    1.光合成水分解機構の解明:光化学系Ⅱ(PSII)単一微結晶中の水分解反応を、顕微フーリエ変換赤外(FTIR)分光法を用いて調べた。その結果、PSIIの微結晶内部でも高い遷移効率が保たれていることが示された。また、Mnクラスターの配位子であるD1-D170のHis変異体、および近傍の水素結合ネットワーク上のD1-S169のAla変異体を作成し、その構造および反応を調べた。その結果、S169A変異はMnクラスターの近傍構造を変化させたが、D170H変異は特異的なアミノ酸変換を起こし、野生株のPSIIに戻ることが示された。
    2.キノン電子受容体の酸化還元電位制御:PSIIにおける第一キノン電子受容体QAの酸化還元電位(Em)は、Mnクラスターによって制御されることが定説となっていたが、これらのコファクター間の相互作用の存在は確認されていない。そこで、FTIR分光電気化学法を用いて、QAのEmへのMn除去の効果を調べ、これまでの蛍光法による結果と比較した。その結果、Mnクラスターの有無によって、QAのEmは変化しないことが示され、これまでの説は蛍光法によるアーティファクトであることが示された。
    3.反応中心クロロフィルの構造と反応:PSIIのChlD2の軸配位子を置換した好熱性シアノバクテリアの組換え体を作製し、ChlD1変異体と共に、4分子の反応中心クロロフィルの正電荷分布の移動、および光阻害への影響について調べた。その結果、ChlD1は光励起後3 psでChlD1+になり、正電荷は350 psで[PD1 PD2]+ に移動することが示された。ChlD2変異体では正電荷はChlD2まで広がったことから、ChlD2の配位子構造が正電荷分布を制御することが示唆された。そして、ChlD2変異体は強い光阻害を受けたことから、副次的電子移動における電子受容体であることが示唆された。
    A01班が作成した光化学系Ⅱ微結晶のFTIR解析を、全反射FTIR法によって微結晶の集合体として行うだけでなく、顕微FTIR分光を用いて単一の微結晶についても行うことに成功し、単一結晶内部の水分解反応の詳細を調べることができたことは大きな成果である。水分解反応におけるプロトン共役電子移動機構やプロトン放出経路の研究も順調に進んでいる。また、電子受容体側については、QAがMnクラスターの有無によって制御されるというこれまでの定説を覆し、QBにおけるpHに依存する電子移動制御の機構を明らかにした。こうした光化学系Ⅱの電子伝達系に存在する電子移動制御機構を明らかにしたことは、非常に重要である。さらに初発電荷分離を起こす反応中心クロロフィルおよび副次的電子移動経路を形成するCytb559についても、これまでに多くの重要な結果が得られている。
    1.光合成水分解機構の解明:Mn2+の光酸化過程を光誘起フーリエ変換赤外(FTIR)差分光法で調べ、さらにquantum mechanics/molecular mechanics (QM/MM)計算によってそのスペクトルをシュミレーションすることによって、最初のMn2+結合位置および光活性化の機構を調べる。また、Mn4CaO5クラスターのカルボキシル配位子の変異体を用いて、昨年度に見出したD1-D170Hのアミノ酸変換の分子機構を調べ、光活性化における、カルボキシル配位子の役割を明らかにする。
    2.鉄-キノン電子受容体の反応機構:異なるpHにおいて非ヘム鉄およびキノン電子受容体QBの酸化還元反応を、FTIR法や遅延蛍光を用いて調べ、鉄-キノン電子受容体のpHに依存する反応制御機構を明らかにする。また、QB近傍に存在するヒスチジンD1-H252の変異体を用いて同様の実験を行い、QBの電子・プロトン移動反応およびそのpH依存性におけるこのヒスチジン残基の役割を調べる。
    3.反応中心クロロフィルの電荷分離反応機構の解明:ChlD1とChlD2の両方を同時に変えた組換え体を完成させ、これまでに作製した変異体と共に反応中心クロロフィルの速度論およびエネルギー論的な反応機構を明らかにする。またChlzD1およびChlzD2変異体を作製し、P680変異体と合わせて吸収分光測定を行い、副次的電子移動経路に関わるクロロフィル分子を特定する。昨年度、組換え体作製を試みたが致死した変異体があったため、光従属栄養下で生育可能な組換え系を完成させ、これを宿主に用いて変異体を完成させる。

