    Photosynthetic water oxidation is performed through a light-driven cycle of five intermediates (S0-S4states) in photosystem II (PSII). The S2→ S3transition, which involves concerted water and proton transfer, is a key process for understanding the water oxidation mechanism. Here, to identify the water and proton transfer pathways during the S2→ S3transition, we examined the effects of D1-N298A mutation and NO3-substitution for Cl-, which perturbed the O1 and Cl channels, respectively, on the S2→ S3kinetics using time-resolved infrared spectroscopy. The S2→ S3transition was retarded both upon NO3-substitution and upon D1-N298A mutation, whereas it was unaffected by further NO3-substitution in N298A PSII. The H/D kinetic isotope effect in N298A PSII was relatively small, revealing that water transfer is a rate-limiting step in this mutant. From these results, it was suggested that during the S2→ S3transition, water delivery and proton release occur through the O1 and Cl channels, respectively.

    © 2020 American Chemical Society. Photosynthetic water oxidation takes place at the Mn4CaO5 cluster in photosystem II. Although the atomic structures of its intermediates called S states have recently been reported, the catalytic mechanism of water oxidation has not been well understood. Here, to investigate the involvement of the O4 site of the Mn4CaO5 cluster and a water channel from O4 in the water oxidation reaction, we examined the effects of D1-S169A mutation, which perturbs the interaction of a water molecule hydrogen-bonded with O4, by thermoluminescence (TL), Fourier transform infrared (FTIR) difference, and time-resolved infrared (TRIR) measurements. The observed upshifts of TL peaks and some changes in FTIR spectra upon S169A mutation revealed the perturbations of the redox potential of the Mn4CaO5 cluster and the interactions of the surrounding hydrogen bond network. In contrast, FTIR oscillation patterns and TRIR traces showed only minor effects of the mutation on the efficiencies and kinetics of individual S-state transitions. It was thus concluded that the O4 site plays a role in retaining the redox potential and the structure of the hydrogen bond network, whereas it is unlikely to be directly involved in the catalytic reaction of substrate water except for proton transfer through the O4 water chain.

    © 2019 The density of homogeneous phase fluid mixtures for various carbon dioxide (CO2)/organic solvent systems was measured in homogeneous phase. Solvents included cyclohexane (C6H12), methylcyclohexane (C6H11CH3), and ethylbenzene (C6H5C2H5). Density measurements were performed using a high-pressure vibration type density meter equipped with a circulation pump and variable volume viewing cell which guarantee the homogeneity of mixtures. The density was measured at temperatures ranging from 313 K to 353 K and pressures up to 20 MPa, and the CO2 composition varied from (0 to 80) mol%. The obtained experimental data were correlated according to a density equation. The resulting correlation agrees well with the experimental data.

    The phase behaviors of carbon dioxide (CO2)/tetramethyl orthosilicate (TMOS)/poly(methyl methacrylate) (PMMA) and CO2/TMOS/poly(styrene) (PS) ternary systems were measured. The measurements were performed using a synthetic method involving observations of the bubble point and cloud point. The phase boundaries were measured at temperatures ranging from 313.2 K to 393.2 K and CO2 weight fractions ranging from 0.01 to 0.40. At a constant TMOS:polymer weight ratio of 9:1, both systems showed similar CO2 mass fraction dependence of their phase behaviors. When the vapor-liquid (VL) and vapor-liquid-liquid (VLL) phase boundaries of the ternary systems were compared with those of the binary CO2/TMOS system using polymer-free bases, they were found to be nearly identical. On the other hand, the two systems showed completely opposite polymer mass fraction dependence of their phase behaviors at a constant CO2 ratio.

