    本研究ではヒト化CD20抗体を細胞外ドメインとして用いたCARを作成・評価し前臨床試験までを行う。既に抗体のAffinityが報告されているヒト化CD20抗体の遺伝子情報を用いるため、すぐに開発に着手できる。ヒト化抗体を用いたCARの開発はまだ報告が少ないが、導入した遺伝子産物に対する免疫反応が起こりにくいと考えられるヒト化抗体を用いて免疫反応を避ける必要性は高い。本研究で用いるCD20抗体はAffinityが既知であるために、標的細胞側の抗原発現量がリガンド結合後のT細胞機能に及ぼす影響を、異なるAffinityのCARを用いて検討可能である。さらにAffinity/avidityを変化させたときにCD28/CD27/4-1BBの細胞内ドメインの違いが及ぼす影響について、とくに抗原刺激後のT細胞の増殖・メモリー化に及ぼす影響についてin vivoで検討する。
    CD20刺激後のサイトカイン分泌や細胞分裂能から最適なCARの構造を決定し、これを用いた臨床試験の準備を行う。

    キメラ抗原受容体遺伝子改変T細胞療法 (CAR-T細胞療法)は、従来の治療が無効な血液腫瘍に対し治癒をもたらし得る免疫細胞療法であるが、半数以上の患者で再発を来し、他の治療法への移行決断に必要な効果・再発予測因子の同定や、効果を規定するCAR-T細胞の作用機序の解明が必要である。これまでの研究で、CAR-T細胞の長期維持や輸注後晩期の増幅が治療効果を規定することが示唆され、T細胞の免疫記憶 (メモリー化)が重要な役割をはたすことが想定されるがその詳細は明らかにされていない。本研究では、CAR-T細胞のメモリー化に注目し、その動態、分子学的機序、さらに、治療効果予測因子としての意義を明らかにする

    CD19に対するキメラ抗原受容体遺伝子を導入したT細胞療法 (CAR-T細胞療法)はこれまでの治療と比べて格段に良好な治療成績を示していますが、それでも効果が不十分な患者さんがおられます。そのため、我々は細胞の機能強化を目的としてこの研究を行います。すでにスクリーニングの結果、CUL5という遺伝子をノックアウトすることによりCAR-T細胞がCD19抗原陽性腫瘍細胞と出会ったあと、よく増えるようになることがわかっています。その効果のメカニズムを明らかにし、ヒトTリンパ球におけるCUL5の働きを解明することがこの研究の目的です。
    E3 ユビキチンリガーゼであるCullin-5 (CUL5) を、我々独自のGenome wide CRISPR screeningから標的遺伝子として同定した。これまでCUL5のノックアウト (KO) の効果について、CD19CAR-T細胞で機能を検討してきた。CUL5-KO CD19CAR-Tにおいて、CUL5-KOによりインターロイキン-2およびT細胞レセプターの刺激の下流である、JAK-STAT pathwayのタンパクのリン酸化が遷延することを示した。これにより抗原刺激後の細胞増殖が遷延・増強し、T細胞レセプター刺激後のエフェクター細胞分画が増加する。この効果は免疫不全マウスにヒト由来腫瘍細胞を移植したモデルを用いた in vivo実験でも確認された。
    CUL5-KOを行う場合には、guide RNAとCas9タンパクを遺伝子導入するために一度電気穿孔を行う必要があるが、これにより細胞が一旦かなりの割合で死滅する。その後生き残った細胞で前述のような効果が見られるが、その効率は決してよくない。この課題に対して、short hairpin RNAを用いて電気穿孔を行わずにCUL5の働きを落とすシステム(shCUL5)を作成した。このシステムでは一度のレンチウイルスによる遺伝子導入でCARとshCUL5の両方を発現させることが可能であり、効率よく細胞を作成することできる。細胞作成効率の上昇とともに、CUL5-KOの効果も見られることを確認している。現在、shCUL5-CD19CARやshCUL5-CD37CAR, shCUL5-固形癌特異的CARなどを作成して同様の働きを示すことを確認している。
    Cullin-5 KOによるどのような変化が起こるかについては、現象についてもメカニズムについてもはっきりしてきた。現在、その応用に関する検討を行っており、概ね順調に進展している。
    shCUL5-CD19CARやshCUL5-CD37CAR, shCUL5-固形癌特異的CARなどを作成して、in vivoの実験を進めている。同時にshCUL5-CD19CARと同様の働きを示すことをin vitroでも確認を進める。

    In this study, we attempted to develop a novel vector that enables gene transfer of TCR and adapter molecule (ATAM) into T cells by a single gene transfer. One virus vector method (1vv method) using all-in-one vector and two virus vector (2vv) method using two vectors used in the previous study showed that the expression of ATAM was about 3 times higher in 2vv method. The expression of ATAM was about 3-fold higher in the 2vv method. Comparison of the function of the TCR-T cells generated by the 2vv method showed that the cells generated by the 2vv method proliferated significantly better after antigen stimulation, and the intracellular signals after stimulation showed that the signals of phosphorylated Erk and p38 were significantly enhanced in the TCR-T cells generated by the 2vv method. The results showed that it is necessary to transfer three times more ATAM than TCR.

    腫瘍が醸成する免疫抑制的環境においても細胞傷害活性を発揮する腫瘍特異的免疫担当細胞の作成とその機能解析を行う。
    1.PD-L1に対するchimeric signaling receptor (PD-L1 CSR)の開発および機能解析
    腫瘍微小環境に発現し、腫瘍免疫に対して抑制的な環境を作る分子であるPD-L1 に着目し、その抑制的なシグナルを、逆に免疫に対する賦活化シグナルに変換する方法を開発する。
    2.CTLA-4 knock-out Tリンパ球の開発
    T リンパ球に備わっている自己抑制的メカニズムの中で中心的な役割を果たしているCTLA-4をノックアウトすることによって
    、腫瘍特異的T リンパ球が患者体内で自己抑制的に収縮する
    ことを防ぐ方法を開発する。

    Based on the amino acid sequence information of the antibody against the molecule PD-L1 thought to be expressed on tumor cells in order that tumor cells escape from immune cells, a single-chain antibody binding to PD-L1 was prepared. We further created a novel molecule with CD28 or 4-1BB or both as a part of domain. When this antibody was stained by fluorescent-labeled PD-L1 molecule, it was found that it stained well, indicating that it binds well to the antigen. This gene was transfected into T lymphocytes and its function was examined in vitro and in vivo in mice. On the other hand, we tried to knock out CTLA-4 molecule, which is expressed after activation of T cells and supposedly acts suppressively on self.
