    Rosa rugosa Thunb. ex. Murray possesses unique and value traits including: large, fragrant flowers, a long flowering period, disease resistance, and winter hardiness. An improved regeneration system is essential to exploit the potential of this species utilizing transgenic applications. The development of an efficient plantlet regeneration system was explored using different combinations and rates of 2,4-dichlorophenoxy acetic acid (2,4-D) and trans-zeatin (ZT) during the shoot induction stage. Immature seeds (14-28 days after pollination) were cut transversely in half and placed onto a medium containing Murashige and Skoog (MS) basal salts plus vitamins, 0.25% (w/v) Gellan gum, and varying concentrations and combinations of 2,4-D and ZT for 33 to 49 days. The treatment producing the highest frequency of explants with calli containing shoot primordia (14.4%) contained 2.0 mg/l 2,4-D and 1.5 mg/l ZT. Explants were transferred to the same medium without phytohormones for 56 to 67 days, each differentiating ~12 adventitious shoots. The shoots were easily separated and rooted after 28 days on medium without phytohormones or medium with 0.1 mg/l 1-naphthaleneacetic acid (NAA).

    We investigated S-RNase genotypes of 21 wild apples with Neville Corpman, and King of Tompkins 1, 2 and 3 by the PCR-digestion method. M. sylvestris 392390 (T1-2-66) did not contain any known S-RNase allele, and seemed to be useful as a pollinizer. Thirteen individuals (M. baccata (S1-7-15), M. fusca, M. fusca F 50 (T1-16-51), M. orientalis (W1-11-13), M. pumila Mill, M. pumila Pendula var. elise rathka, M. prunifolia USSR 18, M. prunifolia USSR 24, M. prunifolia USSR P, M. sieversii, M. sieversii (W1-10-49), M. sieversii sdl.2250 and M. sylvestris) contained an unidentified S-RNase allele with a known allele. Although M. baccata 4433 (79091) contained two known alleles, the S16a does not frequently occur in domestic Japanese cultivars. These wild apples also could be useful as pollinizers of cultivars in Japan, except for cultivars having an identical S-RNase allele. We have selected M. baccata 4433 (79091) as a pollinizer for the cultivar `Fuji'.

We selected `Maypole' and `Dolgo' as pollinizers for the cultivar `Fuji', and investigated the rate of fruit and seeds in `Fuji' fruits produced by pollen of the pollinizers. We developed a method for tracing pollen flow based on the leaf color of progeny and S-RNase allele of `Maypole', and on Simple Sequence Repeat (SSR) analyses of `Maypole' and `Dolgo'. These were powerful tools for determining the distance insects (mainly Osmia cornifrons) carry pollen from the pollinizers to `Fuji'. Although the fruit set of `Fuji' apples was not reduced with increasing distance between `Fuji' apple trees and pollinizers, (probably due to pollen flow from other commercial cultivars planted outside the area), the rate of `Fuji' apple fruit produced by the pollen of the pollinizers decreased with increasing distance. The rate of fruit produced by the pollinizers was 84% and 77% when `Fuji' was 2.5 m from `Maypole' and `Dolgo', respectively, and 71% and 64% when `Fuji' was 5 m from `Maypole' and `Dolgo', respectively, but was reduced to 47% and 39% when `Fuji' was 10 m from `Maypole' and `Dolgo', respectively. However, the spacing could cause reduced fruit size, and require extra fruit thinning for producing large fruits. It could also cause reduction of the yield for increasing the planting area of pollinizers. We recommend that pollinizers should be planted no more than 10 m from `Fuji'.