    The phase behaviors of carbon dioxide (CO2)/trimethoxy(methyl)silane (MTMS) and CO2/Methylsilicate 51 (MS-51: methylsilicate tetramer) binary systems were determined. The experiments were performed using a synthetic method by observing the bubble point. The phase boundaries were determined at temperatures of 313.2 K-393.2 K and CO2 molar fractions of 0.2-0.9. The experimentally obtained bubble, points of the CO2/MTMS binary system were correlated using the Peng-Robinson equation of state (PR EoS), and the molecular interaction parameters k(ij) and l(ij) were determined. The results of the CO2/ MS-51 binary system were correlated using the PR EoS and the Sanchez-Lacombe equation of state (SL EoS). In the SL EoS, two types of mixing rules, originally proposed by Sanchez et al. which uses k(ij) as a fitting parameter and modified by McHugh et al. which uses two fitting parameters k(ij) and l(ij), were used. For this system, the experimental results were correlated better using the SL EoS than using the PR EoS, and the latter mixing rule was more suitable than the former. (C) 2017 Elsevier B.V.     Carbon nanotubes are a novel material for next-generation applications. In this study, we generated carbon nanotube and green fluorescent protein (GFP) conjugates using affinity binding peptides. The carbon nanotube-binding motif was introduced into the N-terminus of the GFP through molecular biology methods. Multiple GFPs were successfully aligned on a single-walled carbon nanotube via the molecular recognition function of the peptide aptamer, which was confirmed through transmission electron microscopy and optical analysis. Fluorescence spectral analysis results also suggested that the carbon nanotube-GFP complex was autonomously formed with orientation and without causing protein denaturation during immobilization. This simple process has a widespread potential for fabricating carbon nanotube-biomolecule hybrid devices. (C) 2017 The Japan Society of Applied Physics

    The ultrasonic degradation of methylene blue was carried out in the absence and presence of dimethyl sulfoxide (DMSO) as a radical scavenger for various frequencies, and the effects of DMSO addition on the degradation rate constant estimated by assuming first-order kinetics were investigated. The degradation reaction rate decreased with DMSO addition, and hydroxyl radicals were observed to play important roles in the degradation of methylene blue. However, the degradation reaction did not stop with DMSO addition, and the degradation rate constant in the presence of DMSO was not affected by ultrasonic frequency.     Ultrasonic emulsification is known to be useful in preparation of nanoemulsions, because use of surfactant can be reduced. Moreover, nanoemulsions with droplet diameters of around 100 nm can be prepared by sequential ultrasonic irradiation from low to high frequency. In this study, oil-in-water systems of toluene-water emulsions were prepared by ultrasonic emulsification and mechanical emulsification. Ultrasound emulsification was found to be more suitable than mechanical emulsification with a homogenizer. In addition, a two-step emulsification method was applied to preparation of emulsion. Crude emulsion was prepared mechanically in the first stage, and this was irradiated with ultrasound in the second stage. The droplet size of the emulsion was found to be influenced by the relationship between droplet size of the crude emulsion and ultrasonic frequency in the second stage. (C) 2015 The Authors.     The ultrasonic degradation of hazardous organic compounds in an aqueous solution is an attractive technology for wastewater treatment. The kinetics of ultrasonic degradation has been investigated. However, there have been only a few quantitative studies of the effect of ultrasonic frequency on degradation rate. In this study, the ultrasonic degradation of methylene blue was performed at various frequencies, especially in the high frequency region, and various initial concentrations. From the results of the time dependence of the methylene blue concentration in this study, ultrasonic degradation was considered to be a pseudo-first-order reaction. The apparent degradation rate constant and sonochemical efficiency have a linear relationship, and the apparent rate constant and initial concentration have an inversely proportional relationship. These phenomena well agree with our proposed formula for estimating the apparent degradation rate constant. In addition, our proposed model is also applicable to the ultrasonic degradation of nonvolatile and hydrophilic compounds. (C) 2014 The Japan Society of Applied Physics

