    When C4 plants are exposed to environmental stresses, mesophyll chloroplasts scattered at the cell periphery undergo "aggregative movement" toward bundle sheath cells. In this study, we aimed to elucidate the induction mechanism and physiological role of the aggregative movement, and obtained the following results.
    (1) The universality of aggregative movement in C4 plants was confirmed in a total of 36 C4 plant species across 11 different evolutionary lineages, including three C4 subtypes. (2) Decrease in CO2 concentration in leaves promotes aggregative movement under blue light. (3) The aggregative movement is also induced by low temperature stress under light. (4) Photorespiration may be involved in the induction of aggregative movement. (5) A different maldistribution of chloroplasts from the aggregative arrangement occurs in mesophyll cells of C3 plants.

    The organelle arrangement is different between mesophyll and bundle sheath cells, which are two types of photosynthetic cells of C4 plants, and is closely related to metabolic pathways. In this study, we aimed to elucidate the molecular mechanism and physiological significance of the organelle arrangement, and obtained the following results. (1)The intracellular arrangement and shape of bundle sheath chloroplasts in gramineous plants were three-dimensionally understood. (2)The three-dimensional changes in the intracellular arrangement and morphology of the mesophyll chloroplasts were revealed. (3)The acquisition step of the aggregation movement of mesophyll chloroplasts during the evolution of C4 plants was clarified. (4)The opposite distribution of Rubisco and starch granule was revealed in the bundle sheath chloroplasts of NAD-ME type C4 plants.

    In this research, we aim to clarify details of aspartate-mediated crosstalk between NADP-ME-type and PCK-type C4 cycles. In sorghum and green foxtail, NADP-ME-type C4 plants, there was no facilitative effect of aspartate on malate transport to chloroplasts and the PCK enzyme activity in leaf tissues was low. Therefore, it is inferred that the cooperative mechanism of the C4 cycle involving malate transport has been established in NADP-ME-type C4 plants having PCK-type C4 cycle such as maize. In addition, we found four kinds of Echinochloa C4 plants in Poaceae with markedly high PCK enzyme activity and they were expected to be good materials for clarifying the cooperation between both the C4 cycles.

    The invagination structure of a chloroplast that surrounds organelles such as mitochondria and peroxisomes within a thin layer of chloroplast stroma is called a chloroplast pocket. Since it is expected that stromules, which are long tubules including stroma, might be involved in the formation, we constructed the three-dimensional architecture of chloroplast pockets by using serial images obtained by TEM and FIB-SEM. Three types of chloroplast pockets were observed by TEM: organelles were completely enclosed in a chloroplast pocket (enclosed type), a chloroplast pocket with a small gap in the middle part (gap type), and a chloroplast pocket with one side open (open type). Mitochondria and peroxisomes were often in contact with the chloroplast pockets. FIB-SEM revealed chloroplasts with a sheet structure partially surrounding peroxisomes. This fact suggests that chloroplasts might construct large sheet structures that would be related to the formation of chloroplast pockets.
