    皮革産業は世界貿易額が10兆円を越える巨大産業である。皮革製品は「クロムなめし」と「その他」に分類されるが、生産コストが低く関税が格安なので、世界の皮革製品の90%以上はクロムなめしで生産される。経済グローバル化に伴い、先進国が環境汚染・健康障害を誘発するクロムなめしの工程を途上国に押しつける構図が新たな地球規模問題をうみ出している。本研究は、皮革製品輸出国のバングラデシュ(途上国)と、主要輸入国である日本(先進国)に焦点をあて、A. 環境汚染の把握→B. 汚染物質の健康影響評価→C. 解決策の提案からなる問題解決型研究を両国が協働で実施し、本問題の解決をめざす。

    代表者等は、開発途上国の飲用井戸水に含まれる多種多様な元素の単独および複合経口曝露による健康被害を報告してきた。上記知見は、経口する井戸水が異なれば、汚染されている元素も異なる可能性を示している。本研究では、まず、開発途上国の飲用水元素汚染の現状に基づき、飲用水に含まれる元素の健康リスクをオーダーメイドで評価できる技術を開発し、浄化すべき元素を特定する。次に、飲用水に含まれる有害元素を過不足なく除去できる浄化技術を開発・実用化することで飲用水に起因する健康被害の緩和をめざす。また、本研究で開発された健康リスク評価技術を用いて、日本人における元素の健康リスクを評価し、健康増進に貢献する。
    元素に汚染された飲用水の健康リスクを正確に評価するためには、「元素曝露状況を解明するフィールドワーク研究」と「環境の影響を受けた個体に対する実験研究」の両方を推進することが必要である。本研究では、元素の経口曝露により誘発される健康被害を予防することを目的とし、1)飲用水元素汚染の現状を把握するとともに、2)疫学研究と実験研究を組み合わせて、元素の健康リスクをオーダーメイドで評価できる技術を開発して浄化すべき元素を特定し、3)オーダーメイドで有害元素を吸着できる浄化技術を開発する 。さらに、途上国における研究成果を用いて、4)日本人における元素の健康影響を評価することで、日本に成果を還元することを目的としている。
    本研究では、バングラデシュの飲用井戸水では、鉄濃度が高いことを示した。さらに、鉄の過剰摂取は、住民の聴力障害を誘発する可能性を報告した(Sci Rep 2019)。また、アフガニスタンの飲用井戸水では、リチウム及びボロンの濃度が高いことを示すとともに、リチウム(Sci Total Environ 2020)及びボロン(Environ Pollut 2020)の潜在的発癌毒性を示した。さらに、iPS細胞を用いて、水銀を例として元素の発生毒性をin vitroで評価できるシステムを開発(Chemosphere 2020)した。こうした研究実績が評価され、2019年度の日本衛生学会・学会賞を受賞し、飲用井戸水の健康リスクに関する研究成果は、学会賞受賞記念総説論文(Environ Health Prev Med 2020)としてまとめられた。
    コロナウイルス感染症のパンデミックにより、開発途上国におけるフィールドワーク研究は、余儀なくペンディング状態になっている。しかし、過去に採取した資料を、より新しい技術で、より詳細に解析することにより、この難局を乗り越える努力をしている。実際に、環境学におけるTop 10% Journalを数編公表しており、本研究は、おおむね順調に研究を進展していると判断できる。
    元素に汚染された飲用水の健康リスクを評価することを目的として、「元素曝露状況を解明するフィールドワーク研究」と「環境の影響を受けた個体に対する実験研究」の両方を推進する。
    1)飲用水元素汚染の現状把握:コロナウイルス感染症の状況を勘案しながら、開発途上国におけるフィールドワーク研究を推進する。フィールドワーク研究が難しい場合は、過去に採取した検体を、より新しい技術で解析することで乗り越える。
    2)元素の健康リスク評価と浄化すべき元素の特定:ICP-MSを用いて、元素取り込みに着目した細胞生物学実験を進める。さらに、LA-ICP-MSを用いて、臓器の元素分布を調べる動物実験を推進する。
    3)浄化技術の開発:日本の基準に対応できる浄化材を開発し、開発途上国への技術移転の準備を進める。
    4)日本人における元素の健康影響を評価:日本人の元素曝露状況を調べるとともに、微量元素曝露が健康に与える影響について、疫学実験で解明する。さらに、ヒトで得られた成果を、動物実験で検証する。