    Gloeobacter violaceus PCC 7421 is considered, by molecular phylogenetic analyses, to be an early-branching cyanobacterium within the cyanobacterial clade. G. violaceus is the only known oxygenic photosynthetic organism that lacks thylakoid membranes. There is only one report on the development of a transformation system for G. violaceus [H. Guo, X. Xu, Prog. Nat. Sci. 14 (2004) 31-35] and further studies using the system have not been reported. In the present study, we succeeded in introducing an expression vector (pKUT1121) derived from a broad-host-range plasmid, RSF1010, into G. violaceus by conjugation. The frequency of transformation of our system is significantly higher than that described in the previous report. In addition, luciferase heterologously expressed in G. violaceus functioned as a reporter. The established system will promote the molecular genetic studies on G. violaceus.     We report on the assembly process for single-walled carbon nanotubes (SW-CNTs) bundle on the tip of cantilever for atomic force microscopy (AFM) by dielectrophoresis (DEP). In DEP process, an ac electric field (20 Vp-p, 5 MHz) was applied between a tungsten probe and the cantilever tip to form the SW-CNTs bundle bridge. We can monitor the SW-CNTs bridging process in real time under an optical microscope and successfully assembled SW-CNTs bundle with a diameter of approximately 100 nm on the tip. To compare the performance of our fablicated CNT cantilever with conventional cantilevers, we scanned a porous aluminum surface using AFM. From AFM measurements, we confirmed that our cantilever by DEP allowed us to obtain high resolution images similarly to conventional cantilevers with strong mechanical strength and good stability of scanning.

    Using an artificial peptide library, we have identified a peptide that has strict selective affinity for ZnO surfaces. The binding affinity of the peptide on the ZnO surface can be controlled simply through changes in phosphate concentration at constant pH and temperature. In this study, we functionalized inorganic nanoparticles by orderly conjugating ZnO-binding peptides (ZnOBPs) on the surface of cadmium selenide (CdSe) nanoparticles and performed spontaneous and reversible nanopatterning of ZnOBP-displayed nanoparticles on lithographed ZnO films. Conjugation of ZnOBPs on CdSe nanoparticles caused spontaneous adsorption of the nanoparticles on a ZnO film, and fluorescence and cathodoluminescence images clearly showed specific adsorption of nanoparticles on the ZnO films lithographed on nano-and micrometer scales. The selectively bound nanoparticles on ZnO films were completely released by changing the phosphate concentration in solution; such release did not require heat or mechanical applications. Repeated capture and release of nanoparticles were achieved on the micrometer scale. Our results show the potential of material-binding peptides for nanopatterning and dynamic microarrays.

    Photosynthetic oxygen evolution by plants, algae, and cyanobacteria is performed at the Mn4Ca cluster in photosystem II (PSI!) by light-driven water oxidation. It has been proposed that CP43-Arg357, which is located in the vicinity of the Mn4Ca cluster, plays a key role in the O-2 evolution mechanism; however, direct evidence for its involvement in the reaction has not yet been obtained. In this study, we have for the first time detected the structural coupling of CP43-Arg357 with the Mn4Ca cluster by means of isotope-edited Fourier transform infrared (FTIR) spectroscopy. Light-induced FTIR difference spectra upon the S-1 -&gt; S-2 transition (S-2/S-1 difference spectra) of the Mn4Ca cluster were measured using isolated PSII core complexes from Synechocystis sp. PCC 6803 cells, where the Arg side chains were labeled with either [eta(1,2)-N-15(2)]Arg or [zeta-C-13]Arg. Bands due to Arg side chain vibrations, which were extracted by taking a double difference between the S-2/S-1 spectra of isotope-labeled and unlabeled samples, were found at 1700-1600 and 1700-1550 cm(-1) for [eta(1,2)-N-15(2)]Arg- and [zeta-C-13]Arg-labeled PSII, respectively. These frequency regions are in good agreement with those of the CN/NH2 vibrations of a guanidinium group in difference spectra between isotope-labeled and unlabeled Arg in aqueous solutions. The detected Arg bands in the S-2/S-1 difference spectra were attributed to CP43-Arg357, which is the only Arg residue located near the Mn4Ca cluster. The presence of relatively high frequency bands arising from unlabeled Arg suggested that the guanidinium N(eta)H2 is engaged in strong hydrogen bonding. These results indicate that CP43-Arg357 interacts with the Mn4Ca cluster probably through direct hydrogen bonding to a first coordination shell ligand of a redox-active Mn ion. This structural coupling of CP43-Arg357 may play a crucial role in the water oxidation reactions.