    令和元年7月に大津市で領域会議を、11月20-24日に広島で第3回公開シンポジウムを兼ねた第3回太陽燃料国際会議・人工光合成に関する国際会議2019(ISF-3/ICARP2019)を開催し、領域内での研究成果の発表・交流や、領域外への研究内容の紹介・成果の発信を広く行った。特にISF-3/ICARP2019では、光合成や人工光合成分野で多数の国際的に著名な研究者を含む約400名の参加者があり、成功裏に終了した。上記2回の会議に合わせて総括班会議を開催し、領域全体の研究方針・計画を確認し、領域内での異分野融合研究を推進するための方策・体制を確立した。また、ISF-3/ICARP2019の開催に合わせて、地球温暖化解決のための国際イニシアチブ(24カ国が参加)であるMission Innovation(MI)における人工光合成分野の提言のロードマップを作製するため、ホスト国としてInnovation Challenge 5(IC5)/Japan Sunlight Conversion Road mapping workshopを開催し、10カ国から人工光合成研究のキーパーソンである30名程度の参加があり,活発な議論がなされた。
    総括班の中で、国際相互派遣支援班、国際招聘支援班、領域内共同研究支援班を設置し、領域内から広くそれぞれの内容に合った提案を募集し、審査を経て、それぞれの提案を採択し、総括班経費から実施を支援した。それらの結果、英国グラスゴー大学のRichard J. Cogdell教授、オランダ・アムステルダム自由大学の Rienk van Grondelle 名誉教授等を班員の研究室に招聘し、共同研究についての打ち合わせや情報交換を行った。また、多数の国内共同研究を実施した。
    月1回の領域ニュースレターを発行し、領域内外への情報発信・異分野交流を行った。
    第3回太陽燃料国際会議・人工光合成に関する国際会議2019(ISF-3/ICARP2019, 3rd International Solar Fuel Conference/ International Conference on Artificial Photosynthesis-2019)では、22国から光合成・人工光合成分野の主要な研究者を含む約400名の参加者があり、極めて成功裏に終わった。特にISF-3はこのシリーズの国際会議の第3回目で、前2回はそれぞれスウェーデン、米国で開催され、今回はアジアでの初開催となったが、参加者数は今回が最も多かった。また、これに合わせて開催されたMission Innovation(MI)会議でも、人工光合成分野でInnovation Challenge 5(IC5)のための政策提言に関して活発な議論が行われ、ロードマップ作製に関するいくつかの重要な合意が得られた。
    総括班内で設置した国際相互派遣支援班、国際招聘支援班、領域内共同研究支援班により、光合成や人工光合成分野で著名な研究者を招聘し、共同研究に関する打ち合わせを実施したほか、班員同士で活発な共同研究の支援が行われ、異分野共同研究を促進した。
    引き続き国際共同研究や領域内での異分野融合研究を強力に推進するため、総括班内で設置した国際相互派遣支援班、国際招聘支援班、領域内共同研究支援班により、領域内でそれぞれの提案を公募し、その中から優れたプロジェクトに対して総括班予算から支援する。
    年2回の総括班会議、1回の領域会議、1回の公開シンポジウム開催を継続して開催し、月1回のニュースレター発行を維持する。ホームページ上では研究成果、論文発表、プレスリリース、研究関連報道などの情報を随時更新し、領域内外への情報発信を積極的に行う。加えて、公募班メンバーを加えた各班(A班、B班、C班)のメンバー会議、及び領域メンバーの主催・共催による各種国内・国際学会・シンポジウムを積極的に支援する。

  5. 光合成による水分解:プロトン共役電子移動の分子機作

    光化学系ⅡのMn4CaO5クラスターにおいて行われる光合成水分解反応の分子機構について、近傍アミノ酸の変異体の赤外分光解析により、以下の成果を得た。
    1. 好熱性シアノバクテリアT. elongatus のYD欠損変異体(D2-Y160F)から調製した光化学系Ⅱタンパク質について、S2-S3遷移の電子・プロトン移動速度のpH依存性を時間分解赤外分光法を用いて調べた。その結果、より低いpHにおいて、~350 μsの速度成分が著しく遅延することが示され、この成分がプロトン移動律速の電子移動反応によることを明らかにした。
    2.架橋酸素O4に水素結合する水分子と相互作用するセリン残基(D1-Ser169)をアラニンに置換したD1-S169A変異体を作成し、その反応を光誘起フーリエ変換赤外(FTIR)差分光法、時間分解赤外分光法および熱発光を用いて調べた。その結果、この変異により、Mn4CaO5クラスターの酸化還元電位は低下するものの、各中間状態遷移の効率および速度には大きな変化は見られなかった。このことから、O4サイトは水分解の触媒反応に直接関与していないが、S0-S1遷移でのプロトン放出経路としての役割を果たしている可能性が示された。