    【背景】 我々が日常的に使用している皮革製品は、「クロムなめし」と「その他のもの」に分類される。生産コストが低く、輸入税率も格段に安いので、世界の皮革製品の80%以上は、クロムなめしを用いて作製されている。経済グローバル化に伴い、環境汚染を誘発するクロムなめしの作業工程を途上国が担当する構図が、新たなグローバル・イシューを産み出している。
    【目的】 クロムなめし工場では、皮革製品の製造に大量の水を必要とする。クロムなめし工場から高濃度のクロム等の有害元素を含む大量の廃液が流れ出ると、工場内だけでなく、周辺水域に甚大な環境汚染と健康被害を誘発する可能性がある。本研究は、皮革製品を世界約70ヶ国に輸出しているバングラデシュと、主要輸入国の1つである日本に焦点をあて、1)環境汚染の現状把握→2)汚染物質の健康影響評価→3)解決策の提案・実践からなる研究を推進する。
    【研究実績】 バングラデシュの皮革工場集積地域(ハザリバーグ)は、ブラックスミス研究所の報告において、「世界で最も環境汚染が深刻な10地点」の1つに選別されたこともある重大汚染地域である。本研究は、1)ハザリバーグにおける環境汚染の現状把握(Chemosphere 2018a)を報告するとともに、2)皮革工場労働者(Chemosphere 2019;Environ Res 2020)及び細胞(Chemosphere 2018b)を用いた有害元素の健康影響評価に関する成果を国際科学誌に報告してきた。2021年度は、ハザリバーグにおける有機物による環境汚染を解明するとともに、有機物除去に有効なオリジナルの浄化材を提案した(Chemosphere 2021)。
    本研究は、バングラデシュの皮革工場集積地域における無機物質の汚染に加えて、有機物質の汚染を解明しただけでなく、解決策の1つとなりえる浄化材の開発に成功しているため。
    バングラデシュの皮革工場集積地域(ハザリバーグ)における有害元素汚染に焦点を当て、現状を確認するとともに、解決策となりえる浄化材の開発を推進する。さらに、皮革工場労働者における疫学調査を推進し、皮革工場に起因する有害元素が、ヒトの健康に与える影響を調べる。以上のようにして、1)環境汚染の現状把握→2)汚染物質の健康影響評価→3)解決策の提案・実践からなる研究を推進する。

    In this study, we first developed a device that can produce low frequency sound (low frequency sound stimulator). Then, our experimental study proposed a threshold at which low frequency sound induced health disorders (balance and motor dysfunction), and identified the potential mechanisms and damaged sites (otoliths and otolith membranes) in the vestibule. We also developed a novel system that quickly assesses the health risk of low frequency sound by cultured utricle. Moreover, our epidemiological studies evaluated the health effects of low frequency sound in young adults. Finally, our animal study showed that increased expression level of heat Shock protein 70 expressed in the otolithic membrane can prevent a damage of otolithic membrane and dysfunction of equilibrium and motor caused by low frequency. Thus, we proposed a preventive method for health disturbances caused by exposure to low frequency noise.

    Contamination of toxic elements in well drinking water has caused various element-originating diseases in tens of millions of people in Asian countries. In this study, the comprehensive study was conducted to quickly resolve the international environmental problems related to contamination of toxic elements in well drinking water. First, the present situation of elemental contamination of well drinking water was investigated in the areas including Afghanistan. Then, harmful elements that may induce carcinogenesis and/or disorders of sensory organs (melanosis, hearing loss, etc.) were detected by our multidisciplinary health risk assessment system consisted of experimental and epidemiological analyses. Finally, we developed a new remediation system that can remove harmful elements from well drinking water based on our results of health risk assessment.

    We previous developed RET-transgenic mice (RET-mice), in which development of hyperpigmented skin and benign melanocytic tumors at about 100% and development of melanoma at about 65%. In this study, we first selected molecules after performing microarray analysis using benign tumor and melanoma developed in RET-mouse. We then performed functional analysis for these three molecules. The molecules are involved in the regulation for anchorage- independent growth, invasion and metastasis. Thus, we have not only proposed plural candidate molecules that are available as a biomarker of melanoma but also provided basic data for developing a molecular target therapy that suppress melanoma proliferation, infiltration and metastasis.