    Peptides with affinity for inorganic materials hold promise for immobilizing functional nanostructures on a solid surface. In this study, a peptide with strict selective affinity for ZnO surfaces was utilized. We functionalized cadmium selenide (CdSe) nanoparticles by conjugating ZnO-binding peptides (ZnOBPs) on the surface of the nanoparticles and performed spontaneous and reversible patterning of ZnOBP-displayed nanoparticles on lithographed ZnO films. Conjugation of ZnOBPs on CdSe nanoparticles caused spontaneous adsorption of the nanoparticles on a ZnO film, and fluorescence and cathodoluminescence images clearly showed specific adsorption of nanoparticles on nano- and micrometer scales. The selectively-bound nanoparticles on ZnO films were completely released by changing the phosphate concentration in solution
    such release did not require heat or mechanical applications. Our results show the potential of material-binding peptides for nanopatterning and dynamic microarray applications. © 2011 IEEE.

    Water oxidation by photosystem (PS) II in oxygenic photosynthetic organisms is a major source of energy on the earth, leading to the production of a stable reductant. Mechanisms generating a high oxidation potential for water oxidation have been a major focus of photosynthesis research. This potential has not been estimated directly but has been measured by the redox potential of the primary electron acceptor, pheophytin (Phe)a. However, the reported values for Phe a are still controversial. Here, we measured the redox potential of Phe a under physiological conditions (pH 7.0; 25 degrees C) in two cyanobacteria with different special pair chlorophylls (Chls): Synechocystis sp. PCC 6803, whose special pair for PS II consists of Chl a, and Acaryochloris marina MBIC 11017, whose special pair for PS II consists of Chl d. We obtained redox potentials of -536 +/- 8 mV for Synechocystis sp. PCC 6803 and -478 +/- 24 mV for A. marina on PS II complexes in the presence of 1.0 M betaine. The difference in the redox potential of Phe a between the two species closely corresponded with the difference in the light energy absorbed by Chl a versus Chl d. We estimated the potentials of the special pair of PS II to be 1.20 V and 1.18 V for Synechocystis sp. PCC 6803 (P680) and A. marina (P713), respectively. This clearly indicates conservation in the properties of water-oxidation systems in oxygenic photosynthetic organisms, irrespective of the special-pair chlorophylls.

    We constructed a mutant (CP43-Glu354Gln) of the cyanobacterium Synechocystis sp. PCC 6803 in which the glutamic acid at position 354 of the 43 kDa chlorophyll protein (CP43) was replaced with glutamine. To determine the effect of this mutation on the reaction processes of the Mn cluster in the oxygen-evolving complex, we mainly analyzed the spectroscopic properties, including Fourier transform infrared (FTIR) spectroscopy, of photosystem II core complexes. Mutant cells exhibited a lower oxygen-evolving activity than wild-type cells, and an altered pattern of flash-dependent delayed luminescence. This phenotype differed somewhat from an earlier report of the same mutant [Strickler, M. A., et al. (2008) Philos. Trans. R. Soc. London, Ser. B 363, 1179-1187]. FTIR difference spectroscopy revealed that CP43-Glu354 functions as a ligand to the Mn cluster, most likely with bridging bidentate coordination to two Mn ions in the S(1) state and chelating bidentate coordination to a single Mn ion in the S(2) state. A single water molecule was bound to the same Mn atom to which CP43-Glu354 was ligated, and this Mn atom was oxidized in the S(1)-to-S(2) transition. This is the first report on a binding site of a water molecule relevant to a specific amino acid ligand. We found that the Mn ion or ligand that is oxidized in the S(2)-to-S(3) transition was not directly coupled to CP43-Glu354. While the definitive assignment or ligation to the Mn atoms is still under debate, our identification of a novel water binding site will lead to new insights into the oxygen evolution mechanism.