    3. Mn4CaO5クラスターにおいてMnとCaを架橋するD1-Asp170をヒスチジンに置換したD1-D170H変異体を作成し、その光化学系Ⅱタンパク質を光誘起FTIR差分光および質量分析によって解析した。その結果、変異導入されたHis側鎖が、主に元のAsp側鎖に変換され、野生株と同様のFTIRスペクトルが観測されることが示された。これは、これまでに知られていない、Mn結合サイトに特異的な光誘起アミノ酸変換である可能性がある。
    水分解反応における水分子・プロトン移動経路については、D1-Asn298やD1-Ser169などのMnクラスター近傍の変異体の赤外分光解析が予定通りに進んでいる。時間分解赤外法を用いた、反応機構の研究も予定通りに進み、S2-S3遷移における、プロトン移動律速の電子移動機構、およびS0-S1遷移での電子移動律速のプロトン共役電子移動を初めて検出することができた。さらに、アミノ酸配位子についての研究において、その変異体の光変換の観測という予想外の成果も得られている。
    引き続き、シアノバクテリアの変異体を用いて、水分解反応における、プロトン移動・水分子取り込み機構とその経路、およびアミノ酸配位子の構造についての研究を進める。
    (1)S2-S3遷移におけるプロトン・水分子移動機構:YZチャネル上に位置するD1-N298をAlaに置換したD1-N298A変異体を用いてフーリエ変換赤外分光解析、時間分解赤外解析、熱発光測定を行い、S2-S3遷移におけるN298A変異の効果を調べる。さらに、野生株およびD1-N298A変異体の光化学系ⅡにおいてClチャンネルを形成するClイオンを硝酸イオンに置換した試料について同様の測定を行い、硝酸イオン置換の効果を調べる。これらの結果から、S2-S3遷移におけるプロトン放出および水分子取り込みがどの経路で行われるかを明らかにする。
    (2)S0-S1遷移におけるプロトン・水分子移動機構:異なるプロトン化構造を持つS0状態のモデルについて、Mn4CaO5クラスターの近傍タンパク質を取り込んだ、quantum mechanics/molecular mechanics(QM/MM)計算を行う。その結果から、S0-S1遷移の赤外差スペクトルをシュミュレーションし、実測の光誘起FTIR差スペクトルと比較によって、S0状態におけるMn4CaO5クラスターのプロトン化構造を明らかにする。そのプロトン化構造に基づいて、S0-S1遷移におけるプロトン移動経路を予測し、それを検証するための変異導入を計画する。
    (3)アミノ酸配位子の光変換:Mnイオンのカルボキシル配位子の変異体におけるアミノ酸変換の機構を、アミノ酸の同位体置換体を用いた解析や、他のアミノ酸への変異の効果などによってさらに追求する。

    By using the developed cryogenic microscope system, intracellular inhomogeneous distributions of photosynthetic proteins of Chlamydomonas, a model organism for the photosynthesis study, were observed. The analysis clarified the extent of co-localizations of photosystem proteins and light-harvesting antenna proteins. This results demonstrated that antenna proteins actually shuttles between the two photosystem proteins upon state transition, which is a regulation mechanism of excitation balance of the two photosystems.
    A system enabling scanning of the excitation wavelength was developed. By using the system, the fluorescence excitation spectra of single photosynthesis proteins were obtained for the first time. Information about the excitation-energy flow within the protein was obtained.
