    In this study, we showed that the new mass spectrometry PESI/MS/MS (Probe Electrospray Ionization-Tandem Mass Spectrometry) is a rapid, laborless and useful technology for the metabolite analysis of plants. By PESI/MS/MS, metabolites can be analyzed in seconds by simply sticking a needle into the sample, and local metabolite analysis and imaging analysis are also available. In addition, we developed the method to analyze 130 metabolites in 5 minutes and the platform to process the data automatically within 1 minute.

    Liver X receptor (LXR) activity is expected to have an innovative effect of extracting cholesterol from once formed atherosclerotic lesions and regressing arteriosclerosis, but it also induces a lipid synthesis system gene (SREBP1c). It has the drawback of further exacerbating hepatic lipid deposition. In this study, in order to overcome this drawback, the applicants have a special purpose (1): the inhibitory effect of bile acid on SREBP1c activity and the purpose (2): the efficacy of vabagenin, which is a novel ligand candidate for LXR activity that does not activate SREBP1c. As a result of examination using the NASH- myocardial infarction onset model (SHRSP5 / Dmcr rat), the lipid deposition in the arteries was improved without aggravating fatty liver.

    The purpose of the present study is to develop methods for sensitive identification and precise quantification of natural toxins in human biological specimens using deuterium labeled natural toxins by deuterium exchange reactions. Although we have not obtained deuterium labeled natural toxins, we have developed two methods for screening, identifying and quantification natural toxins in human biological specimens: a screening system for 56 natural toxins by liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (LC-QTOFMS), and a sensitive quantification method for 12 plant toxins by LC-tandem mass spectrometry, combined with simple solid-phase extraction.

    In this study, our research team have developed a real-time metabolome analytical technique using probe electrospray ionization tandem mass spectrometry (PESI/MS/MS) with time-series multivariate analyses. We optimized analytical conditions for real-time mass spectrometry, achieving successfully monitoring of 40 metabolites in a living mouse. Our research team has also developed a data pipeline using the R software, and this data pipeline can automatically execute bioinformatics analyses such as multivariate analysis. Moreover, we have developed a new approach using a state space model based on Bayesian statistical modeling for time-series data obtained from real-time monitoring. Finally, we applied this technique to a living mouse, demonstrating the practicality of our technique.

    In this study, we attempted to develop metabolome analysis techniques and create a model mouse of hypothermia to observe body temperature, activity and energy metabolism in infant exposed to undernutrition during gestation. Metabolome analysis methods in liver, muscle and brain were developed using GC/MS/MS and probe electrospray ionization/tandem mass spectrometry (PESI/MS/MS). We tried to create a model mouse of hypothermia induced by food restriction. Mice exposed to food restriction for 10 days or more did not give birth. In 5 days restriction, mice could give birth, but body temperature of the offspring in weaning period was almost same in control group. Therefore, we continue to consider creation of a model mouse of hypothermia.

    The aim of this project is to develop simple and comprehensive methods for determining novel abused drugs including synthetic cannabinoids by combining ultra-sensitive and/or highly specific mass spectrometers, and conventional mass spectrometers. We developed a simple method for determining several synthetic cannabinoids and their metabolites using a high-performance liquid chromatography-quadrupole time-of-flight mass spectrometer (LC-QTOFMS). The combination of an LC-QTOFMS and an LC-QTrap enabled the first identification of 5-fluoro-ADB (5-F-ADB) in the blood of a fatal intoxication case of 5-F-ADB and diphenidine. We then proposed possible metabolic pathways of the compounds. We are now investigating the difference of 5-F-ADB levels among organs in which the expression levels of carboxyesterase differ.