    Spectral properties, particularly fluorescence spectra and their time-dependent behavior, were investigated for a mutant of the cyanobacterium Synechocystis sp. PCC 6803 lacking the 43 kDa chlorophyll-protein (CP43, PsbC). Lack of CP43 was confirmed by a size shift of the corresponding gene and by Western blotting. The CP43-deletion mutant grown under heterotrophic conditions accumulated a small amount of photosystem (PS) II, but virtually no PS II fluorescence was observed. A 686-nm fluorescence band was clearly observed by phycocyanin excitation, coming from the terminal pigments of phycobilisomes. In contrast, no PS I fluorescence was detected by phycocyanin excitation when accumulation of PS II components was not proved by a fluorescence excitation spectrum, indicating that energy transfer to PS I chlorophyll a was mediated by PS II chlorophyll a. Direct connection of phycobilisomes with PS I was not suggested. Based on these fluorescence properties, the energy flow in the CP43-deletion mutant cells is discussed.

    Acaryochloris marina is a unique cyanobacterium that is able to produce chlorophyll d as its primary photosynthetic pigment and thus efficiently use far-red light for photosynthesis. Acaryochloris species have been isolated from marine environments in association with other oxygenic phototrophs, which may have driven the niche-filling introduction of chlorophyll d. To investigate these unique adaptations, we have sequenced the complete genome of A. marina. The DNA content of A. marina is composed of 8.3 million base pairs, which is among the largest bacterial genomes sequenced thus far. This large array of genomic data is distributed into nine single-copy plasmids that code for &gt;25% of the putative ORFs. Heavy duplication of genes related to DNA repair and recombination (primarily recA) and transposable elements could account for genetic mobility and genome expansion. We discuss points of interest for the biosynthesis of the unusual pigments chlorophyll d and a-carotene and genes responsible for previously studied phycobilin aggregates. Our analysis also reveals that A. marina carries a unique complement of genes for these phycobiliproteins in relation to those coding for antenna proteins related to those in Prochlorococcus species. The global replacement of major photosynthetic pigments appears to have incurred only minimal specializations in reaction center proteins to accommodate these alternate pigments. These features clearly show that the genus Acaryochloris is a fitting candidate for understanding genome expansion, gene acquisition, ecological adaptation, and photosystem modification in the cyanobacteria.

    Heterologous expression of the PufX membrane protein from purple photosynthetic bacterium Rhodohacter sphaeroidcs was attempted by using Escherichia (E.) coli cells. The PufX was overexpressed as a recombinant protein with a histidine tag added to the carboxyl terminus, and can be extracted from the cell membrane by various detergents. Circular dichroism measurements showed that the expressed PufX protein had alpha-helix contents of 29% in organic solvents and 22-26% in 0.8-2.0% (W/V) n-octyl beta-D-glucopyranoside solutions, suggesting that the PufX contains a substantial alpha-helical region composed of 18-22 amino acids. The PufX expressed in E. coli was examined by reconstitution experiments with LH1 alpha- and beta-polypeptides and bacteriochlorophyll a. It was shown that the PufX inhibited not only the reconstitution of the LH1 complex, but also the formation of the B820 subunit type complex at high concentrations, indicating that the expressed PufX is biologically active. Large-scale expression of the functional Puff membrane protein provides sufficient quantity for further biophysical and structural analyses of its biological function, and adds another example for producing highly hydrophobic integral membrane proteins using the E. coli expression system.

