    It has been believed that intracellular signaling via receptors, like proteins, is not possible for glycans. We have determined the downstream signals of recently discovered glycan receptors and the core structure of their glycan ligands, and elucidated the regulation of neuronal axon regeneration by them. The focus of this study was to establish a new concept of “glycan signaling” by taking a multifaceted approach. We found that in addition to the receptor tyrosine phosphatase PTPσthe receptor tyrosine kinase ALK acts as a glycan receptor, and that the long glycans chondroitin sulfate and heparan sulfate send opposite intracellular signals via a common substrate under these receptors and show opposite phenotypes for axon regeneration.

    Basigin (Bsg) promotes intracellular transport of monocarboxylic acids. Upon high-fat diet loading, Bsg deficiency ameliorated diabetes and insulin resistance, and reduce renal damage and fatty liver disease. To develop new therapeutic strategies for diabetic kidney disease and fatty liver disease, this study aimed to elucidate the detailed involvements of Bsg in intracellular metabolic mechanisms and to conduct therapeutic experiments with artificial nucleic acids using high-fat diet-loaded mice. In primary cultured cells of liver and renal proximal tubules, intermediate metabolites in the TCA circuit were reduced by 30% to 70% by Bsg inhibition. Systemic administration experiments in high-fat diet-loaded mice showed a trend toward improvement in serological indices in the liver, but it was difficult to conclude that the treatment had a significant effect on diabetes mellitus.

    The principal investigators searched for a new kinase that opposes the receptor-type tyrosine phosphatase PTPRσ, which inhibits neuroaxonal regeneration, and discovered that ALK (anaplastic lymphoma kinase) promotes neuroaxonal regeneration. In addition, the study found that dermatan sulfate (DS), a sulfated glycan, strongly activates ALK and promotes neuroaxonal regeneration in an ALK-dependent manner. In the current study, we demonstrated that DS has sufficient affinity for ALK and induces its clustering. Furthermore, we identified a group of molecules interacting with ALK and PTPRσ by the proximity-dependent labeling method BioID. The results revealed that there is a group of substrate molecules common to these two enzymes with opposing functions. Future studies can be developed on the physiological significance of these molecular switches.

    We were able to establish a Patient-derived Tumor Xenograft (PDX) model using tumors derived from neuroblastoma patients, and established PDX libraries in 5 cases from 18 neuroblastoma patients. Our established PDX model is used in the multicenter prospective clinical study (JN-H-20) nationwide.

    In the present study, we aimed to elucidate the underlying mechanisms of chronic kidney disease (CKD) and hepatic steatosis caused by nutritional overburden more in details. Therefore, we investigated the role of CD147/Basigin (BSG) to maintain intracellular energy homeostasis, using high fat diet (HFD)-induced wild-type and BSG gene-deficient (BSGKO) mice. In the kidneys of BSGKO mice, vacuolar degeneration of tubular epithelial cells and accumulation of lipofuscin-like substances were remarkably suppressed. These findings might be due to the phenomena that transport of the substrates such as pyruvate and lactate in the cells was suppressed by BSG deficits subsequently leading to the the reduction of TCA cycle intermediates and enhanced ketogenesis through activation of free fatty acid beta oxidation. In the setting, augmented BSG protein might be due to the promotion of autophagy, but not translocation to nuclei.

    We identified the actin-binding protein cortactin as a substrate of protein tyrosine phosphatase σ (PTPRσ). PTPRσ was activated by chondroitin sulfate and dephosphorylated cortactin. As a consequence, the autophagy flux was disrupted at the step of autophagosome-lysosome fusion. In addition, we succeeded in comprehensive screening of PTPRσ substrates.
    Our study provided solid evidence of the direct relationship between PTPR and autophagy. Considering that autophagy dysfunction is involved in various disorders such as neurodegenerative diseases, our results may influence a broad spectrum of research fields.

    Extracellular accumulation of amyloid beta peptides is a character of cerebral amyloid plaques in Alzheimer's disease. Heparan sulfate is an extracellular carbohydrate chain found in amyloid plaques in the brain of transgenic Alzheimer's model mice and patients with Alzheimer's disease. Highly sulfated subdomains within heparan sulfate chains are abundant in amyloid plaques of Alzheimer's mouse brains. Degradation of these subdomains may be a potential approach to activate microglial clearance of the plaques in Alzheimer's brain. We tested if the transgenic expression of an extracellular sulfatase in Alzheimer's model mice could facilitate removal of the subdomains and attenuate cerebral amyloid plaque formation. We have shown that a cell type-specific expression of an extracellular sulfatase resulted in negative regulation of amyloid plaque formation. An enzymatic remodeling of extracellular heparan sulfates in the brain can perhaps control Alzheimer’s pathogenesis.