    栽培イチゴの苗増殖に関わる栄養成長期と果実生産に関わる生殖成長期の転換機構を分子レベルで解き明かし、超促成栽培に応用することを目的とする。具体的には、栽培イチゴの生殖成長すなわち花芽分化に関連する遺伝子群、栄養成長に関わる遺伝子群を特定して構造・機能解析を行なう。また、花芽分化時に発現変動する遺伝子群を指標とした超促成栽培技術を確立する。さらに、プラズマ照射による栄養・生殖成長転換系を実現し、新たな周年栽培の確立を目指す。
    栽培イチゴに普遍的な花芽分化誘導(促進)遺伝子FaFT3の機能解析を行うため、‘とちおとめ’から単離したFaFT3、FaFT1と野生種イチゴの花芽分化誘導遺伝子FvFT1をカリフラワーモザイクウイルス35Sプロモーター制御下でシロイヌナズナに導入し、過剰発現体を作出した。35S::FvFT1が短日条件下でのみ早期開花したのに対し、35S::FaFT3は短日・長日両条件下で早期開花した。一方、FvFT1と1アミノ酸のみ異なるFaFT1 (FaFT3ホモログ)を導入した35S::FaFT1では短日条件下で開花遅延が認められたことから、機能分化に関する知見を得るためにタンパク質モデリング解析を行った。花芽未分化状態と花芽分化初期状態の‘とちおとめ’クラウン茎頂部をレーザーキャプチャーマイクロダイセクション法にて取り出し、これを用いて実施したRNA-seq解析結果について再解析した。花芽分化時に発現上昇する702遺伝子についてGOエンリッチメント解析を行ったところ、生物学的プロセスでは、“非生物的、刺激、内生刺激への応答”、“花器官分化”、“生殖器官発生”に関わるGOアノテーションを持つ遺伝子が優先的に含まれており、具体的には、FaFT3に加えて花器官分化に関わるFaAP1、LEAFY、GID1などが見出されたが、Histone H4は見出されなかった。これに対し、花芽分化時に発現低下する449遺伝子からは、FaTFL1、FaAP2などの花芽分化抑制に関わる遺伝子が見出された。栽培イチゴ‘紅ほっぺ’クラウン部位に低温プラズマを直接照射したところ、プラズマ処理区において無処理区には見られない、既存の花芽分化誘導方法である間欠冷蔵処理区と同等の早い段階での収穫ピークが確認された。また、プラズマ処理区では、花芽分化初期にあたるクラウン頂部でのFaFT3発現レベルの上昇も確認された。
    今年度は、FvFT1とFaFT1についてタンパク質モデリング解析により立体構造を比較したところ、1アミノ酸置換部位においてFvFT1のH80の水素結合が2本であったのに対してFaFT1のN80では3本となっていた。これまでに、シロイヌナズナFTの1アミノ酸変異(Y84H)による花芽分化促進から抑制への機能変換が知られているが、この場合も水素結合の数が1本から2本と増加しており、タンパク質構造の安定性などが機能に重要であるのかもしれない。RNA-seq解析により、昨年度までに見出した栽培イチゴに普遍的な花芽分化誘導(促進)遺伝子FaFT3、抑制遺伝子FaTFL1に加えて、シロイヌナズナなどで見出されている遺伝子ホモログを確認することができ、シロイヌナズナと同様のカスケードにより栽培イチゴ花芽分化促進・抑制が制御されている可能性が示された。また、これまでの促成栽培のための低温短日処理、間欠冷蔵処理による花芽分化誘導に対し、今回、新たに低温プラズマ処理により花芽分化時期、収穫期を制御(前倒し)できる可能性が示された。これまでの方法では、冷蔵に関わるコストや多大な労働力の問題が指摘されているが、今後、プラズマ処理条件、方法の検討や改良などにより、より安価で効率的な促成栽培技術確立に繋がる可能性がある。さらに、クラウン茎頂部位から花芽未分化状態と花芽分化初期状態を確認した苗の葉を用いたRNA-seq.によるトランスクリプトーム解析を実施した。以上、前年度に引き続きいくつかの興味深い知見が得られており、おおむね順調に進展している。
    実施した葉組織のRNA-seqによるトランスクリプトーム解析結果から、葉で栄養・生殖成長転換期に発現変動する遺伝子群の同定を行い、葉組織に花芽分化誘導に関わる遺伝子群の発現変動が見られるかを検証して、葉組織の花芽分化への関わりを明らかにする。また、花芽分化時に特異的に発現上昇する遺伝子をバイオマーカーとして用いる超促成栽培法を確立する。さらに、プラズマ直接照射に対してプラズマ処理液を施用する方法による花芽分化誘導についての知見を得る。基礎研究としての栽培イチゴ花芽分化の分子機構の一端に関するこれまでのクラウン茎頂部、葉組織を用いた研究成果を取りまとめるとともに、応用研究としての発現変動遺伝子を指標とした超促成栽培や間欠冷蔵、プラズマ処理による新規促成栽培法についてもまとめる。

    We have investigated the wild origin of the continuous-flowering rose Rosa chinensis 'Old Blush' based on fieldwork in China for wild roses and genetic analyses of their nucleotide sequence diversity and similarity to Rosa chinensis. The results demonstrate hybrid origin of Rosa chinensis and one ancestral species is Rosa chinensis var. spontanea. We have also performed a large-scale screening of genetic resources for continuous-flowering roses and identified a rose cultivar with novel mutation for continuous-flowering characteristics.

    Forcing culture of June bearing type strawberries is carried out to harvest fruits at high demand season from late autumn to winter. Accurate identification of flower bud differentiation is essential for forcing culture. In this study, we identified the genes involved in the promotion and suppression of flower bud differentiation in cultivated strawberries, and obtained data that would lead to the practical application to super-forced cultivation using the expression changes of these genes as indicators. In addition, we have identified genes that are highly expressed during vegetative growth or runner development.