    Photosynthetic reaction centers (RCs) and their core light-harvesting complexes (LM-RCs), purified from a thermophile, Thermochromatium (T.) tepidum, and a mesophile, Allochromatium (A.) vinosum, were reconstituted into liposomes. The RC and the LH1-RC in the reconstituted liposomes were found intact from the absorption spectra at about 4 and 40 degrees C respectively. The thermal stability of the RCs of T. tepidum in the liposome was dependent on whether they were surrounded directly by lipids or-by the core light-harvesting complexes. The. results show that the RC of T. tepidum gains its thermostability through interactions with the LH1. These results are consistent with the result that the thermal stability of the LH1 in T. tepidum is similar in both the reconstituted LH1-RC liposome and ICM. This is clearly different from the mesophilic bacterium, A. vinosum. The thermal stability of RC was also affected by its subunit constitution: the RC containing a cytochrome subunit was more thermostable than the cytochrome-detached RC. This suggests that the cytochrome subunit might play a role in protecting the special pair pigments from denaturation. The thermal denaturation showed a second-order reaction dependence on time. The interaction of the pigments with proteins and/or lipids might be the cause of the second-order reaction profile.

    Heterologous expression of a bacterial light-harvesting (LH) integral membrane protein was attempted using Escherichia coli cells and cell-free synthesis systems prepared from E. coli extracts. The alpha-apoprotein of LH1 complex from purple photosynthetic bacterium Rhodospirillum rubrum was overexpressed as a recombinant protein with a histidine (His(6)) tag added to the carboxyl terminus. Both of the expression systems produced alpha-apoprotein in a fully functional form as can judged by its ability to form a structural subunit with native beta-apoprotein and the pigment molecule bacteriochlorophyll a. The expression product in E. coli appears to be located in the inner cell membrane and can be almost completely extracted by 0.5% (w/v) Triton X-100. Circular dichroism measurement indicated that the expressed alpha-apoproteins from both systems had alpha-helical contents essentially identical with that of the native one. About two thirds of the alpha-apoprotein expressed in E. coli was found to have the amino terminal methionine residue modified by a formyl group. About one third of the alpha-apoprotein expressed in the cell-free system was found to be oxidized at the side chain of the amino terminal methionine residue. Functional expression of the alpha-apoprotein using the cell-free system provides an useful example for producing highly hydrophobic integral membrane proteins with relatively large quantities sufficient for biophysical and structural analysis.

    A. marinaに関するゲノム情報解析と、他のシアノバクテリアの光合成機能の中でPS IIに関する解析が進展した。(1)Chl dを主要な色素として持つシアノバクテリア、A. marinaについての全ゲノム情報の解読が終了した。シアノバクテリアとしては例外的に9つのプラスミドを持ち、そのサイズの分布も374 kbpから2 kbpに渡り極めて稀な分布を持つこと、全ゲノムサイズが8.3 Mbpという例外的に大きいものであることが明らかになった。(2)反応中心タンパク質について、PS Iはアミノ酸置換が多く、PS II、特に光化学反応に関与する2サブユニットは小さいことが判明した。この事実は維持すべき色素の酸化還元電位との関連であると考えられる。(3)PS IIの電子受容体は他のシアノバクテリアと同じくフェオフィチンであることを我々は証明した。同時にその還元電位が他のシアノバクテリアに比べて高くなり、Chlの置換により獲得できるエネルギーが小さくなったことを補償する機構として極めて妥当な設計であることが判明した。この電位の変化に対応して、電子受容体の近傍に存在すると考えられるアミノ酸に多くの置換が見いだされた。(4)Gloeobacter violaceus PCC 7421において酸素発生系で機能する3種の膜表在性タンパク質(PsbO, PsbU, PsbV)の立体構造予測を行うと、ユニークなアミノ酸配列から予想されるとおりに、他のシアノバクテリアとは異なる構造が考えられた。他のタンパク質との相互作用のサイトなどが異なっており、酸素発生型光合成生物の間では極めて保存性が高いと考えられている水分解系に例外的な事例があることが強く示唆された。このことは、G. violaceusの水分解系が細胞膜外、ペリプラズムで行われることと高い相関があると考えられる。さらにG. violaceusの酸素発生活性は阻害剤であるDCMUによって約70%しか阻害されないことを見いだした。DCMU結合サイトのアミノ酸レベルでの変異はないので、その理由がまったく判然としていない。今後の大きな研究課題となった。
