    <p>Various pseudoscientific theories regarding health and food sciences have spread widely among both the general public and healthcare professionals. This study examined the degree of infiltration of such theories, particularly in the professional medical community, through an investigation targeting students attending a medical training school. In addition, to compare if these students’ perceptions would be corrected by proper education, the investigation targeted a fixed group of students from the time of their entrance until graduation from medical training school. The result revealed that various pseudoscientific theories regarding health and food sciences had infiltrated deeply among many students, and that some of their perceptions of several such theories had not been altered between their entrance to and graduation from the school. At the same time, significant findings were obtained regarding differences in the educational effect among students due to differences in learning content in the subject of basic medical sciences according to students’ major fields of study. The importance of education with concrete context has been demonstrated in previous studies, along with the importance of enhancing the level of students’ science literacy; the results of this study suggest the necessity of the same approach, of ensuring concrete context regarding pseudoscientific theories on health and food sciences.</p>

    Obesity is a serious international health problem that increases the risk of several diet-related chronic diseases. The genetic factors predisposing to obesity are little understood. Rcan2 was originally identified as a thyroid hormone-responsive gene. In the mouse, two splicing variants that harbor distinct tissue-specific expression patterns have been identified: Rcan2-3 is expressed predominately in the brain, whereas Rcan2-1 is expressed in the brain and other tissues such as the heart and skeletal muscle. Here, we show that Rcan2 plays an important role in the development of age- and diet-induced obesity. We found that although the loss of Rcan2 function in mice slowed growth in the first few weeks after birth, it also significantly ameliorated age- and diet-induced obesity in the mice by causing a reduction in food intake rather than increased energy expenditure. Rcan2 expression was most prominent in the ventromedial, dorsomedial and paraventricular hypothalamic nuclei governing energy balance. Fasting and refeeding experiment showed that only Rcan2-3 mRNA expression is up-regulated in the hypothalamus by fasting, and loss of Rcan2 significantly attenuates the hyperphagic response to starvation. Using double-mutant (Lep(ob/ob) Rcan2(-/-)) mice, we were also able to demonstrate that Rcan2 and leptin regulate body weight through different pathways. Our findings indicate that there may be an Rcan2-dependent mechanism which regulates food intake and promotes weight gain through a leptin-independent pathway. This study provides novel information on the control of body weight in mice and should improve our understanding of the mechanisms of obesity in humans.

    The neurological mouse mutation shambling (shm) exhibits ataxia and hindlimb paresis. Positional cloning of shm showed that it encodes contactin-associated protein (Caspr), which is required for formation of the paranodal junction in myelinated nerves. The shm mutation is a TT insertion in the Caspr gene that results in a frame shift and a premature stop codon at the COOH-terminus. The truncated Caspr protein that is generated lacks the transmembrane and cytoplasmic domains. Here, we found that the nodal/paranodal axoplasm of shm mice lack paranodal junctions and contain large mitochondria and abnormal accumulations of cytoplasmic organelles that indicate altered axonal transport. Immunohistochemical analysis of mutant mice showed reduced expression of Caspr, contactin, and neurofascin 155, which are thought to form a protein complex in the paranodal region; protein 4.1B, however, was normally distributed. The mutant mice had aberrant localization of voltage-gated ion channels on the axolemma of nodal/paranodal regions. Electrophysiological analysis demonstrated that the velocity of saltatory conduction was reduced in sciatic nerves and that the visual response was attenuated in the primary visual cortex. These abnormalities likely contribute to the neurological phenotype of the mutant mice.

    The mechanism of diabetes-induced congenital malformation remains to be elucidated. It has been reported that -lipoic acid (LA) prevents neural tube defects (NTDs) in offsprings of rats with streptozotocin-induced diabetes. Here, we evaluate the protective effect of LA against diabetic embryopathy, including NTDs, cardiovascular malformations (CVMs), and skeletal malformations, in mice.
    Female mice were rendered hyperglycemic using streptozotocin and then mated with normal male mouse. Pregnant diabetic or non-diabetic mice were treated daily with either LA (100 mg/kg body weight) or saline between gestational days 0 and 18. On day 18, fetuses were examined for congenital malformations.
    Plasma glucose levels on day 18 were not affected by LA treatment. No congenital malformations were observed either in the saline-treated or LA-treated non-diabetic group. In the saline-treated diabetic group, 39% of fetuses had external malformations and 30% had NTDs. In the LA-treated diabetic group, the corresponding proportions were 11 and 8%, respectively. LA treatment also decreased the incidence of CVMs from 30-3% and of skeletal malformations from 29-6%.
    We conclude that LA can reduce NTDs, CVMs and skeletal malformations in the offspring of diabetic mice at term delivery.

    健康や食品に関する疑似科学的な言説のうち、特にいわゆる「健康食品」の新聞広告を対象として、テキストマイニング分析によって明らかにする。さらに、宣伝を受け取る立場にある市民や医療従事者、学生がどのようにして人体や健康に関する知識や考え方を身につけてきているかを、初等中等教育課程の学習内容を対象として、教科横断的に検討する。
    1)「健康食品」に関する新聞内容の分析:2年分の全国紙1紙と地方紙1紙の新聞掲載広告並びに折り込み広告のうち、「コラーゲン」、「グルコサミン」、「コンドロイチン」のいずれかを含有成分とする商品の広告を対象とした。該当する広告は、36商品、広告数339であり、広告回数の多かった2商品を対象として、KH coderを使用して分析した。この結果、タイトルなと&#12441;に科学用語ではない語を使用したり、使用者の声などエビデンスレベルの低い内容をくり返し取りあげたりするほか、論理的というよりも情緒的な説明が特徴的であった。この成果はヘルスコミュニケーションウィーク広島(2021年10月)で発表した。
    2)教育課程の分析:小学校、中学校、高等学校の理科、社会科、技術家庭、保健体育を中心にして、分析を行っている。共同研究者からの依頼によって、気候変動や環境問題に関する教育内容の分析もおこない、第80回日本教育学会(2021年8月)で発表した。
    3)医療従事者への教育:理学療法士や鍼灸師などを対象としたセミナー(「臨床のための生理学講座」)を5回実施した。おもな内容は、ミネラルやビタミンについての栄養学的な基本知識とともに、それらを含有している栄養機能食品やいわゆる「健康食品」についての話題を提供した。関連して、視覚機能に関わる効能をとく食品注目して、エビデンスとされる文献内容を紹介した。

    Many foods are advertised as effective in promoting health and preventing diseases despite no scientific basis. Our research has shown that the influence of such advertisements are widespread among not only the general public (non-professionals) but also a substantial portion of medical professionals. For instance, one-third of the survey respondents, including ordinary citizens and medical professionals, selected the option “Seems correct” for the statement, “Greater intake of collagen provides moisture to the skin.” However, we found that students who are in the process of training to become medical professionals have some of their misconceptions changed by the education they receive after entering school. Medical professionals are supposed to possess correct knowledge and understanding in their fields of expertise and are responsible for giving medical advice to the general public. It has become clear how important education is.
