    This chapter outlines advances in the ruthenium-mediated alkyne [2+2+2] cycloaddition reactions. These can be classified into two major categories in terms of the type of products: (1) syntheses of benzene derivatives via alkyne [2+2+2] cycloadditions and (2) syntheses of heteroaromatics via [2+2+2] co-cycloadditions of alkynes with nitriles or heterocumulenes. Benzene ring-forming reactions are essentially prototypes of the corresponding heteroaromatic annulations. Therefore, the first class of reactions will be reviewed in the next section and followed by a discussion of the second class of reactions. The mechanistic aspects and synthetic applications of ruthenium-catalyzed [2+2+2] cycloadditions are also described to exemplify the scientific and practical significance of ruthenium catalysis.

    Syntheses of polysubstituted benzenes have often encountered difficulties in controlling regioselectivity and incompatibility to labile functionalities. Highly substituted benzeneboronic acid esters were synthesized by using ruthenium-catalyzed [2 + 2 + 2] cycloaddition of alkynylboronates under mild conditions. Multiply substituted iodobenzenes were also efficiently synthesized with defined substitution patterns via ruthenium-catalyzed [2 + 2 + 2] cycloaddition of iododiynes.

    遷移金属カルベン錯体は、有機合成において極めて有用なツールとなっている。本計画では、末端アセチレンとカチオン性ルテニウム錯体から発生するビニリデンカルベン錯体への酸素原子移動反応によるケテンの発生と、同過程を組み込んだ新規触媒系の創成を目指す。旧来のカルベン錯体からのケテン発生法は、予め調製されたカルベン錯体を用いる非触媒的手法であったことに加え、有毒な一酸化炭素の挿入に頼らざるを得なかった。本計画の達成によって、入手容易な末端アセチレンよりビニリデンカルベン錯体を発生させ、その酸化により所望のケテンを触媒的に発生させる実用的な手法が確立される。本手法は学術的に興味深いことに加え、極めて応用範囲の広いものである。さらに、本研究を通して開発される触媒的分子変換プロセスを駆使して、生理活性物質の合成を展開する。
