    Salt intrusion into paddy fields decrease rice growth and yield, thus it is a serious problem to be solved. Rice is a salt-sensitive crop and it is important to improve its salt tolerance. Under saline conditions, salt is absorbed by root, transported to the leaf blades and accumulated salt inhibit photosynthesis and growth in rice. Therefore, removing salt from xylem vessels in root and leaf sheath is a key mechanism to decrease salt accumulation in leaf blade. In this study, we found that leaf sheath has a salt removal ability to decrease salt transportation to leaf blade. We also found that leaf sheaths unload Na and Cl ions from xylem vessels and transport them to the fundamental parenchyma cells. Using around 300 rice varieties, we performed genome wide association analysis regarding salt removal ability in leaf sheath. As a result, we found that the key gene may be localized on chromosome 5 for Na removal. Also, several SNPs were found to be involved in Cl removal ability.

    Zea nicaraguensis, which is a wild relative of maize (Z. mays ssp. mays), has higher tolerance of soil waterlogging than maize. We found that Z. nicaraguensis (but not maize) can form a barrier to radial oxygen loss (ROL). To identify a locus controlling ROL barrier formation in Z. nicaraguensis, we used a library of introgression lines (ILs), each containing a chromosome segment from Z. nicaraguensis in the maize inbred line Mi29. By fine-mapping and phenotyping, we discovered that the gene controlling ROL barrier formation was located on about 232 kb region of chromosome 3. We conducted gene expression analyses and then finally found one candidate gene. We also carried out suberin staining of root cross sections. However, we were not able to find any relationship between suberin staining pattern and root ROL barrier formation.

    Under waterlogged conditions, soybean forms secondary aerenchyma (SA) in the hypocotyl. In this study, I used 2 cultivars, Asoaogari and Enrei. The SA formation is 1-2 days earlier in Asoaogari than in Enrei. The grafting of the 2 cultivars showed that the SA formation-promoting factors of Asoaogari exist in the hypocotyl, but not in leaves. On the other hand, I found that the SA formation was inhibited in the darkness or CO2 deficiency even under waterlogged conditions. The SA formation was inhibited under waterlogged conditions when we cut seedlings above the epicotyl, but sucrose loading to the cut surface recovered its formation. In the heat-girdled seedling under waterlogged conditions, the SA was formed only above the heat-girdled position. Moreover, sucrose was highly accumulated in the hypocotyl under waterlogged conditions. These results suggest that sucrose transport from leaves is important for the SA formation in soybean hypocotyls under waterlogged conditions.

    ダイズの耐湿性付与に重要な役割を果たす二次通気組織形成機構を解明することが本研究の目的である。これまでの解析により、二次通気組織形成には葉で合成された糖(スクロース)が湛水条件下で胚軸に輸送されることが重要であることを明らかにした。そのことを確認するために、明条件または暗条件で湛水処理をしたダイズの胚軸内のスクロースの蓄積量を測定した。その結果、明条件下で湛水処理をした場合にのみ、胚軸内でスクロースの蓄積量の増加が観察された。また、湛水条件下で、上胚軸を切断した幼植物体の切断面に糖を添加することによって、二次通気組織形成が観察されるかどうかを調査したところ、グルコース処理時には部分的に、スクロース処理時には非切断時と同程度まで二次通気組織形成能が回復することが明らかとなった。それに対して、ネガティブコントロールの糖として用いたマンニトールで処理した場合では二次通気組織は形成されなかった。これらの結果から、湛水条件下の二次通気組織形成には、葉で光合成によって生合成される糖(スクロース)が師部を通して胚軸に供給されることが、二次通気組織の形成に重要であることを確認できた。
    また、二次通気組織形成に関与する調節因子としてCLEペプチドが考えられているので、二次通気組織形成時に特異的に発現するCLE遺伝子を同定して、組織別の定量RT-PCRを実施した。また、人工合成したCLEペプチドを投与して、二次通気組織形成への効果を確かめた。
    今後、これらの解析を通して、糖やCLEペプチドによる二次通気組織形成の制御機構を明らかにする予定である。

    This study was conducted with the object of analyzing the possibility of cropping system in rice and soybean in Swamp which spreads in Indonesia.     Cultivated rice (Oryza sativa) produces not only diterpene phytohormone gibberellins but also labdane-related diterpene phytoalexins, including momilactones, phytocassanes and oryzalexins. Our studies indicated that enzyme genes responsible for early-step biosynthesis of these diterpenoids express according to their biological roles, one is growth regulation and another is defense. Our studies also showed that momilactones and phytocassanes were accumulated in leaf blade samples of wild species Oryza rufipogon.

