    近年のビッグデータにより、脳腫瘍には多彩なゲノム・エピゲノム異常が蓄積していることが明らかになった。今後脳腫瘍の新規治療戦略を開発するためには、マスターレギュレーターとして中心的な役割を果たす分子異常を同定することが重要である。本研究では脳腫瘍オルガノイドモデルのゲノム編集を行うことで組織学的な悪性転化が誘導されるモデルを作成する。本モデルを用いて経時的にゲノム・エピゲノム異常を解析することで、悪性転化においてマスターレギュレーターの役割を果たす分子異常を同定する。予後不良な脳腫瘍の未だ明らかになっていない重要な弱点を同定し、新規治療戦略の開発につながる研究になることを期待する。

    研究代表者はこれまで、もやもや病病変部に対する網羅的遺伝子発現解析を行い、もやもや病患者では脳動脈に限局してCOL15A1遺伝子の低発現化が顕著であることを見出した。COL15A1蛋白は動脈組織の重要な構成成分であり、これまでの報告や病変の限局性からエピゲノム機序の存在が予想される。その低発現化は血管内皮や平滑筋細胞の変性、血管新生の促進など、もやもや病の様々な病態を矛盾なく説明し得ると考えた。今研究では、COL15A1遺伝子のエピゲノム制御機構と感受性遺伝子RNF213がそこに及ぼす影響を臨床検体を用いて、低発現となることで脳動脈に与える影響を細胞株や動物モデルを用いて明らかにする。
    もやもや病好発年齢、進行期である20歳以下の患者検体は、中高年以降の患者と比較して病変部動脈壁の菲薄化がさらに進行しており、western blotを行う上で十分量の蛋白が得られないと判断した。そこで2022年度は次世代シークエンシングを用いて、20歳未満の患者群、対照群についてトランスクリプトームを行うことで、病変部におけるCOL12A1の発現について検索を行うこととした。20歳以下のもやもや病患者を疾患群、焦点切除術を受けた難治性てんかん患者を対照群とした。脳表の動脈壁からtotal RNAを抽出し、NGSによるRNA-Seqを行った。データのマッピングと標準化ののち2群比較を行った。q-value<0.05かつ|Log2 fold change|>2のものをdifferentially expressed gene(DEG)とした。もやもや病10例、てんかん5例の比較に対しアッセイを行った。男女比と年齢分布は両群で一致させた。対照と比較し、もやもや病群では42遺伝子が高発現、144遺伝子が低発現、合計166遺伝子がDEGとなったが、CAL12A1はこれに含まれなかった。DEGsに対するエンリッチメント解析ではマトリソーム、細胞接着、血管新生に関連する遺伝子セットが有意に多く見られた。しかしながら特にもやもや病群において、次世代シークエンシングにおいてマッピング率が50%を下回るなど極端に低い値となっていた。開始total RNA量が微量であることが想定された。現在はこれまでのプロトコールよりもさらに微量の開始toral RNA量からライブラリ作成が可能なsingle cellを対象としたNGSライブラリ作成キットを用いたプロトコールを作成中である。
    特にもやもや病患者の病変部検体に含まれる蛋白やtotal RNA量が想定よりも微量であったことが原因と考えている。
    現在はこれまでのプロトコールよりもさらに微量の開始toral RNA量からライブラリ作成が可能なsingle cellを対象としたNGSライブラリ作成キットを用いたプロトコールを作成中である。

    We have focused on a molecule named Podoplanin (PDPN) as a therapeutic target of malignant brain tumors. PDPN, a type I transmembrane mucin-like glycoprotein, is abundant in several solid tumors including squamous cell carcinoma, malignant mesothelioma, Kaposi sarcoma, angiosarcoma, testicular seminoma, and brain tumor. The expression level of PDPN is reported to be in accordance with tumor malignancy. We can relieve patients from suffering potential side effects if new measures are developed by which PDPN-expressing tumor cells are only killed.

    Despite the accumulation of big data, the mechanism of tumorigenesis of most malignant brain tumors is still unclear, and it is essential to explore novel mechanistic concepts that have never been explored before. Epigenomic mechanisms are precisely regulated by modifications that activate (on) and repress (off) gene expression, and an imbalance between these modification contributes to cancer development. Recently, super-enhancer formation has been proposed as a novel driver mechanism of cancer, in which transcription factors such as Myc molecules alter the three-dimensional structure of the genome and enhance gene expression of cancer-related genes. In this study, we will elucidate the abnormal chromatin conformational changes during tumor formation using a mouse model which spontaneously develops brain tumor.

    The transcriptome analysis of injured Xenopus laevis tadpole and mice suggested that Neurod4L.S., a basic-helix-loop-helix transcription factor, was the most promising transcription factor to exert neuroregeneration after spinal cord injury (SCI) in mammals. We generated a pseudotyped retroviral vector with the neurotropic lymphocytic choriomeningitis virus (LCMV) envelope to deliver murine Neurod4 to mice undergoing SCI. SCI induced ependymal cells to neural stem cells (NSCs) in the central canal. The LCMV envelope-based pseudotypedvector preferentially introduced Neurod4 into activated NSCs, which converted to neurons with axonal regrowth and suppressed the scar-forming glial lineage. Neurod4-induced inhibitory neurons predominantly projected to the subsynaptic domains of motor neurons at the epicenter, and Neurod4-induced excitatory neurons predominantly projected to subsynaptic domains of motor neurons caudal to the injury site suggesting the formation of functional synapses.

    We established MADM mouse model which generates malignant glioma spontaneously. From an early stage of tumor formation, expression level of EZH2 was highly upregulated. Using glioma cell lines which are derived from MADM mice tumors, we found that EZH2 inhibitor (EPZ6438) inhibited cell growth of tumor cells. Also MRI imaging revealed that both oral administration of EPZ6438 and genetic depletion of EZH2 inhibited tumor growth in MADM mice significantly. We harvested tumor tissue of EZH2 knock-out MADM mice tumors, EZH2-intact MADM mice tumors and normal mice brains. Gene expression analysis with microarray revealed that expression levels of genes which are associated with neuronal differentiation are retained by knock out of EZH2 gene. These data indicated that EZH2 plays pivotal roles in glioma formation via dysregulation of histone modifications.
