特許取得は困難であった。今年度も引き続き探索を進めることとなった。
    ②ADAM10阻害によるOA治療薬開発
    定法通りADAM10のpromoter領域にreporter遺伝子を組み込んだコンストラクト作成し、それをHCS細胞株に遺伝子導入し、Drug Repositioningの一次スクリーニング系を立ち上げた。二次スクリーニングを進めたが結果が安定していない。別にADAM10の酵素活性を測定する方法を検討した。軟骨細胞に過度な力学的刺激を加えると①ADAM10活性化により引き起こされるCD44の断片化が起こること、②CD44断片化の産物である細胞内ドメイン(以下ICD)が細胞内で代謝変化を引き起こすことを明確にした。この方法でADAM10活性化によるCD44断片化を推定することが出来る。この機械的刺激によるCD44断片化はヒアルロン酸添加によって抑制されるのでADAM10活性化機序を考察する上で重要な知見を得た。
    ①Wnt/βカテニン阻害によるOA治療薬の開発
    当初の目標であるWnt阻害薬の探索を行った。今回は探索範囲をDKK-1分子まで広げることにより精密に標的とする化合物を見出す可能性を検討した。しかし、軟骨細胞では当初の予想とは異なりWntとDKK-1の活動性が平行して動いていた。そのためにWnt,DKK-1抑制の両面から探索してRaloxifeneを見出した。しかし、本物質は既に我々が軟骨保護効果を報告済みであった。この系を利用して追求することにより残っている候補化合物を再度検証する予定である。一部に変更は必要であったが、結果として軟骨保護効果のある薬剤を見出している。
    ②ADAM10阻害によるOA治療薬開発
    ADAM10発現阻害の実験系はADAM10 promoter領域にreporter遺伝子を組み込んだ改変細胞を構築することに手間取った。一方でADAM10の酵素活性を測定可能な実験系をCD44ICDの生理活性を軟骨細胞の代謝変化として測定可能な系を構築した。これによりmRNA発現ではなくて、より直接的に問題となるADAM10の酵素活性が測定出来ることになった。これにより研究が加速できるものと思われる。
    ①Wnt/βカテニン阻害によるOA治療薬の開発
    今までの知見からDKK-1抑制からもスクリーニング系を構築できる可能性が示された。今後はDKK-1抑制に働く化合物を含め、より広範囲に探索を進める予定である。今回見出したRaloxifeneを検討した時の候補化合物についても更にDKK-1の関連から検討する。
    ②ADAM10阻害によるOA治療薬の開発
    ADAM10酵素活性を活性産物の細胞活性から間接的に評価できることを明確にした。発現調節と同時に酵素活性抑制を検討することにより二つの経路から探索可能となり、標的化合物がより確実に見出しやすくなった。このままの方向性で検討を続ける予定である。