    In rice, the hypodermis in the outer part of roots forms a suberized cell wall (Casparian strip and/or suberin lamellae), which inhibits the flow of water and ions. We examined if an ABC transporter RCN1 is involved in the Casparian strip and suberin lamellae formations and tried to understand the mechanism of suberization in Casparian strip and suberin lamellae by identifying transcription factors, which control expressions of suberin biosynthetic genes and suberin transporter genes. Under stagnant deoxygenated conditions, well-suberized hypodermis developed in the wild type but not in the rcn1 mutant. An apoplastic tracer, periodic acid, was blocked at the hypodermis in the wild type but not in rcn1, indicating that the apoplastic barrier in the mutant was impaired. Moreover, we identified some transcription factors, which might control the formations of Casparian strip and suberin lamellae.

    本研究では、イネの酸素漏出(ROL)バリアの構成成分の解明を目指した。そのために、好気条件下で生育させたイネの幼植物体をさらに嫌気還元条件で14日間処理した根を根端から1㎝毎に分離して、代謝産物を抽出した。各々の代謝産物をNMR (800MHz)解析を行ったところ、糖類、アミノ酸類などの様々な代謝物の蓄積量が、根の部位または生育条件によって異なることが明らかになった。特にリンゴ酸の蓄積パターンがROLバリアの形成パターンと非常に似ていたことから、リンゴ酸がROLバリア形成に関与する可能性が考えられた。スベリンの前駆体になる脂肪酸に関しては、GC-MS分析をした結果、ROLバリア形成条件において、極長鎖脂肪酸の蓄積量が増加していることが明らかになった。この結果から、嫌気還元処理をすることによって、イネの根では極長鎖脂肪酸の生合成が促進され、それらの極長鎖脂肪酸由来のスベリンモノマーが生成されることが示唆された。
    さらに、根の各組織より全RNAを抽出して、マイクロアレイ解析を行ったところ、リンゴ酸代謝関連遺伝子の発現に特徴があることが分かった。つまり、嫌気還元条件下においてプラスチドで働くNADP-Mclic emyme (NADP-ME)とPyruvated ehydrog㎝ase (PDH)をコードする遺伝子の発現が誘導されることが明らかになった。この結果より、細胞内の細胞質で生成されるリンゴ酸がプラスチドに輸送されて、プラスチド内でNADP-MEおよびPDHによってリンゴ酸からアセチルCoAへの変換が促進されることが示唆された。アセチルCoAは脂肪酸生合成の最初のステップの基質となる物質であるので、嫌気還元条件下ではリンゴ酸の蓄積量が増加することで、脂肪酸の生合成が促進されるという可能性が考えられた。
    これらの発見によつて、イネの根で嫌気還元条件特異的に蓄積するスベリンが生合成されるまでにどのような代謝経路を経るのかを示唆することができた。

    Aerenchyma enables internal aeration between shoots and roots, and its formation is therefore important for the adaptation of plants to excess water environments. I examined optimal conditions for comprehensive proteome analysis using the cortex isolated by laser microdissection (LM) from maize roots. I determined an optimal fixative for fixation of tissues. Moreover, I detected Alcohol dehydrogenase protein by western blotting using LM-isolated cortex proteins. However, I failed to obtain enough amounts of proteins that are required for comprehensive proteome analysis.

    Genetic engineering of chloroplast genome in higher plants has been attempted since 1980’s, and has been successful in tobacco and several species. Chloroplast transformation has advantages over nucleargenome, because i) maternal inheritance of the genome eliminates dispersal of a transgene through pollen, ii) no gene silencing allows transgene to be expressed at highlevel, and iii) homologous recombination targets a transgene into a desired genomic region. Despite such advantages, efficiency of chloroplast transformation is extremely low in rice and other crops. We reasoned that one obstacle may be a small size of chloroplasts in these species. In this study, we propose to utilize a mutant that has enlarged chloroplasts. Isolation and characterization of a rice mutant showing large chloroplasts, identification of the gene responsible for this mutation, and an attempt to use this line for rice chloroplast transformation, have been conducted.

    畑作物では通気組織はできるものの、Radial O_2 Loss(ROL)バリアが形成されないために、酸素の大部分が根の基部から土壌へと漏洩してしまい、根端が酸欠になることが湿害発生の原因となる。ROLバリアとは、根の基部からの酸素漏洩を防ぐバリアのことで、通気組織を介した酸素輸送過程での酸素損失を最小限に抑えて根端まで効率的に酸素を供給する役割を担っている。本研究の材料であるHordeum marinumは畑作物と異なり、通気組織のほかにROLバリアを形成できるために、根端まで効率よく酸素を運ぶことにより過湿環境に適応することができる。
    平成23年度はH.marinumのROLバリアを形成する系統のH21と形成しない系統のH90を用いて、低酸素処理を行い、ROLバリアの形成パターンを調査した。その結果、H21では顕著にROLバリアが形成されていたのに対して、H90ではROLバリアの形成はほとんど観察されなかった。また、外皮のスベリンラメラの染色を行った結果、H21ではスベリンの蓄積が観察されたのに対して、H90では観察されなかった。さらに、ガスクロマトグラフィーを用いて、H21とH90の根におけるスベリン含量の分析を行った。しかし、スベリンモノマーの組成については、スベリン染色のときほどの明確な差がH21とH90の間で見られなかった。その結果の再現性を確認するために、再度ガスクロマトグラフィー用の試料を調製したので、今後、再度スベリン分析を行う予定である。H.marinumの根の外皮付近の組織のみをレーザーマイクロダイセクションで単離後、網羅的な遺伝子発現解析を行うことになっていたが、これについては今後発展させる予定である。

    Endocytosis performs a wide range of functions in animals and plants. Clathrin-coated vesicle (CCV) formation is an initial step of endocytosis, and in animal cells is largely achieved by dynamins. However, little is known of its molecular mechanisms in plant cells. To identify dynamin-related proteins (DRPs) involved in endocytic CCV formation in plant cells, we compared the behaviors of two structurally different Arabidopsis DRPs, DRP2B and DRP1A. DRP2B and DRP1A colocalized and assembled/disassembled together at the plasma membrane in Arabidopsis cells. DRP2B and DRP1A participate together in endocytic CCV formation in Arabidopsis cells despite the difference of their molecular properties.

    This study aimed to reveal the molecular mechanisms of self-incompatibility in the Brassicaceae and the Solanaceae. In the Brassicaceae, we have analyzed the downstream signaling cascades after the perception of the pollen determinant(SP11 ligand) by the pistil determinant(SRK receptor kinase), and identified some candidate molecules and some mutants related to the cascades. In the Solanaceae, we have revealed that the pollen determinant comprises multiple types of F-box proteins(SLFs), and proposed the "collaborative non-self recognition model" suggesting that SLFs collectively recognize and detoxify entire suite of non-self S-RNases.

    研究材料として用いているBrassica junceaのCMS系統は、細胞質雄性不稔の形質の他に、雄蕊の一部が花弁のような組織に変化する形質が観察されるユニークな材料である。これまで、CMS系統と稔性のある維持系統間のミトコンドリアゲノム構造および遺伝子発現の比較解析を通して、細胞質雄性不稔を引き起こす原因遺伝子orf220を同定した。植物ミトコンドリアへの形質転換系はまだ確立されていないので、orf220遺伝子のCMSへの関与を証明するために、orf220遺伝子にミトコンドリア輸送シグナルを接続し、アグロバクテリウム法でBrassica junceaの稔性系統に導入した。その結果、形質転換体では、CMS系統と同様に雄性不稔のほか雄蕊の形態変化が観察され、orf220遺伝子がこのCMS系統の雄性不稔ミトコンドリア遺伝子であることの証明に成功した。Brassica junceaのCMS系統では雄蕊の形態変化が観察されていることから、雄蕊内の特定の組織でCMSに研異的な遺伝子発現の変化が起きていると予想されるので、さらに、このCMS系統の生殖器官形成過程でどのようなRetrograde調節がなされているかの解明を試みた。そのために、雄蕊の特定の組織を純粋に単離できるLaser Microdissection(LMD)を利用した。これらの組織に対するLMDの実験系を確立し、マイクロアレイ解析を行うために必要なRNAを単離できた。研究期間の途中でYang博士が帰国したために、現時点での本研究の成果はLMDによってRNAを単離できたところまでであるが、今後、マイクロアレイやcDNAの大規模塩基配列解析を進めることで、雄蕊内の特定の組織でCMSに特異的に発現する遺伝子の同定を目指す。
